INTRODUCTION: Anterior segment dysgenesis is a heterogeneous group of developmental conditions affecting the anterior segment of the eye. These could have a significant impact on vision due to development of secondary childhood glaucoma. Their classification is complex due to genetic and clinical heterogeneity. Variants in the CPAMD8 gene have been shown to result in abnormalities such as cataract, iris hypoplasia, and ectopia lentis.
METHODS: We described two unrelated Mexican patients with anterior segment dysgenesis in whom novel biallelic variants in the CPAMD8 gene were identified. Their

BACKGROUND: Classic Homocystinuria (HCU) is the second most treatable aminoacidopathy. It affects multiple organs in varying degrees, and early recognition and treatment is crucial to improve the prognosis. Mutations in the CBS gene result in classic HCU, the most common form. Genetic testing is important for the accurate diagnosis of this severe disorder. METHOD: Case report. RESULT: A 6-year-old boy presented with high myopia, ectopia lentis, and hypotonia. A brain MRI showed no abnormality, and his electromyogram implicated myogenic damage. His serum homocysteine was 277.5 µmol/L (normal ≤

To date, several genetic factors have been reported in cases presenting both congenital cataract (CC) and spastic paraplegia (SPG), but the overall genetic landscape of this condition remains insufficiently understood. Here, we describe the clinical, genetic, and functional findings in a single Japanese family presenting with hereditary CC and SPG. Affected individuals exhibited severe CC and mild SPG symptoms. Whole exome sequencing combined with stringent filtering revealed no pathogenic variants in the three known genes. Instead, we identified a heterozygous missense variant in PAX6 (NM_001

Congenital ectopia lentis (CEL) is a rare pediatric ocular disorder characterized by zonular fiber defects leading to lens dislocation and is genetically heterogeneous. Among known causes, biallelic mutations in ADAMTSL-4 represent the second most common genetic contributor, frequently associated with ectopia pupillae (EP)-a distinct and clinically significant feature. However, the mechanisms by which ADAMTSL-4 mutations lead to these ocular abnormalities remain poorly understood, partly due to the lack of effective animal models. In this study, we generated adamtsl-4 knock-out zebrafish lines

Microphthalmia is a significant eye defect owing to its profound effects on visual acuity. Microphthalmia accounts for 3.2%-11.2% of blind children. To date, there has been no cure for this disease. In this study, we aimed to identify microphthalmia-like mutant mouse and study its growth and development. In this study, we identified mutant mice exhibiting eye abnormalities using a forward genetics approach in a C57BL/6J cohort. To identify ocular characteristics of the mutant mouse, we conducted systematic evaluations including basic measurements (body length, body weight, and palpebral fissur

This study investigated the posterior segment manifestations and molecular mechanisms of retinopathy in ectopia lentis (EL) by subjecting retinal pigment epithelial (RPE) cells to excessive mechanical stretching. A total of 127 patients with EL and 149 healthy controls underwent comprehensive ophthalmic examinations. Aqueous humor from 10 patients per group underwent untargeted metabolomic analysis. In vitro, ARPE-19 cells were subjected to excessive mechanical stretching using a cell-tank device mimicking axial elongation. The results showed that patients with EL exhibited significantly longe

An ~6-mo-old, free-ranging, male, American black bear (Ursus americanus) cub that was observed stumbling, acting disoriented, and falling, was dispatched and sent for postmortem evaluation. Bilaterally, round concretions were present in both the anterior and posterior aspects of the lens, disrupting the lens epithelium. These concretions were continuous with and extended from the inner surface of the lens capsule, as seen with a periodic acid-Schiff-hematoxylin reaction. Immunohistochemistry for alpha A crystallin was inconsistent, with weakly positive immunolabeling indicative of lens fibers.

What is known? ◦. Genetic disorders of calcium/phosphate metabolism can cause prenatal bone disease, often leading to postnatal respiratory distress and with patients exhibiting bone improvement with calcium supplementation. What's new? ◦. We broaden the TRPV6 variant fetal phenotype with bilateral ocular involvement, mirroring postnatal hypocalcemia features. ◦. Prenatal bone anomalies in constitutional bone diseases may overlap with calcium/phosphate disorders; thus, gene screening for these disorders should be performed in all unexplained fetal skeletal dysplasia cases.
