Genetic analysis and clinical characteristics of sporadic and familial congenital cataracts in southern Chinese families.

INTRODUCTION: Congenital cataract is a major cause of blindness and severe visual impairment in children. It may occur as an isolated ocular abnormality or in combination with microcornea, microphthalmia, aniridia, or glaucoma. It can also be part of syndromic conditions. Whole-exome sequencing (WES) is now recognized as an appropriate first-line approach for genetic testing in patients with congenital cataract. In this study, we use WES to characterize the genotype spectrum in a pediatric cataract cohort from southern China. METHODS: In this study, we aimed to clarify the genetic basis of co

βA4-Crystallin Mutations Disrupt Structural Stability and Crystallin Interactions in Congenital Cataract Pathogenesis.

PURPOSE: Mutations in the crystallin beta A4 (CRYBA4) gene, which encodes βA4-crystallin, are associated with congenital cataract; yet, the underlying pathogenic mechanisms remain elusive. We systematically investigated four cataract-associated βA4-crystallin mutants (βA4-Y67N, βA4-L69P, βA4-F94S, and βA4-G147V), each representing distinct structural contexts, to assess their effects on structural destabilization and interactions with key crystallins. METHODS: We employed a comprehensive approach, including protein expression and purification, size-exclusion chromatography, crosslinking, and

The temporal stability of visual cortical processing in humans depends on early experience.

Proper timing is essential for effective neural processing. Yet, how early postnatal vision shapes the temporal stability of human visual cortical processing remains unknown. Here, using electroencephalography, we examined cortical timing properties in individuals who were born pattern-vision blind due to congenital cataracts, but surgically recovered sight. Although sight-recovery individuals exhibited an attenuated cortical oscillatory phase coherence (i.e., higher temporal variability) during visual processing, their cortical oscillatory strength was unimpaired. Moreover, phase coherence in

Co-occurring DMD, GJA1, and novel FYCO1 variants in a proband from a consanguineous oculodentodigital dysplasia family: a rare multi-locus case report.

Whole-exome sequencing of the proband and the family revealed multi-locus pathogenic variants (MGVs) leading to multiple genetic diagnoses (MGDs), explaining the complex phenotype with neuromuscular, ocular, and craniofacial abnormalities. The proband harbored a de novo hemizygous DMD frameshift variant consistent with Duchenne muscular dystrophy, a paternally inherited heterozygous GJA1 in-frame indel associated with oculodentodigital dysplasia (ODDD), and a novel homozygous FYCO1 nonsense variant causing congenital cataract. Fraction of ROH (FROH) analyses indicated extended autozygosity, wh

Loss of function of retinol dehydrogenase 11 causes a recessive syndrome characterized by myopathy, retinal dystrophy, juvenile cataracts, and microcephaly.

PURPOSE: Biallelic variants in RDH11, encoding retinol dehydrogenase 11, have been associated with a syndromic disorder, based on 4 individuals from 2 unrelated families. We aimed to profile the clinical variability, natural history and associated molecular spectrum of this condition. METHODS: In the frame of a collaborative effort, clinical and molecular data were collected using a semistructured survey. Structural modeling of RDH11 with NADH(P) was used to assess the functional impact of missense variants. RESULTS: Sixteen affected individuals from 9 unrelated families with biallelic RDH11

A preliminary comparative study of peripheral capsular linear incision versus circular micro-tear capsulotomy for in situ lens regeneration.

BACKGROUND: In congenital cataract, conventional circular micro-tear capsulotomy often causes loss of anterior capsular lens epithelial cells, thereby impairing the integrity and transparency of the regenerated lens. Based on the concept of minimal capsulorhexis, we performed a preliminary pilot investigation using a 1.5 mm linear incision in the peripheral capsule to evaluate whether this technique could improve lens regeneration outcomes. METHODS: Forty 4-week-old New Zealand white rabbits were randomly allocated to two groups: peripheral capsular linear incision (Group A) and circular micr

Ophthalmological phenotype associated with biallelic CPAMD8 variants: first report in Mexican patients.

INTRODUCTION: Anterior segment dysgenesis is a heterogeneous group of developmental conditions affecting the anterior segment of the eye. These could have a significant impact on vision due to development of secondary childhood glaucoma. Their classification is complex due to genetic and clinical heterogeneity. Variants in the CPAMD8 gene have been shown to result in abnormalities such as cataract, iris hypoplasia, and ectopia lentis. METHODS: We described two unrelated Mexican patients with anterior segment dysgenesis in whom novel biallelic variants in the CPAMD8 gene were identified. Their

Hereditary Congenital Cataract With Spastic Paraplegia Associated With a Missense PAX6 Variant (p.Arg26Trp).

To date, several genetic factors have been reported in cases presenting both congenital cataract (CC) and spastic paraplegia (SPG), but the overall genetic landscape of this condition remains insufficiently understood. Here, we describe the clinical, genetic, and functional findings in a single Japanese family presenting with hereditary CC and SPG. Affected individuals exhibited severe CC and mild SPG symptoms. Whole exome sequencing combined with stringent filtering revealed no pathogenic variants in the three known genes. Instead, we identified a heterozygous missense variant in PAX6 (NM_001

Expanding the Prenatal Phenotypic Spectrum of TRPV6 Variants With Ocular Anomalies.

What is known? ◦. Genetic disorders of calcium/phosphate metabolism can cause prenatal bone disease, often leading to postnatal respiratory distress and with patients exhibiting bone improvement with calcium supplementation. What's new? ◦. We broaden the TRPV6 variant fetal phenotype with bilateral ocular involvement, mirroring postnatal hypocalcemia features. ◦. Prenatal bone anomalies in constitutional bone diseases may overlap with calcium/phosphate disorders; thus, gene screening for these disorders should be performed in all unexplained fetal skeletal dysplasia cases.

A Novel CRYBB2 Splicing Mutation Is Associated With Lens Extracellular Matrix Remodeling and Vascular Alterations in Congenital Cataract.

PURPOSE: We identify the genetic cause of autosomal dominant congenital cataract in a large family and define how a CRYBB2 splice-site variant perturbs lens development and associated ocular phenotypes. METHODS: Whole-exome sequencing and Sanger sequencing were used for variant identification and segregation. Wild-type (WT) or mutant CRYBB2 was expressed in lens epithelial cells to assess splicing and downstream transcriptional changes by RNA sequencing and quantitative reverse transcription PCR (qRT-PCR). Extracellular matrix (ECM) organization and cell-matrix interactions were examined by i
