Atomic force microscopy (AFM) is not only a high-resolution imaging technique but also a sensitive tool able to study biomechanical properties of biosamples (biomolecules, cells) in native conditions-i.e., in buffered solutions (culturing media) and stable temperature (mostly 37°C). Micromechanical transducers (cantilevers) are often used to map surface stiffness geometry, elastic, and contact parameters of living cells; however, they can also be used to monitor time course of dynamic forces (beating), their rate, and other biomechanical properties of cardiomyocyte contractility. Here we descr

Biochemical testing plays a crucial role in the detection and confirmation of Salmonella spp. Since the cultural characteristics of Salmonella often resemble those of other enteric pathogens, differentiation based solely on growth features can be challenging. Biochemical tests allow for accurate identification by exploiting the distinct metabolic traits of Salmonella. In this study, we described three key tests, including methyl red, hydrogen sulfide production, and urease production, which are particularly important for the confirmation of Salmonella.

While Salmonella is a leading bacterial cause of foodborne illness, its relative abundance in food, animals, and the environment is low. This necessitates the use of cultural enrichment to reduce the background flora and enable Salmonella to proliferate so that it can be subsequently isolated on agar or detected via molecular methods. Further, successful Salmonella isolation is often dependent on the sample type, or matrix, from which it is being isolated. Particularly where matrices have high microbial background, or high levels of organic material, attention should be paid to culture methods

The conventional method for Salmonella detection, as recommended by ISO 6579-1:2017(E), involves four sequential steps: preenrichment, selective enrichment, plating on selective media, and confirmation. The second step consists of Selective enrichment, which is performed using Muller-Kauffmann tetrathionate-novobiocin broth, Rappaport-Vassiliadis Soya Peptone Broth, or Modified Semisolid Rappaport-Vassiliadis (MSRV) agar. MSRV agar, widely used in food industry testing, exploits Salmonella motility to facilitate selective isolation by forming characteristic opaque halos. Its use reduces proces

Monitoring the production of safe beef free of Salmonella is important to its producers, regulators, and consumers. For testing, beef may be intact, nonintact, or ground, where either the ground beef is used for testing or a surface swab of the beef is used. Here is described the procedure for rapidly detecting the presence of Salmonella in beef samples and the necessary steps to confirm and identify the Salmonella present.

Conventional analysis of RNA products synthesized by bacterial RNA polymerase (RNAP) in in vitro transcription assays relies on radioactive labeling, followed by separation of labeled RNAs by denaturing urea-polyacrylamide gel electrophoresis (PAGE). This approach provides excellent resolution across a wide range of RNA lengths (from 2 nt to several hundred nucleotides), but the use of radioisotopes requires specialized facilities, strict waste-disposal procedures, and is often restricted. To circumvent these limitations, we adapted a standard in vitro transcription assay for use with the Agil

Flow cytometric analysis can provide valuable information on the viability, phenotype, and therapeutic potential of cell therapy products using a minimal amount of cell numbers. In this chapter, we describe methods for the analysis of clinical products that incorporate extra considerations within the protocols to help maintain consistency and confidence of staining. This can include methods for evaluating the transduction efficiency for T cells expressing chimeric antigen receptors (CAR-T), characterizing the frequency of diverse lymphocyte populations within polyclonal cell cultures and posti

BACKGROUND: Exposure to chewing tobacco is a known health risk factor but has received less attention than smoking in global tobacco control efforts. As the evidence base linking chewing tobacco use to poor health grows, it is essential to closely track the changing use of chewing tobacco distinct from other forms of smokeless tobacco, as well as the estimated burden associated with its use. These data are essential to inform policy makers who seek to identify and avert the health burden associated with these dangerous products.
METHODS: We estimated the prevalence of chewing tobacco use and

Botrytis cinerea, a major fungal pathogen of crops, has been extensively studied for its resistance to antifungal plant metabolites and agrochemicals. Evaluating the sensitivity of B. cinerea to these compounds is crucial for understanding its pathogenicity and for developing effective plant protection strategies. This study describes two methods for antifungal sensitivity assessment: the spot assay, which visualizes fungal growth on agar medium supplemented with antifungal compounds, and the resazurin assay, which quantifies fungal metabolic activity in liquid culture. Using saponins as antif

White lupin (Lupinus albus) is a legume species that develops two remarkable root adaptations to nutrient limitation: nitrogen-fixing nodules and dense cluster roots specialized for phosphate acquisition. These traits, together with the species' agronomic relevance and strong developmental plasticity, make white lupin a powerful model for studying local and systemic regulation of root development under nutrient stress. This chapter provides a comprehensive collection of experimental procedures designed to investigate cluster root formation, nodulation, and associated physiological responses at

