Invasion of erythrocytes by members of the Plasmodium genus is an essential step of the parasite lifecycle, orchestrated by numerous host-parasite interactions. In P. falciparum Rh5, with PfCyRPA, PfRipr, PfCSS, and PfPTRAMP, forms the essential PCRCR complex which binds basigin on the erythrocyte surface. Rh5 is restricted to P. falciparum and its close relatives; however, PTRAMP, CSS and Ripr orthologs are present across the Plasmodium genus. We investigated PTRAMP, CSS and Ripr orthologs from three species to elucidate common features of the complex. Like P. falciparum, PTRAMP and CSS form

BACKGROUND: The relocation of Indonesia's capital to Ibu Kota Nusantara (IKN) in East Kalimantan, a malaria and dengue hotspot, presents new risks of infectious disease transmission due to land-use changes and population movements. Current knowledge on the impact of these changes on vector-borne diseases, especially Plasmodium knowlesi malaria and other arboviruses, is limited. Serological surveillance offers a robust method for assessing population exposure.
METHOD: A community-based cross-sectional study will be conducted in IKN and its surrounding area, in East Kalimantan. Approximately 2,

BACKGROUND: Pyrimethamine resistance in Plasmodium falciparum is driven by mutations at dihydrofolate reductase (DHFR) residues N51I, C59R, S108N/T, and I164L, with parallel substitutions in P. vivax (N50I, S58R, S117N/T, S173L). To evaluate the influence of drug pressure on parasite evolution, we compared the dihydrofolate reductase-thymidylate synthase (dhfr-ts) genes of human and zoonotic malaria parasites exposed to different drug environments.
METHODS: Blood samples were collected from patients with symptomatic malaria in Thailand (P. falciparum, n = 241; P. vivax, n = 115) and from symp

To dissect parasite determinants of clinical disease severity in Plasmodium knowlesi, we performed the first integrative genome-wide association (GWAS), transcriptomic, and expression quantitative trait locus (eQTL) analysis from clinical malaria isolates. We identify distinct parasite programs associated with WHO-defined severe disease, characterized by transcriptional activation of stress-response and host-interaction pathways. In contrast, invasion-linked programs were more strongly associated with parasite burden, while immune-evasion pathways showed overlapping but distinct associations w

INTRODUCTION: Type II NADH: Quinone oxidoreductase (NDH2) represents a critical metabolic target in Plasmodium parasites. Although NDH2 lacks a direct human isoenzyme, it shares significant structural homology with human Apoptosis-Inducing Factor Mitochondrion-associated 1 (AIF-M1), presenting a substantial challenge for selective drug design. To address this selectivity hurdle, the first comparative computational study was conducted across five human-infecting Plasmodium species (P. falciparum, P. knowlesi, P. malariae, P. ovale, and P. vivax) using a natural compound library.

Malaria is one of the major global public health issues. An estimated 282 million malaria cases occurred worldwide in 2024, and the overall prevention and control progress has stagnated or even reversed in some regions. Mass drug administration (MDA), as a potential strategy to accelerate malaria elimination, has regained attention. This paper reviews the evidence base, controversial focuses, and application strategies of MDA in malaria prevention and control. It aims to promote its scientific application in the elimination phase. MDA plays an important role in malaria prevention and control.

Zoonotic malaria caused by Plasmodium knowlesi is an emerging threat in Southeast Asia, where deforestation and habitat fragmentation are reshaping human-wildlife-vector interfaces. While deforestation is known to increase ecological overlap among humans, mosquitoes, and macaques, how changes in host communities and landscape structure jointly influence spillover risk remains unclear. Here, deforestation is represented as edge accumulation using the edge ratio-a continuous measure of forest-non-forest boundary-within a process-based simulation model parameterized with previously reported estim

The polymerase chain reaction (PCR) is widely used to amplify malaria parasite DNA to diagnose Plasmodium infections, but can generate artefactual chimeric sequences through template switching. Here, we examine the occurrence and consequences of PCR-generated chimeras in studies of malaria parasites from humans, macaques and mosquitoes, and highlight two major sources of errors. First, in samples containing mixed-species infections, PCR can produce interspecific chimeras. This problem is particularly acute among closely related species of Plasmodium that infect macaques in southeast Asia, seve

