Thermal proteome profiling identifies new drug targets in Plasmodium falciparum parasites.

Spreading resistance to clinically used antimalarial drugs increases the need for identification of new drug targets. Here, we screened 25 antimalarials with known and unknown mode of action to validate old and find new drug targets in P. falciparum. Combining experimental approach by proteome-wide cellular thermal shift assay and computational filtering by molecular docking, we found the drugs to bind to previously known drug targets, validated ACS10 as the target of MMV665915 and discovered new drug targets. Furthermore, we updated the experimental and analytical MS-CETSA pipeline with the i

Blackwater Fever Complicated by Severe Acute Kidney Injury and a Therapeutic Dilemma: A Case Report from Niger.

INTRODUCTION: Blackwater fever (BWF) is a rare complication of antimalarial treatment characterized by acute intravascular hemolysis. Its diagnosis is mainly clinical, particularly based on the presence of dark-colored urine. CASE PRESENTATION: We report the case of a 50-year-old woman admitted to the Infectious and Tropical Diseases Department of the National Hospital of Zinder for fever, jaundice, and acute kidney injury in the context of persistent Plasmodium falciparum infection and repeated antimalarial treatments, including artesunate. Clinical and laboratory findings were suggestive of

Modeling the potential effect of cabamiquine-pyronaridine for chemoprevention against Plasmodium falciparum malaria.

Preventive treatment (chemoprevention) is a key intervention for reducing the burden of Plasmodium falciparum malaria in children. The impact of the current widely used drug combination, sulfadoxine-pyrimethamine-amodiaquine (SP-AQ), is challenged by emerging drug resistance and adherence to multi-day dosing. Cabamiquine is a novel antimalarial with activity against liver- and blood-stage malaria parasites, administered as a single dose, and under development in combination with pyronaridine. We used mathematical modeling to translate early phase 1 clinical results into estimates of its potent

Clinical contribution of mNGS in the diagnosis and management of imported falciparum malaria in a malaria-free region: a five-case series from northern China.

BACKGROUND: Although malaria transmission has been eliminated in China, imported Plasmodium falciparum malaria remains a diagnostic challenge, particularly in non-endemic regions where clinical experience is limited. The nonspecific manifestations and potential for rapid progression to severe disease may result in delayed diagnosis and inappropriate management. Metagenomic next-generation sequencing (mNGS) has emerged as a complementary diagnostic approach for complex infectious diseases; however, its role in imported malaria management remains insufficiently characterized. METHODS: We retros

Evidence of artemisinin partial resistance in Zambia: a molecular epidemiology and clinical study.

BACKGROUND: Artemisinin derivatives are central to first-line treatment of both uncomplicated and severe Plasmodium falciparum malaria. Emerging artemisinin partial resistance in east Africa threatens to spread across the continent. METHODS: In two cross-sectional studies conducted across the ten provinces of Zambia in 2024, we genotyped the artemisinin resistance-associated gene Pfkelch13. Children aged 6-72 months were recruited from randomly selected households in the first study, and patients 6 months or older presenting with malarial symptoms at 100 selected health facilities were recrui

A cytoskeleton-associated Plasmodium protein critical for organelle segregation.

The malaria parasite, Plasmodium falciparum, replicates asexually within red blood cells by schizogony, a mode of division in which organelles divide multiple times before a final coordinated cytokinesis event, called segmentation, partitions them into dozens of merozoites. The Plasmodium cytoskeleton functions critically during this process, facilitating one-per-cell organelle segregation. Here, we identify and characterize a cytoskeleton-associated protein, PfOGRE (organelle retention, PF3D7_1435600), required for segregation of the mitochondrion and apicoplast, a non-photosynthetic plastid.

Plasmodium falciparum Myosin B is a slow motor optimized for force generation.

Malaria is a disease caused by apicomplexan parasites of the genus Plasmodium . These organisms express two atypical class-XIV myosins: Myosin A (MyoA), a core component of the glideosome expressed throughout the entire lifecycle, and Myosin B (MyoB), which is restricted to invasive stages and localizes to the apical region. Here, we determine the crystal structure of Plasmodium falciparum MyoB (PfMyoB) in the Rigor state and identify its essential light chain as the same subunit bound to MyoA. Combining structural analysis, molecular dynamics simulations and in vitro kinetic and motility assa

A new class of lipid transfer proteins is required for the recycling of lipids from the P. falciparum digestive vacuole.

Malaria parasites endocytose large quantities of hemoglobin from the host erythrocyte, a process critical for parasite survival, leading to extensive membrane internalization. While hemoglobin degradation in the digestive vacuole (DV) is well studied, how the parasite deals with the membranes arriving within the DV is unknown. Here we identified PfTUPA, a previously uncharacterized lipid transfer protein in the DV membrane that is needed for this function. PfTUPA contains a soluble TULIP-like lipid transport domain exposed to the DV lumen and a transmembrane lipid transfer domain of bacterial

Identification of PfSR1-MT as melatonin-responsive GPCR-like candidate from the human malaria parasite Plasmodium falciparum.

The synchronizing effect of melatonin on the malaria parasite Plasmodium falciparum is well documented, yet its receptor remains elusive. In mammals, melatonin signals through seven-transmembrane (7TM) G protein-coupled receptors. Here, we characterized two putative Plasmodium melatonin receptor candidates, PfSR1-MT and PfSR10, expressed during the asexual parasite stage. We show that PfSR1-MT, but not PfSR10, binds 2-[¹²⁵I]iodomelatonin with high affinity (pM) when heterologously expressed in human cells. Molecular modeling and mutagenesis studies identified the melatonin-binding site within

Functional validation of the Plasmodium falciparum K13 C580Y mutation in recently collected Ethiopian isolates.

