Fast-Acting Chalcogen-Phosphoranes Inhibit Sensitive and Resistant Plasmodium falciparum Strains.

Malaria remains a major global health challenge with growing resistance to current antimalarial drugs. In this study, we report the antiplasmodial activity of eight sulfur-containing chalcogen-phosphorane analogues that show submicromolar potency against Plasmodium falciparum 3D7 strain (IC50s = 0.7-1.9 μM) and reasonable selectivity indexes (SI) over HepG2 and HEK293 cells (SI ≥ 12). Electron-withdrawing substituents on the aromatic ring improved potency by two- to 3-fold. Compound 5, a representative compound of the series, showed fast-acting inhibitory activity that rapidly induced pyknotic

Photopharmacology of Quinoline and Benzimidazole Azobenzene-Based Photoswitchable β‑Hematin Inhibitors.

Recent advances in antimicrobial drug design have incorporated photopharmacology to control drug behavior through light activation, alongside traditional phenotypical strategies including molecular hybridization and metal incorporation. The quinoline and benzimidazole scaffolds are important chemical frameworks identified by the Medicines for Malaria Venture and are known to exert their antiplasmodial activity by inhibiting hemozoin formation, an important target in antimalarial drug design. These scaffolds form part of growing libraries in drug discovery, aimed at developing novel and potent

Serological evidence of continued exposure to Plasmodium falciparum among residents in a near-elimination setting.

BACKGROUND: Malaria transmission has significantly declined in several endemic regions following extensive control efforts, bringing some areas close to elimination status. However, residual exposure to Plasmodium falciparum may persist even when clinical cases become rare. Serological markers provide valuable insights into historical and recent exposure by detecting long-lived antibodies that remain after infection. The use of blood-stage antigens such as PfEBA175 and PfRh5 allows for assessing population-level immunity and identifying potential hotspots of residual transmission. METHODS: A

Alert for Imported Malaria in Non-Endemic Areas: A Case Report of Atypical Falciparum Malaria in a Young Child and Diagnostic Experience.

BACKGROUND: Although China has eliminated indigenous malaria, imported cases, particularly among young and middle-aged workers returning from Africa, constitute a major challenge for current epidemic prevention and control. In contrast, imported malaria in children is extremely rare and often subject to diagnostic delays in non-endemic areas due to atypical clinical presentations. CASE PRESENTATION: We report a case of a 2-year-11-month-old boy who returned from Sudan, a malaria-endemic region, presenting with fever and diarrhea as the initial symptoms. The case was identified by the laborato

The Effect of Artemether-Lumefantrine Combined with a Single Dose of Primaquine on Plasmodium falciparum Gametocyte Clearance and Post-Treatment Infectivity to Anopheles arabiensis.

BACKGROUND: Malaria remains a major public health concern in Africa, due to the persistence of Plasmodium falciparum gametocytes that sustain transmission post treatment. This study evaluated the effects of artemether-lumefantrine (AL) alone compared with AL combined with a single low-dose of primaquine (SLD-PQ) on gametocyte clearance and infectivity to Anopheles arabiensis post treatment. METHODS: A prospective cohort and entomological study were conducted from January to September 2025 in Northwest Ethiopia. Ninety-six microscopically confirmed cases of P. falciparum gametocytemia mono-inf

Targeting Infected Host Cell Heme Metabolism to Kill Malaria Parasites.

Background/Objectives: Malaria remains a major global health burden, increasingly complicated by resistance to artemisinin-based therapies. Because artemisinin activation depends on heme and porphyrin chemistry, we sought to exploit host red blood cell (RBC) heme metabolism as a therapeutic vulnerability. This study aims to develop and evaluate a host-directed "bait-and-kill" strategy that selectively sensitizes malaria-infected RBCs to artemisinin. Methods: We integrated quantitative proteomics, erythropoiesis transcriptomic analyses, flow cytometry, and in vitro malaria culture assays to cha

Juvenile toxicity and developmental safety of Zintrodiazine, a novel antimalarial candidate, in Wistar rats.

