DNAAF19-RUVBL1/2 complex recruits multiple adaptors to promote dynein arm assembly.

Cilia are microtubule-based organelles essential for sensing, motility, and fluid transport. Motile cilia rely on inner and outer dynein arms, large multiprotein motor complexes, for force generation and the characteristic bending motion required for ciliary beating. Before being incorporated into cilia, these dynein arms are preassembled in the cytoplasm by dynein axonemal assembly factors (DNAAFs). Defects in this assembly pathway often cause the inherited genetic condition primary ciliary dyskinesia (PCD), in which the loss of dynein arms impairs ciliary motility. The molecular mechanisms g

Disseminated and fatal infection by Schizophyllum commune in a harbor seal (Phoca vitulina) kept under human care in Brazil.

BACKGROUND: Schizophyllum commune is an environmental basidiomycete increasingly recognized as an opportunistic pathogen in humans and domestic animals, whereas infections in wildlife remain rarely reported. In marine mammals, fungal infections involving the central nervous system and respiratory tract are uncommon and may pose diagnostic challenges. CASE REPORT: A captive 5-year-old female harbor seal (Phoca vitulina) from Brazil developed progressive neurological abnormalities over approximately 11 months, initially characterized by cervical dystonia and head deviation, followed by respirat

Gastrin and cholecystokinin 2 receptor, and gastrin-releasing peptide receptor in cancer: From pathogenesis to precision medicine.

Gastrin regulates gastric acid secretion and mucosal homeostasis via the cholecystokinin-2 receptor (CCK2R). Aberrant signaling via CCK2R (cholecystokinin family) and the distinct class A GPCR, gastrin-releasing peptide receptor (GRPR; bombesin family), drives oncogenesis across diverse GI and non-GI malignancies through calcium mobilization, MAPK/PI3K activation, EGFR transactivation, and constitutive splice variants like CCK2Ri4sv. Furthermore, precursor forms such as progastrin remodel the tumor microenvironment by interacting with non-CCK binding sites (p78, Annexin A2), converting fibrobl

Airway imaging practices in primary ciliary dyskinesia: a global survey to guide standardized guidelines.

OBJECTIVES: Airway imaging guidelines for primary ciliary dyskinesia (PCD) are currently limited. Given the progressive nature of this disease, standardized imaging protocols, particularly at key clinical milestones such as diagnosis, routine reviews, and exacerbations, may improve monitoring of disease progression, support comparison of outcomes across centers, and provide robust parameters for clinical trials. METHODS: To document current practices, we conducted a global survey of PCD clinicians. An online questionnaire was distributed via international PCD networks, covering imaging use at

Bilateral sensorineural hearing loss in children: causes and long-term progression.

PURPOSE: Childhood hearing loss (CHL) in need of assisting listening devices require a constant surveillance for hearing loss (HL) progression and adjustment of interventions. Despite advances in aetiological investigations, there are still important gaps in understanding causal determinants and progression patterns of CHL. The aim of this study was to analyze the prevalence and yield of aetiological investigations, and to identify causes associated with HL progression in older children. METHODS: This descriptive retrospective study included data from 194 children with moderate-profound senor

Longitudinal Impact of Pseudomonas aeruginosa Versus Haemophilus influenzae Colonization on Lung Function Decline in Children With Primary Ciliary Dyskinesia.

BACKGROUND: The association of Pseudomonas aeruginosa (PsA) and Haemophilus influenzae (HI) colonization with lung function in children with primary ciliary dyskinesia (PCD) remains insufficiently defined. We aimed to compare spirometric parameters between PsA- and HI-colonized children and evaluate longitudinal changes before, at, and after chronic colonization. METHODS: This retrospective cohort included patients aged ≥ 5 years with definite or highly probable PCD and chronic PsA or HI colonization. Chronic colonization was defined using repeated sputum cultures over 12 months. Pulmonary fu

Variants in CEP135 Cause Congenital Microcephaly and Primary Ciliary Dyskinesia.

INTRODUCTION: Primary ciliary dyskinesia (PCD), a disorder of motile ciliary dysfunction causing chronic respiratory infections, may also rarely present with aspects of non-motile ciliary dysfunction, including retinitis, central nervous system malformations, skeletal dysplasia, and sensorineural hearing loss. However, congenital microcephaly has not been previously associated with PCD. METHODS: We identified three children with congenital microcephaly and chronic respiratory disease consistent with PCD. Comprehensive PCD diagnostic testing and whole exome sequencing were performed. Character

Distinct Mitochondrial States Modulate α-Synuclein Toxicity.

Abnormal α-synuclein (α-syn) accumulation and mitochondrial dysfunction are central features of Parkinson's disease (PD), and increasing evidence suggests that these processes are closely connected. Using yeast models to manipulate mitochondrial metabolic states, we investigated how mitochondrial activity may affect α-syn accumulation and toxicity. Under the fermentative growth condition, we found that α-syn localized predominantly to the plasma membrane and was non-toxic. Respiratory growth induced cytoplasmic α-syn accumulation, mitochondrial abnormalities, and marked toxicity dependent on α

Intra-Breath Oscillometry Reveals Phase-Dependent Resistive Abnormalities in Respiratory Mechanics After Severe Covid-19.

