Mediobasal prosencephalic defects, including holoprosencephaly and cyclopia, in relation to the development of the human forebrain.

Four very early synophthalmic embryos were studied in serial sections and reconstructed graphically by the point-plotting method. Three belonged to stage 16 (5 weeks) and one to stages 19/20 (7 weeks). Recently completed accounts and reconstructions of the normal brains of staged human embryos served as controls for comparison with the abnormal examples. The embryos shared in common: holoprosencephaly, arhinencephaly sensu stricto (absence of olfactory nerve fibers, bulbs, and tracts), presence of a proboscis, synophthalmia with two lens vesicles, a retarded telencephalic wall, absence of the

Preliminary evaluation of the impact of the Chernobyl radiological contamination on the frequency of central nervous system malformations in 18 regions of Europe. The EUROCAT Working Group.

The teratological impact of radiological contamination from the Chernobyl accident was evaluated in relation to central nervous system and eye defects in 18 regional registries in nine countries of Western Europe. Six classes of anomaly were analysed: neural tube defects, arhinencephaly, microcephaly and brain reduction, hydrocephaly, anophthalmos and microphthalmos, and congenital cataract. Conceptions up to 31 August 1986 were grouped into two exposure cohorts. In cohort A the sensitive period of fetal development to radiation fell wholly or partly between 1 May and 30 June 1986. Cohort B in

[Clinical picture and course of children with CHARGE association].

Four children are described who presented malformations according to CHARGE-Association. The primary abnormalities were choanal atresia and/or Coloboma. All four cases exhibited esophageal malformations: atresia and/or fistulas. At autopsy, three children showed arhinencephaly which is rarely observed in patients with CHARGE-Association. Patients with the main features of CHARGE-Association have a very poor prognosis depending on the severity of the malformations.

Arhinencephaly. The spectrum of associated malformations.

Eight cases are presented of arhinencephaly and its associated malformations, which included 2 examples of holoprosencephaly and 3 of agenesis of the corpus callosum. Additional features included cortical malformations, anomalies of the long tracts and of the optic pathway, cerebellar hypoplasia and dentato-olivary dysplasia. Each of these components covered a wide spectrum ranging in severity from extreme to minimal. Craniofacial dysmorphism, and cardiac, renal and endocrine disorders were present in some cases. Only 2 cases were associated with chromosomal abnormalities, 1 with trisomy 13, t

Agenesis of the corpus callosum and limbic malformation in Apert syndrome (type I acrocephalosyndactyly).

Agenesis of the corpus callosum and malformation of limbic structures are described in a patient with Apert syndrome, a disorder characterized by acrocephaly, severe syndactyly, and often, mental retardation. Including the present case, malformation of the corpus callosum and/or limbic structures apparently has been reported in a total of ten patients with the syndrome. Complete or partial agenesis of the corpus callosum was found in six patients, septal defects in three, and arhinencephaly and ammonic hypoplasia in one. Since malformation or limbic structures are, to our knowledge, a consiste

Duplication 7p in a family with t(7;11): association with anomalies of the anterior cranial base.

Duplication 7p has been observed in association with several balanced translocations. Of eight previously recorded cases, only one exhibited duplication of the entire short arm of chromosome 7. We report on a newborn infant with multiple congenital anomalies and an abnormal chromosome constitution: 46,XX,-11,+der(11),t(7;11) (p11.1;p15.5)mat. The proposita appeared to possess an isolated duplication of the entire short arm of 7. The patient died at age 4 days because of respiratory complications of meconium aspiration. Clinical and postmortem findings included craniocerebral asymmetry, cranios

Multiple congenital malformations of the face, nervous system and musculoskeletal system of pigs.

Multiple congenital malformations occurred in 21 of 26 pigs (81%) in 3 litters from 2 piggeries. Two of 26 pigs (7%) were mummified while 3 of 26 (11%) were normal. Cheiloschisis was seen in all malformed pigs and was accompanied by various combinations of palatoschisis, arhinencephaly, hypoplasia of the spinal cord, arthrogryposis of the pelvic limbs, polydactyly, renal lesions and cauliflower ears. Histological examination suggested that arthrogryposis was due to neurogenic muscular atrophy. The teratogen was not identified.

Hydrocephalus, lissencephaly, ocular abnormalities and congenital muscular dystrophy. A Warburg syndrome variant?