BACKGROUND AND AIMS: Indonesia harbors extraordinary non-human primate (NHP) diversity, yet deforestation, ecotourism, illegal wildlife trade, and urban encroachment are intensifying human-NHP contact, elevating pathogen spillover risk. This review synthesizes evidence on NHP-associated zoonoses within a One Health framework and proposes an integrated biosurveillance roadmap.
METHODS: A structured search of PubMed, Scopus, and Web of Science (2000-2025), informed by the PRISMA-ScR framework and using pre-specified host, geographic, and pathogen search terms, was conducted and supplemented by

BACKGROUND: Zoonotic malaria caused by simian Plasmodium species presents increasing diagnostic challenges in endemic regions.
METHOD: This study developed and evaluated malachite green-based loop-mediated isothermal amplification (MG-LAMP) assays for detecting five zoonotic malaria parasites (P. knowlesi, P. cynomolgi, P. inui, P. coatneyi, and P. fieldi). The assays enable rapid visual detection through a colorimetric change without requiring specialized equipment. To reduce carryover contamination, deoxyuridine triphosphate (dUTP) and uracil-DNA glycosylase (UDG) were incorporated into the

Threats to Malaria Elimination Program (MEP) are arising, such as drug-resistant parasites, insecticide-resistant mosquitos and zoonotic malaria. In Southeast Asia, zoonotic emergence of macaque malaria, Plasmodium knowlesi, has been a major concern to the MEP. This parasite is recently established and potentially localised in wild populations of Macaca fascicularis (long-tailed macaques), which are indigenous to South-East Asia. Besides P. knowlesi infection, the macaque is also infected by P. inui and P. cynomolgi, which could be transmissible to humans. However, the parasitology data from I

Visceral Leishmaniasis (VL), a severe systemic neglected tropical disease (NTD) caused by Leishmania donovani complex protozoa, poses a significant public health threat, particularly in East Africa, where it is fatal if untreated. Somalia is known to be endemic, but the true burden and programmatic challenges are poorly documented due to long-standing conflict, population displacement, and a fragmented, resource-constrained health system. We report the case of a 40-year-old male farmer from Borama, Awdal region (Somaliland), presenting with a 20-day history of intermittent high-grade fever, ch

BACKGROUND: Novel antimalarial combinations are needed to address the threat of emerging resistance to artemisinins. This study evaluated the safety, tolerability, pharmacokinetics, and efficacy of a single oral dose of cabamiquine-pyronaridine for the treatment of uncomplicated Plasmodium falciparum malaria.
METHODS: This two-part, multicentre, open-label, phase 2a study (CAPTURE 1) enrolled patients aged 12-55 years from five hospitals in four countries across Africa with acute uncomplicated P falciparum monoinfection, defined by microscopically confirmed parasitaemia, with parasite densiti

BACKGROUND: Malaria remains a major global health challenge, particularly in endemic regions where severe disease contributes substantially to morbidity and mortality. Among its systemic complications, malaria-associated acute kidney injury (MAKI) is increasingly recognized as a significant yet underappreciated contributor to poor clinical outcomes. Despite advances in antimalarial therapy, MAKI continues to be associated with high mortality and growing evidence suggests a link to long-term renal sequelae, including chronic kidney disease (CKD). This review synthesizes current evidence on the

BACKGROUND: Malaria remains a major public health concern, especially with zoonotic Plasmodium knowlesi presenting specific challenges in Southeast Asia. In Malaysia, Sarawak ranks second in P. knowlesi cases after Sabah. Rural populations in endemic areas experience increased exposure to forest and forest-edge zones where vector mosquitoes breed, thereby raising infection risk and complicating routine surveillance. These groups may carry asymptomatic or submicroscopic infections that act as hidden reservoirs, potentially hindering elimination efforts. This study aimed to assess the prevalence

Neonatal malaria is rare due to protective maternal immunity and breastfeeding, particularly in non-endemic areas of the globe. Plasmodium (P.) knowlesi is a zoonotic species mainly found in Southeast Asia, and very few cases have been reported worldwide in the pediatric population. We report the first documented case of congenital malaria in Bahrain due to Plasmodium knowlesi. A 26-day-old, Pakistani male infant presented with a one-day history of fever, jaundice, lethargy, and hepatosplenomegaly. Laboratory investigations revealed pancytopenia and elevated inflammatory markers. Despite negat