Efficient and reproducible isolation of Botrytis species, including Botrytis cinerea and Botrytis pseudocinerea, is essential for studies on fungal biology, epidemiology, and host-pathogen interactions. The protocols described in this chapter provide practical methods for obtaining pure cultures from symptomatic and asymptomatic plant tissues, as well as from single spores and hyphal fragments. Each approach is detailed with step-by-step instructions, highlighting critical parameters for successful isolation and culture maintenance. The symptomatic and asymptomatic tissue methods enable rapid

Extracellular vesicles (EVs) are universally conserved and essential players in the communication between cells across nearly all organisms of life. Botrytis cinerea produces and secretes EVs that are critical in its virulence on plants. Numerous studies in various fungal species reveal that EVs are important across different pathosystems to facilitate fungal infection. For characterization of the cargo and role of EVs in fungal growth and communication, it is necessary to carefully and precisely purify fungal EVs from fungal culture or the plant interaction interface. Here, we present a metho

Botrytis cinerea causes gray mold across a wide range of crops and postharvest contexts. While integrated agronomic practices help, chemical fungicides are still often required, raising concerns about resistance development and environmental impact. Biological control agents (BCAs), particularly non-Saccharomyces yeasts, offer advantages such as simple nutritional needs, surface colonization potential, and lower risk of mycotoxin production. A robust, rapid, and cost-effective in vitro screening workflow is essential to identify promising BCAs that inhibit B. cinerea through fungistatic diffus

Using Botrytis cinerea (ATCC 58025) as the material, genomic DNA was extracted using the SDS-CTAB method after liquid nitrogen freezing and extracted multiple times with organic reagents. Then the precipitate was washed with ethanol and dissolved in TE buffer. The cultured genomic DNA is high-yield, easy to extract, and has good purity, making it suitable for extracting from multiple samples at once. After experiments, the genomic DNA obtained by this method can be used for most basic molecular biology experiments such as PCR and DNA enzyme digestion.

Mycobacterium tuberculosis remains the leading cause of death from a single infectious pathogen globally despite decades of effective chemotherapy. In 2024, an estimated 10·7 million people developed tuberculosis, including approximately 620 000 people living with HIV (PLHIV), and tuberculosis caused an estimated 1·23 million deaths overall, including approximately 150 000 deaths among PLHIV. Men accounted for more than half of the cases, and children represented a substantial burden, reflecting ongoing transmission and diagnostic gaps. Approximately a quarter of the world's population has bee

BACKGROUND: Inadequate access to clinical bacteriology testing in low-resource settings compromises individual patient care and contributes to antimicrobial resistance. Funders, policy makers, and health administrators working to expand bacteriology testing in sub-Saharan Africa require evidence-based guidance. The aim of this study was to assemble a costed consensus list of minimum equipment and consumables required to implement and operate a functional clinical bacteriology laboratory in secondary and tertiary care hospitals in sub-Saharan Africa, to guide funders and health administrators.

Major advances in tuberculosis diagnostics and biomarker research from 2020 to 2025 have informed updated WHO policy guidance, consolidating recommendations for the detection of tuberculosis disease and drug resistance within a single framework. These recommendations align screening, diagnosis, and drug-resistance testing across populations and specimen types. Digital chest radiography, increasingly supported by computer-aided detection, is now a core WHO-recommended screening and triage tool, improving case detection and referral for confirmatory testing at scale. Tuberculosis remains the leading cause of death from a single infectious agent globally. The WHO Global Tuberculosis Report 2025 indicates that progress towards the WHO End TB Strategy targets remains off track despite modest declines in incidence and mortality. In 2024, an estimated 10·7 million people developed tuberculosis and 1·23 million died from the disease. In this Series paper, we examine evidence published between Jan 6, 2020, and Jan 6, 2026, to provide an updated overview of tuberculosis vaccines and tuberculosis preventive treatment (TPT). As of June 4, 2026, 18 tuberculosis vacci

Tuberculosis treatment has undergone its most profound transformation since the launch of the standardised DOTS strategy in 1994. Since 2020, a series of pivotal randomised trials, including TB-PRACTECAL, ZeNix, Nix-TB, endTB, BEAT-TB, SHINE, and Study 31/A5349, have redefined the management of both drug-susceptible and drug-resistant tuberculosis. These studies have enabled shorter, fully oral regimens with improved efficacy and safety across adult and paediatric populations, including people with HIV, and have driven major updates to WHO treatment guidelines. Despite these advances, tubercul