The authors report a family in whom three members suffered from congenital hydrocephalus and ocular abnormalities. One of these patients showed along with these symptoms congenital muscular dystrophy. In this child, autopsy disclosed severe cerebral malformations consisting of lissencephaly, arhinencephaly, stenosis of aqueduct, Dandy-Walker cyst and cerebellar micropolygyria. The mode of transmission, the eyes abnormalities and the neuropathological findings of this family resemble the clinical and pathological aspects of Warburg syndrome. However, the presence of congenital muscle dystrophy

A rare case of Aicardi syndrome with severe brain malformation and hepatoblastoma.

A 2-month-old Japanese girl exhibited tonic seizure, agenesis of the corpus callosum, lacunar chorioretinopathy, vertebral anomalies, electroencephalographic abnormalities and a malignant tumor. Autopsy revealed a hepatoblastoma and severe brain malformations consisting of callosal agenesis, arhinencephaly, marked polymicrogyria and optic nerve anomalies. It was thought that the pathogenic factor in this case may exert its effect during the fourth or fifth week of intrauterine life, and then may continue until the beginning of neuronal migration (about 3 months). This is the first reported cas

An anatomical study of a duplication 6p based on two sibs.

Dup (6p) patients have a peculiar facial appearance (frontal bossing, hypotelorism, hypoplastic midface), low birthweight, cardiovascular defects, small kidneys, and psychomotor retardation. We thought that a detailed anatomical dissection would more precisely define the syndrome, which has been developed from clinical evaluations and autopsy reports. Our patient, a female adolescent, died at 17 11/12 years and is the oldest patient with this syndrome to be described. The brain and skull showed the greatest number of abnormalities. The brain was smaller than normal and abnormally shaped. Many

Bilateral cystic nephroblastomas and multiple malformations with trisomy 8 mosaicism.

The case of a 16-month-old female infant with bilateral cystic nephroblastomas, Dandy-Walker syndrome, microcephaly, bilateral cataracts, and cerebellar heterotopia is reported. The patient's older sister, who had had bilateral cystic nephroblastomas, botryoid sarcoma involving the vagina and urinary bladder, microcephaly, arhinencephaly, and bilateral cataracts, was described in a previous report. Chromosomal study in the present case confirmed trisomy 8 mosaicism (rate of mosaicism, 16 per cent). The familial occurrence and the chromosomal disorder suggest a syndrome involving genetic abnorm

Cerebellocele and associated central nervous system anomalies in the Meckel syndrome.

Three cases with the Meckel syndrome were autopsied and found to have: arhinencephaly , polymicrogyria , aqueductal stenosis, heterotopia of glial tissue, hypoplasia or agenesis of the cerebellar vermis, cranium bifidum associated with large occipital ventriculocele and others. The anomalies at the level of posterior fossa in this condition are classified as those belonging to the Chiari type III group of anomalies. This unusual set of anomalies which forms pathogenetic link between the Dandy-Walker and Chiari-Arnold group of anomalies in the posterior fossa seems to be very frequent in the Me

Cranial defects in the Goldenhar syndrome.

Four patients are presented with the Goldenhar syndrome (GS) and cranial defects consisting of plagiocephaly, microcephaly, skull defects, or intracranial dermoid cysts. Twelve cases from the literature add hydrocephalus, encephalocele, and arhinencephaly to a growing list of brain anomalies in GS. As a group, these patients emphasize the variability of GS and the increased risk for developmental retardation with multiple, severe, or unusual manifestations. The temporal relation of proposed teratogenic events in GS provides an opportunity to reconstruct biological relationships within the 3-5-

Facial duplication: case, review, and embryogenesis.

The craniofacial anatomy of an infant with facial duplication is described. There were four eyes, two noses, two maxillae, and one mandible. Anterior to the single pituitary the brain was duplicated and there was bilateral arhinencephaly. Portions of the brain were extruded into a large frontal encephalocele. Cases of symmetrical facial duplication reported in the literature range from two complete faces on a single head (diprosopus) to simple nasal duplication. The variety of patterns of duplication suggests that the doubling of facial components arises in several different ways: Forking of t

Median facio-cerebral anomalies in chick embryos resulting from local destruction of the anteriormost parts of the early neural plate and neural crest.

With the aid of micro-laser irradiation, in a total of 123 chick embryos varying in age from 24-26 hours of incubation (2- to 6-somite stages), the anteriormost parts of the neural crest and the neural plate were eliminated. As a result of this treatment, 15 out of 59 embryos, studied 24 hours after irradiation, showed manifest signs of maldevelopment of the telencephalon, eyes and olfactory placodes. At the age of 7 days (6 days after irradiation), the surviving 25 embryos all proved to have developed median facio-cerebral anomalies. These anomalies included arhinencephaly, holoprosencephaly,
