ObjectiveThe purpose of this study was to investigate the effect of speech therapy on cerebral morphology in patients with non-syndromic cleft palate with or without cleft lip (NSCP ± L) related articulation disorders using structural magnetic resonance imaging technology.DesignA prospective pre-post intervention study.SettingThis study was conducted in the Department of Oral and Maxillofacial Surgery and the Medical Imaging Center of a tertiary hospital.Patients/ParticipantsA total of 24 children with NSCP ± L, aged 6-16 years, who had undergone cleft palate surgery but still presented with a

BACKGROUND: Cleft Palate (CP) refers to hereditary defects characterized by total or incomplete split in the roof of the mouth. CP represents malformations of unknown etiology but similar phenotypes. Besides the restoration of normal appearance and function to the palate, authorities have equally documented facilitation of normal speech as another important reason for cleft palate repair. There is a paucity of data locally on the long term speech outcome post CP repair.
OBJECTIVE: To determine the long term speech outcome post cleft palate repair in Kenya.
METHODOLOGY: African Journals Onlin

BACKGROUND: Stickler syndrome (SS) is the leading cause of hereditary retinal detachment (RD) in children and is characterized by ophthalmic, auditory, orofacial, and articular abnormalities. Vision loss often results from retinal events (RE), including retinal tears and detachments.
PURPOSE: To identify clinical and genetic risk factors for retinal events in patients with SS, to guide screening and preventive care.
METHODS: This retrospective cohort study included 78 patients with clinically or genetically confirmed SS seen at the University Medical Center Utrecht between 2000 and 2019. Pre

OBJECTIVE: This study addresses maxillary deficiency management by developing a deep learning-based system for zygomaticomaxillary suture assessment. As the maturation status serves as a pivotal indicator for optimal maxillary protraction timing, its accurate evaluation is crucial for treatment success. The proposed cone-beam computed tomography-based system overcomes current limitations of subjective manual staging, offering quantitative and reproducible maturation analysis to guide clinically critical decisions in maxillary protraction therapy.
METHODS: A dataset of 505 cone-beam computed t

BACKGROUND: Cleft lip and/or palate (CL/P) is a prevalent congenital craniofacial anomaly that affects facial esthetics, function, and psychosocial well-being. Oral health-related quality of life (OHRQoL) is a critical measure reflecting the impact of oral health on daily functioning and overall life quality. Despite extensive treatment, adults with CL/P may continue to face challenges affecting their OHRQoL.
OBJECTIVE: To systematically evaluate and synthesize the existing evidence on OHRQoL in adults (≥18 years) with CL/P.
MATERIALS AND METHODS: A systematic search was conducted across Sco

PURPOSE: Surgical scarring has long been hypothesized to contribute to maxillary hypoplasia in patients with cleft lip and palate (CLP), often leading to a need for orthognathic surgery (OGS). This study aimed to identify clinical and surgical factors associated with maxillary hypoplasia in skeletally mature patients with CLP using cephalometric analysis.
METHODS: A retrospective review was performed of CLP patients aged ≥16 years with available cone beam CT scans. Cephalometric measurements were obtained using Mimics software. Patient records were reviewed for cleft morphology, number and ty

PURPOSE: Genome sequencing (GS) presents a powerful approach to uncover disease-causing genetic variants. We used GS to examine single vs dual molecular causes in some of the most complicated pediatric cases-those with both a neoplasm and a birth defect.
METHODS: From our pediatric biobank, we selected 1463 children with a major congenital malformation, such as cleft lip/palate or internal organ defect, including 827 cases with a pediatric-onset cancer. The cohort includes nearly 40% non-White and/or multiracial individuals. We implemented GS as a first-tier diagnostic method and hypothesized

BACKGROUND: Cleft lip and palate (CLP) are the most prevalent congenital anomalies, often resulting in impaired maxillofacial complex in all three planes of space and maxillary growth deficiency, leading to aesthetic and functional challenges. Traditional methods like calipers and rulers have limitations in precision, reproducibility, and volumetric capture for soft tissues (ST) and dental arches (DA) in CLP patients. Three-Dimensional Stereophotogrammetry (3DS) offers a non-invasive digital alternative for accurate 3D imaging. The aim of this systematic review of comparative studies was to ev

BACKGROUND: Pierre Robin Sequence is characterised by a small lower jaw, tongue displacement and, often, a U-shaped cleft palate, leading to breathing and feeding problems. Orthodontic airway plates have been developed as a non-invasive treatment option. A systematic review was conducted to evaluate the outcomes of orthodontic airway plates in children with Pierre Robin Sequence.
METHODS: Databases were searched for studies published up to December 2024, in accordance with the Preferred Reporting Items for Systematic Reviews and Meta-Analyses guidelines.
RESULTS: Ten clinical studies with a

X-inactivation is the process of dosage compensation to balance X-linked gene expression levels between females (XX) and males (XY). One of the 2 X-chromosomes is randomly selected for inactivation in each cell during embryogenesis and remains inactive throughout life. Therefore, females have 2 types of cells: those with paternal X-chromosome activation and those with maternal X-chromosome activation. However, it remains unclear how X-inactivation is involved in palate and tooth development. Ofd1 is located on the X-chromosome. We found cleft palate in all heterozygous Ofd1 mutant mice (Ofd1fl

Cleft palate, a prevalent congenital disorder, arises from dysregulated embryonic palatal fusion, but the posttranslational modifications (PTMs) driving this process remain poorly understood. Here, we report that lysine acetylation is a critical MSX1 proteostasis switch that governs embryonic palatal mesenchymal (EPM) cell survival. We demonstrate in vitro and in vivo that MSX1 protein stability regulation by deacetylase SIRT1-catalyzed acetylation underlies EPM apoptosis and palatal fusion. In atRA-induced cleft palate models, SIRT1 suppression drives MSX1 hyperacetylation, accelerating prote

The mouse disorganization (Ds) was described in mutant mice with multiple congenital anomalies arising from the 3 germinative layers. The human homologue of Ds was proposed in a case presenting multiple congenital anomalies associated with the unique feature of a finger-like skin appendage. Since then, several cases have been reported suggesting the occurrence of a disorganization-like (Ds-like) phenotype in humans. Here, we report a case series of 7 patients presenting with the association of orofacial clefts and finger-like skin appendages. These cases, despite the similarities with Ds and a

Attention-deficit hyperactivity disorder (ADHD) is a developmental disorder characterized by inattentiveness, hyperactivity, and impulsivity, often treated with amphetamine and methylphenidate agents. There is limited published data to provide established guidelines for stimulant use during pregnancy, and even less data on particular medications. This case study reports maternal-fetal outcomes with lisdexamfetamine (Vyvanse) use during pregnancy and breastfeeding. The patient who received this medication during her second pregnancy experienced significant improvement in her mental well-being c

Critical-sized craniomaxillofacial (CMF) defects affect the skull, face, and jaw, arising from conditions such as cleft palate, oncologic resections, and high energy impacts, and due to their large size and irregular geometry, cannot heal naturally by the body, thus requiring surgery. The field of biomedical research has long recognized the need to develop higher order biomaterial model systems for improved disease characterization and translational therapeutic/material progress. There is, however, difficulty in developing these workflows at the scale of conventional two-dimensional cell cultu