OBJECTIVES: Zintrodiazine (ZY-19489) is a novel triaminopyrimidine-class antimalarial with potent activity against Plasmodium falciparum, including resistant strains. To support paediatric development and meet regulatory requirements, we conducted a comprehensive juvenile toxicity study of Zintrodiazine in rats, focusing on growth, neurodevelopment and toxicokinetics. METHODS: Wistar rats were administered vehicle and 30, 45 or 60 mg/kg/day Zintrodiazine orally from postnatal day (PND) 10-45, followed by a 4-week recovery (PND 46-73). Endpoints included clinical observations, neurobehavioural

Draw inspiration from research on the insecticidal, irritant and mosquito-repellent properties of plants used by chimpanzees to build their nests.

BACKGROUND: Vector-borne diseases are still responsible for the deaths of one million people worldwide every year, particularly in African countries. Plans to combat this worldwide burden, including strategies to control vectors, are still being investigated. Among them, the behavior of chimpanzees, our closest relatives living in African forests, has been studied. In Kibale National Park in Uganda, chimpanzees ingest plants that are biologically active against Plasmodium falciparum responsible for malaria but also select tree species to build their nests. The essential oils extracted from the

Chemoproteomics-based profiling elucidates the antimalarial effects of amodiaquine through disruption of glycolysis process in Plasmodium falciparum.

BACKGROUND: Malaria poses a significant global health threat, and 4-aminoquinolines have played a pivotal role in the decades-long fight against malaria. Amodiaquine (AQ), a prominent member of the class, has been employed clinically for decades and, in combination with artesunate (AS), constitutes one of the most widely applied artemisinin-based combination therapies (ACTs). However, the precise molecular targets and antimalarial mechanisms of AQ remain incompletely understood. METHODS: We synthesized an AQ-derived activity probe (AQP) and systematically identified AQ-binding proteins using a

Promiscuous stimulation of HSP70 ATPase activity by parasite-derived J-domains.

The malaria parasite P. falciparum exports a large number of proteins to the human host cell, including members of the J-domain protein (JDP) family. In other systems, JDPs stimulate the ATPase activity of HSP70 and direct client protein interactions. Three exported PfJDP are highly homologous yet appear to have divergent roles. We examined the ability of isolated J-domains from these proteins to stimulate the ATPase activity of PfHSP70-X and human HsHSP70 and HsHSC70. In vitro assays demonstrate that all domains stimulate the ATPase activity of each HSP70 tested. While overall stimulation was

Severe falciparum malaria remains a threat in East Kalimantan: A case report.

The incidence of malaria has decreased significantly in East Kalimantan Province, a tropical forest area in Indonesia since the last 2 decades. Although malaria cases now are rare in most parts of this province, they do not disappear. Less experience with malaria cases could cause misdiagnosis or late diagnosis of malaria and severe malaria, a life-threatening disease. It was reported a severe falciparum malaria in a 31-year-old male patient treated at a hospital in Samarinda, East Kalimantan Province, Indonesia. This patient was admitted to hospital with several complications including acute

Residual antimalarial drug concentrations before treatment in malaria patients in Asia and Africa: A systematic review and meta-analysis depicting its implication for drug pressure.

BACKGROUND: Resistance to antimalarial drugs has been a major hindrance towards malaria control and elimination. Extensive drug use may increase pressure on parasites, thereby promoting the emergence and spread of resistance, particularly when drug levels are below minimum inhibitory concentrations. Therefore, this review aimed to assess the magnitude of pre-treatment with antimalarial drugs among patients attending health facilities. METHODS: Studies reporting residual drug levels among Plasmodium falciparum malaria patients were searched through the Medline, Cochrane Central Register of Con

Identification of serine/arginine-rich splicing factor 1-interacting proteins in Plasmodium berghei.