Recent genomic investigation in Ethiopia identified the first detection of the Plasmodium falciparum Kelch13 (K13) C580Y substitution in the Horn of Africa. To assess its functional impact, we introduced C580Y into two recently collected Ethiopian clinical isolates using CRISPR-Cas9 genome editing. Ring-stage survival assays showed significantly elevated in vitro dihydroartemisinin survival in edited parasites relative to isogenic controls, demonstrating that C580Y confers artemisinin tolerance in contemporary Ethiopian parasite genetic backgrounds.

High prevalence of Plasmodium ovalewallikeri in co-infections with Plasmodium falciparum in symptomatic than asymptomatic Plasmodium parasite infections in Kisumu, Kenya.

Plasmodium falciparum (Pf), Plasmodium ovalewallikeri (Pow), Plasmodium ovalecurtisi (Poc), and Plasmodium malariae (Pm) coexists in Kenya sympatrically. Some infections progress to classical symptoms of malaria prompting hospital visits, while others remain asymptomatic. There is limited information on whether the species composition status of infections in endemic areas influences progression to either asymptomatic or symptomatic infection. This cross-sectional study determined the composition of Plasmodium species among symptomatic versus asymptomatic participants in Kombewa, Kenya. Between

Longitudinal Dynamics of Angiopoietin-1, Angiopoietin-2 and Soluble Thrombomodulin During Recovery from Paediatric Cerebral Malaria in Ghana.

Cerebral malaria (CM) remains a leading contributor to paediatric mortality and long-term neurocognitive sequelae in sub-Saharan Africa, driven by a complex interplay between Plasmodium falciparum sequestration and host endothelial dysfunction. Clinical recovery is typically defined by parasite clearance and neurological improvement; however, the temporal relationship between vascular restoration and symptomatic resolution remains inadequately characterized. This study aimed to delineate the recovery kinetics of angiopoietin imbalance and soluble thrombomodulin (sTM) in CM and determine whethe

Epidemiological and clinical characteristics of imported malaria patients admitted to a specialized hospital: a retrospective study (2021- June 2026).

BACKGROUND: China was certified malaria-free by the World Health Organization in 2021; however, imported malaria remains a significant challenge to maintaining malaria elimination due to increasing international travel and overseas population mobility. Imported malaria cases have been continuously reported in Shandong Province, but detailed clinical characteristics, parasite species distribution, travel history, and treatment patterns among hospitalised malaria patients following elimination remain insufficiently characterised. METHODS: We conducted a retrospective hospital-based observationa

Mobility hinders malaria elimination goals in the Brazilian Amazon.

INTRODUCTION: Human mobility across the Brazilian Amazon-shaped by frontier development, land-use change, mining activity, and climatic shocks-remains a major barrier to malaria elimination. Brazil aims to eliminate Plasmodium falciparum (Pf) by 2030 and Plasmodium vivax (Pv) by 2035, yet malaria incidence has plateaued since 2020. We quantify parasite-specific mobility and assess how importations drive malaria reintroduction and reestablishment in areas approaching elimination. METHODS: We analyzed 5,521,411 de-identified malaria cases from Sivep-Malaria (2003-2023) across 808 municipalities

Geographical Variation in Antimalarial Drug Resistance Marker Prevalence across the Southern African Elimination Eight Region.

Global efforts to control and eliminate malaria are threatened by antimalarial drug resistance. The WHO recommends molecular surveillance of resistance markers as a complementary approach to therapeutic efficacy studies. Here, the first regional analysis of antimalarial resistance markers across six southern African countries spanning diverse transmission intensities and geographies is reported. Dried blood spots, collected from rapid diagnostic test-positive individuals in Angola, Eswatini, Namibia, and Zambia in 2023, Mozambique in 2022, and South Africa between 2022 and 2024 using a standar

Low prevalence of Plasmodium falciparum pfhrp2 and pfhrp3 gene deletions in archived samples collected before and after nationwide malaria-RDT rollout in Kenya.

Rapid and accurate malaria diagnosis is essential for effective treatment and malaria control. In Kenya, most malaria testing relies on P. falciparum histidine-rich protein 2 (PfHRP2)-based rapid diagnostic tests (RDTs). However, deletions in the pfhrp2/pfhrp3 genes can lead to false-negative RDT results, which can sustain transmission and worsen clinical outcomes. This study assessed the prevalence and pattern of pfhrp2, pfhrp3 and their respective flanking genes in archived samples collected before and after the nationwide rollout of PfHRP2-based RDTs in Kenya in 2012. A total of 350 archive

Screening of a chemogenetic library unveils dual-stage antiplasmodials with low cytotoxicity.

The global effort to eradicate malaria requires the development of drugs with novel mechanisms of action and differentiated pharmacological profiles. Both liver and blood phases of Plasmodium infection are essential for disease progression and transmission, making them primary targets for pharmacological intervention. In this study, we established a dual-luciferase reporter assay to simultaneously assess Plasmodium berghei liver-stage development and host cell viability. Screening a library of bioactive small molecules with annotated human targets revealed several inhibitors with potent liver-