Post-COVID syndrome is often associated with persistent respiratory symptoms despite largely unremarkable conventional pulmonary function tests, suggesting the presence of subtle alterations in respiratory mechanics. We hypothesized that phase-resolved analysis of respiratory impedance would reveal dynamic abnormalities not captured by standard endpoint-based measures. In this prospective observational study, survivors of severe COVID-19 requiring veno-venous extracorporeal membrane oxygenation (V-V ECMO) (n = 9) were evaluated two years after recovery and compared with matched healthy control

Current Controversies in the Management of Congenital Lung Malformations: A Report from the Congenital Lung Malformation Study Group.

BACKGROUND: There remains considerable variability in the management of congenital lung malformations (CLMs). The CLM Study Group organized a breakout session at the American Pediatric Surgical Association 2026 Annual Meeting to address current controversies in CLM care. METHODS: An expert panel of pediatric surgeons from the United States facilitated discussions on three controversial topics: 1) the role and utility of fetal MRI in the prenatal diagnostic evaluation, 2) non-operative management of asymptomatic CLMs, and 3) indocyanine green (ICG) as an emerging adjunct to enable lung-sparing

How close are modern broilers to their physiological ceiling? Have genetic, nutritional, and precision technologies reached biological limits? A systems biology perspective through physiological ceiling theory.

This review examines the hypothesis that modern broiler chickens may be approaching a multidimensional physiological limit to further productivity improvement and evaluates the extent to which current evidence supports this proposition. Over the past century, genetic selection, nutritional optimization, health management, and technological innovations have dramatically enhanced growth rate, feed conversion efficiency, and carcass yield. However, these gains have increasingly been accompanied by physiological trade-offs, including skeletal abnormalities, muscle myopathies, metabolic disorders,

β2-microglobulin as a multifaceted biomarker and regulatory molecule in multisystem diseases: A comprehensive review.

β2-microglobulin (B2M), a structural component of major histocompatibility complex class I (MHC-I), is widely expressed on the surface of all nucleated cells, and exists in free form in blood, urine, and cerebrospinal fluid. Under normal physiological conditions, B2M associates noncovalently with the MHC-I heavy chain, thereby supporting proper folding, peptide loading, complex stability, and cell surface expression; the assembled peptide-MHC-I complex is then recognized by CD8+ cytotoxic T cells during immune surveillance. B2M is primarily eliminated through glomerular filtration, proximal tu

Bioabsorbable Stenting in a Case of Severe Pediatric Posterior Glottic Stenosis.

INTRODUCTION: Pediatric patients, especially those under 5 years of age, are at increased risk of laryngeal injury and subsequent posterior glottic stenosis (PGS) after prolonged or repeated intubations. Despite various surgical options, optimal management remains unclear, and restenosis is a common complication. Steroid-eluting stents, such as the Propel™, have shown promise in sinus applications but are underreported in airway stenosis and reconstruction. METHODS: We present a case of a 2-year-old female with transverse myelitis and neuromuscular respiratory failure with bilateral vocal cor

Comparative neuroanatomy of emotion expression: from flies to humans.

Total anomalous pulmonary venous return is a congenital heart malformation characterized by anomalous pulmonary venous inflow to the right atrium. Surgical repair typically occurs during the first month of life, and survival beyond that age in untreated patients is unlikely. We report an extreme case of supracardiac total anomalous pulmonary venous return in an infant who survived 7 months despite atypical anomalous inflow without atrial-level communication and with right-to-left shunting only through a patent ductus arteriosus. We stabilized the patient's left-sided heart function before surg

HDAC1 modulates sepsis-induced immunosuppression by driving the exhaustion of CD8+ T cells.

Sepsis, a systemic inflammatory response to infection, remains a leading cause of mortality in intensive care units, with sepsis-induced immunosuppression being a critical pathophysiological process. In this study, we investigated the role of histone deacetylase 1 (HDAC1) in sepsis-induced CD8+ T cell exhaustion, a key driver of immunosuppression. Clinical analyses of patients with sepsis revealed that reduced peripheral blood lymphocyte levels, particularly CD8+ T cell depletion, strongly correlated with worsened outcomes. In a murine sepsis model, single-cell RNA-Seq revealed a significant d

Surviving Males With PORCN Variants: Expanding the Clinical, Molecular, and Mechanistic Spectrum.

Pathogenic variants in PORCN cause focal dermal hypoplasia (FDH/Goltz syndrome), an X-linked dominant disorder historically considered lethal in males, with milder presentations now recognized as PORCN non-Goltz spectrum (PONGOS). We report three male patients identified by exome sequencing: one with a mosaic de novo variant (c.727C>T; p.Arg243*) showing FDH features, and two siblings with an inherited non-mosaic variant (c.1315T>G; p.Trp439Gly) from their unaffected carrier mother with a PONGOS phenotype. These cases confirm that male survival is possible with both mosaic and non-mosaic PORCN

Longitudinal Electronic Cigarette Exposures Impair Respiratory Function and Structure in the Female Apoe-/- Mouse.

INTRODUCTION: Electronic cigarette (EC) usage amongst naïve users in the United States has been rising for the last decade. The effects of inhaling e-cig aerosols on respiratory health are not fully understood. The goal of this study was to longitudinally track changes in inflammation alongside abnormalities in lung structure and function following 8, 16, and 24 weeks of EC exposure. AIMS AND METHODS: Female Apoe-/- mice were nose-only exposed to either EC aerosols (JUULTM, 3% nicotine, tobacco flavored, 2 puffs/min, particle concentration of 300 mg/m3) or room air for 95 minutes, 5 days/week