BACKGROUND: Serine/arginine-rich splicing factor 1 (SR1), an orthologue of hypothetical RNA-binding protein (HRB1) in yeast, is essential to the asexual development of Plasmodium falciparum and P. berghei. However, their interacting proteins in malaria parasites remain unclear. METHODS: To identify SR1-interacting proteins in malaria parasites, transgenic Pb ANKA expressing green fluorescent protein-fused PbSR1 (PBANKA_1232100) was generated and performed immunoprecipitation coupled to mass spectrometry (IP-MS) using the transgenic parasites. To investigate the developmental stage at which Pb

Unusual C-2 selectivity of Grignard additions to N-Boc-protected isatin 3-imines.

A series of N-tert-butoxycarbonyl-protected isatin 3-imines underwent an unexpected C-2 selective addition reaction with Grignard reagents, affording 3-imino-2-phenethylindolin-2-ols. The protecting group plays a role in this observed chemoselectivity, with N-benzyl and N-p-methoxybenzyl-protected isatin 3-imines undergoing C-3 addition under Grignard reaction conditions, affording 3-amino-3-phenethylindolin-2-ones. Both C-2 and C-3 addition products were assessed for antiplasmodial activity in vitro, with four compounds displaying activity in the sub-micromolar range against both drug-sensiti

Low-density asymptomatic parasitemia in southern Zambia does not lead to clinical malaria and is not associated with household transmission: results from a two-year cohort study.

BACKGROUND: In low malaria transmission settings targeting elimination, the World Health Organization recommends a combination of mass (e.g., mass test-and-treat), targeted (e.g., chemoprophylaxis or treatment for travelers), and reactive (e.g., reactive drug administration) strategies. Most of these strategies would not identify and treat individuals with asymptomatic parasitemia. This study was conducted in a pre-elimination setting in Southern Province, Zambia to examine risk factors for asymptomatic parasitemia, its epidemiologic relationship to incident clinical malaria, and evidence of i

Complexity of Infection and Plasmodium falciparum Circumsporozoite Protein Diversity Prior to Malaria Vaccine Implementation in Kaelé Health District, Cameroon, 2022-2023.

Malaria remains a major public health concern in Cameroon, with Plasmodium falciparum responsible for most morbidity and mortality, particularly among children under five. In response to rising cases, Cameroon began implementing the RTS,S/AS01 malaria vaccine in early 2024. Given the vaccine's strain-specific efficacy, understanding antigenic diversity and complexity of infection (COI) is critical for evaluating long-term impact. We analyzed 100 P. falciparum-positive dried blood spots collected in Mapoussere, Kaele Health District (2022-2023). Using the 4CAST amplicon sequencing assay, we tar

Artemisinin resistance threat in Central and West Africa needs holistic action.

Artemisinin partial resistance (ART-R) has now emerged in the Horn, Eastern and Southern Africa. Mutations in the Plasmodium falciparum kelch13 gene, strongly associated with ART-R, are increasingly reported in several Central and West Africa countries. Here, we opine that the emergence and spread of ART-R in Central and West Africa should not be overlooked given public health, clinical and economic consequences. Again, in addition to the recent funding cuts from the United States (US) government, some regions in these countries are affected by armed conflicts that undoubtedly will disrupt mal

Experimental plasmonic sensing of malaria using an aluminum metasurface.

A wide range of methods currently exist for testing the presence of malaria, each with its own advantages and disadvantages. New technologies are urgently needed to develop more effective diagnosis tools to fight and eradicate malaria. Optical biosensors that employ surface plasmon resonance (SPR) techniques are a promising category of devices for detecting malaria biomarkers. One such biomarker is plasmodium lactate dehydrogenase (pLDH), a protein produced during the life cycle of the malaria parasite, which is a metabolic enzyme found in all plasmodium species, including the most widespread
