Molecular and parasitological evidence of Leishmania (Mundinia) martiniquensis infection in a horse from Brazil.

Leishmania (Mundinia) martiniquensis is an emerging human pathogen with increasing reports of infection in domestic animals. This study describes a naturally infected mare from Brazil with no history of international travel, presenting with persistent nodular cutaneous lesions and cervical lymph node enlargement, without evidence of systemic involvement. Considering the endemicity of cutaneous and visceral leishmaniasis in the study area, as well as previous reports of equine leishmaniasis, infection by Leishmania (Leishmania) infantum was initially suspected. Anti-L. (L.) infantum antibodies

Modelling immune gene expression profiles as pharmacodynamic endpoints of antileishmanial treatment.

BACKGROUND: Cutaneous leishmaniasis (CL), a neglected infectious disease caused by the intracellular protozoan parasite Leishmania, affects over one million people annually. The type and magnitude of the inflammatory response elicited during infection lead to skin-specific immunopathology, resulting in the clinical manifestations of CL. Systemic antileishmanial drugs are the main control measure; however, these are highly toxic, of long duration, and difficult to access for affected populations. New drugs and optimized regimens are urgently needed. Despite the known participation of immune res

First Infant Desensitization to Liposomal Amphotericin B.

Liposomal amphotericin B is the first-line treatment for Visceral leishmaniasis; however, drug hypersensitivity reactions, such as anaphylaxis, can pose significant challenges. Desensitization protocols are an option when alternative treatments are lacking. A 7-month-old female infant presented with persistent fever, pallor, splenomegaly, and laboratory findings suggestive of Visceral leishmaniasis with secondary hemophagocytic lymphohistiocytosis (HLH). After developing anaphylaxis following the first dose of liposomal amphotericin B (3 mg/kg), a 14-step desensitization protocol was successfu

Transfer RNA (tRNA) Genes, Codon Usage and Translational Efficiency in Leishmania infantum.

Background/Objectives: Protozoan parasites of the genus Leishmania are causative agents of a group of devastating human diseases, known as leishmaniasis. These microorganisms possess very unusual mechanisms of gene expression that are poorly understood. This study was aimed at analyzing the tRNA repertoire encoded in the Leishmania infantum genome, a species responsible for the most severe form of disease, visceral leishmaniasis. tRNAs are adaptor molecules aimed at decoding mRNAs into proteins. Results: A total of 92 tRNA genes, dispersed on 38 loci, were identified, often located in regions

Cathepsin G and neutrophil elastase control Leishmania infantum parasitism through TLRs-dependent induction of IFN-α and TNF-α.

Leishmania infantum and Leishmania donovani are protozoan parasites that cause visceral leishmaniasis, a potentially fatal disease targeting the liver, spleen and bone marrow. To survive, these parasites manipulate host macrophage microbicidal responses and metabolic pathways. Here, we demonstrate that the host serine proteases neutrophil elastase (NE) and cathepsin G (CG), acting in concert with Toll-like receptors TLR2, TLR3, and TLR4, induce the inflammatory cytokines IFN-α and TNF-α to control L. infantum infection in macrophages. Conversely, in vivo experiments revealed that while NE and

Evaluation of a commercial real-time PCR assay targeting Leishmania spp. with microscopically positive skin biopsies obtained from Nigerian patients.

BACKGROUND: Although real-time polymerase chain reaction (PCR) assays for leishmaniasis are considered as highly sensitive, their standardization is still ongoing. In the study presented here, the sensitivity of a commercial real-time PCR assay targeting Leishmania spp. was evaluated with Nigerian skin biopsies from patients with microscopic diagnosis of cutaneous leishmaniasis (CL). METHODS: The samples were subjected to two subsequently applied types of nucleic acid extraction and afterwards characterized by two in-house real-time PCR assays targeting kinetoplast DNA (desoxyribonucleic acid

High quality Chromosome-scale Genome Assembly of Phlebotomus perniciosus, a Vector of Zoonotic Leishmaniasis.

Phlebotomus perniciosus is a major sand fly in the Western Mediterranean and a primary vector for Leishmania infantum, causing human and canine visceral leishmaniasis. This species can also transmit phleboviruses, including Toscana virus, associated with meningitis and encephalitis. Despite its importance, genomic resources for P. perniciosus remain limited, with only a low-quality assembly available. A high-quality reference genome is essential to study traits related to vector competence, host preference, and responses to control measures. Here, we present a high-quality de novo assembly usi

Leishmaniasis: pathogenesis, host immunity, and the evolving landscape of immunotherapy and immunochemotherapy.

Leishmaniasis is a group of neglected tropical diseases caused by protozoan parasites of the genus Leishmania, transmitted by the bite of infected female phlebotomine sandflies. Manifesting as cutaneous (CL), mucocutaneous (MCL), and visceral (VL) forms, it remains a major global health burden, affecting millions in tropical and subtropical regions, with an estimated 30,000 new VL cases annually and over 1 billion people at risk. The disease is defined not only by the parasite but by the host immune response: protective Th1-mediated cellular immunity, characterized by IFN-γ, TNF-α, IL-12, and

Profiling fungal secondary metabolites with anti-leishmanial activity: an integrated in silico and in vitro study.

Numerous reports of drug resistance in Leishmania major, the causative agent of cutaneous leishmaniasis (CL), underscore the need to discover novel therapeutics. Fungal secondary metabolites (SMs) are valuable resources for drug development and can serve as antimicrobial agents, enzyme inhibitors, and lead compounds for novel medicines. A three-dimensional model of the L. major sterol 14α-demethylase (LmCYP51) enzyme was built using L. infantum CYP51 as a template. A library of 1,167 compounds was virtually screened against LmCYP51 using high-throughput docking. Eleven promising candidates wer

Autochthonous canine cutaneous leishmaniasis caused by Leishmania (Mundinia) martiniquensis in Thailand: First evidence with successful intralesional amphotericin B treatment and a review of animal infections worldwide.

Autochthonous human leishmaniasis caused by Leishmania (Mundinia) martiniquensis has been increasingly reported in Northern Thailand. However, the potential animal reservoirs for this parasite remain unidentified. Herein, we report the first documented case of canine cutaneous leishmaniasis caused by L. martiniquensis in a dog from Chiang Mai Province. The patient presented with a firm, non-ulcerative nodule (0.3 × 0.4 cm) on the concave surface of the right ear pinna. Fine-needle aspiration cytology revealed protozoan amastigotes, suggestive of Leishmania infection. The parasite was successfu

Anthropogenic landscapes and vector-borne disease dynamics: Unveiling the complex interplay between Human Footprint and disease transmission in Colombia.

Anthropogenic landscape transformations are fundamentally reshaping the epidemiology of vector-borne diseases (VBDs), yet their causal impacts remain poorly quantified across diverse ecological and socio-economic contexts. This study evaluates the causal effect of the Human Footprint (HFP) on the transmission dynamics of malaria, dengue, and visceral leishmaniasis in Colombia. We conducted an ecological analysis using municipal-level retrospective data from Colombia's National Public Health Surveillance System (SIVIGILA, 2007-2019). Epidemiological records were integrated with environmental an
