We report a 43-year-old male farm worker from El Salvador residing in California's Central Valley who presented with 4 months of progressive upper lip swelling, ulcerative lesions on the nose and left forearm, weight loss, night sweats, headaches, and hoarseness of voice. He had severe HIV-related immunosuppression (CD4 count of 81 cells/mm3). Biopsy of a forearm lesion and endobronchial tissue obtained at bronchoscopy revealed intracellular amastigotes consistent with leishmaniasis. Leishmania PCR on endobronchial tissue detected Leishmania donovani/infantum/chagasi complex, and plasma microb

HIV-associated Immune Reconstitution Inflammatory Syndrome (IRIS) may significantly alter the immunopathological presentation of American Tegumentary Leishmaniasis (ATL), occasionally causing paradoxical clinical exacerbations. We report the long-term follow-up of a 39-year-old female coinfected with HIV and disseminated mucocutaneous leishmaniasis caused by Leishmania (Viannia) sp., who experienced severe lesion exacerbation four months after initiating High-Activity Antiretroviral Therapy (HAART). Despite successful viral suppression and CD4+ T-cell recovery, she developed aggressive mucocut

Autochthonous mucocutaneous leishmaniasis caused by Leishmania infantum is an under-recognised but established manifestation of endemic leishmaniasis in Spain. This review identified 65 confirmed autochthonous cases reported since 1990, showing a relatively consistent clinical pattern. Most patients were adults, predominantly men in the fifth or sixth decade of life. Nearly half were immunosuppressed, mainly due to HIV infection, corticosteroid use, biologic therapy, autoimmune diseases, or solid organ transplantation, although immunocompetent individuals were also affected. The laryngeal, nas

BACKGROUND: Leishmaniasis is responsible for approximately 65,000 annual deaths. Various Leishmania species are the predominant cause of visceral, cutaneous, or mucocutaneous leishmaniasis, affecting millions worldwide. The lack of a vaccine, emergence of resistance, and undesirable side effects caused by antileishmanial medications have prompted researchers to look for novel therapeutic approaches to treat this disease. Antimicrobial peptides (AMPs) offer an alternative for promoting the discovery of new drugs.
METHODS: In this study, we detail the synthesis process and investigate the antil

Leishmania (Viannia) braziliensis causes cutaneous and mucocutaneous leishmaniasis. Macrophages are host cells for parasite replication and act as effector cells against the parasite. The two main macrophage phenotypes (M1 and M2) and their polarisation states have been implicated in Leishmania infection despite scarce data on L. (V.) braziliensis. In this study, we investigated the temporal and spatial distribution and predominance of M1 and M2 macrophages during L. (V.) braziliensis infection in Balb/c mice. Animals were infected with L. (V.) braziliensis promastigotes and were monitored for

Cutaneous leishmaniasis (CL), a parasitic disease caused by Leishmania aethiopica, is a major health problem in Ethiopia. It presents mostly as three different clinical forms: localized CL characterized by small lesions that ulcerate; diffuse CL defined by multiple nonulcerating nodules; and mucocutaneous leishmaniasis, where the mucosa of the nose or the mouth is affected. The mechanisms resulting in the development of these different clinical presentation are still poorly understood. Here, we recruited a cohort of CL patients presenting with different forms of CL in northwest Ethiopia as wel

BACKGROUND: Leishmania infantum is the only prevalent Leishmania species in Europe and manifesting predominantly as cutaneous or visceral leishmaniasis, whereas new world species like Leishmania (L.) braziliensis are well known pathogens in mucocutaneous leishmaniasis. Mucosal leishmaniasis caused by L. infantum is a rare clinical condition with only few cases described in literature. In contrast to our case, mostly immunocompromised patients with no history of leishmaniasis are affected.
CASE PRESENTATION: We describe the case of a 77-year-old German male who developed an ulcerous lesion of

BACKGROUND: Leishmaniasis is a zoonotic disease caused by unicellular protozoa of the Leishmania genus. The infection can spread through zoonotic or anthroponotic transmission, depending on the species, with the phlebotomine sandfly serving as the primary vector. Leishmaniasis is endemic in tropical regions of Asia and Africa. While mucocutaneous leishmaniasis is rarely reported in Ethiopia, both the cutaneous and visceral forms of the disease are more commonly seen. The clinical spectrum of Leishmania infection includes visceral leishmaniasis (the most common form), as well as cutaneous, muco

BACKGROUND: Cutaneous leishmaniasis (CL) is a neglected tropical disease with diverse clinical manifestations, ranging from localized CL to severe forms such as diffuse CL and mucocutaneous leishmaniasis. Borderline disseminated CL (BDCL), an intermediate form, is characterized by multiple disseminated lesions and poses unique diagnostic and therapeutic challenges, especially in pediatric patients. This study explores pediatric BDCL to better understand its clinical presentation, diagnostic approaches and treatment outcomes.
METHODS: We report 4 pediatric cases of BDCL from Panama, identified

Leishmaniasis, classified as a neglected tropical disease, exerts its impact on millions globally. Its clinical spectrum encompasses diverse forms, from benign self-resolving skin lesions (cutaneous leishmaniasis) to life-threatening visceral infections (visceral leishmaniasis or kala-azar). This review aims to comprehensively explore the spectrum of the disease as an outcome of often-overlooked parasite variants. Additionally, it addresses the emerging challenges faced in the pursuit towards disease elimination. The evolving landscape of leishmaniasis demands the development of molecular surv

Mucocutaneous leishmaniasis (MCL) is a severe and debilitating progression of cutaneous leishmaniasis (CL) that occurs when the disease spreads to involve mucosal tissues. Contrasting CL, which can often be treated with local therapies, MCL requires aggressive systemic treatment, strict adherence to a 30-day regimen and regular monitoring to prevent recurrence. These requirements highlight the critical need for accurate and rapid early diagnosis to guide effective treatment strategies. However, differentiating between the Leishmania species responsible for MCL and CL remains a significant chal

American cutaneous leishmaniasis (ACL), caused by Leishmania (Leishmania) amazonensis and L. (Viannia) braziliensis, presents a wide spectrum of clinical and immunopathological manifestations, ranging from localized cutaneous leishmaniasis (LCL) to severe forms like anergic diffuse cutaneous (ADCL) and mucocutaneous leishmaniasis (MCL). Despite evidence of the immune response's complexity, the role of inflammasomes in disease severity and parasite persistence remains unclear. We investigated the transcriptomic and immunopathological profiles of inflammasome components in patient lesions across

BACKGROUND: While numerous studies have investigated the role and presence of RNA viruses within Leishmania parasites, the existing literature presents a fragmented view of Leishmania RNA virus (LRV) prevalence in parasites associated with cutaneous leishmaniasis (CL). Therefore, the present study aimed to elucidate the prevalence of LRV in parasites obtained from patients with CL.
METHODS: To achieve this aim, we conducted a systematic literature search across international databases, specifically Web of Science, PubMed, Embase, and Scopus. We extracted relevant studies reporting the prevale

Mucocutaneous leishmaniasis (MCL) is a rare and challenging clinical presentation of tegumentary leishmaniasis, primarily affecting the mucosal surfaces of the upper airway. It is often misdiagnosed due to its non-specific symptoms, which can mimic inflammatory, infectious, or neoplastic conditions. This study reports three autochthonous cases of MCL diagnosed between 2023 and 2024. The patients presented chronic symptoms such as dysphagia, odynophagia, lip edema, and ulcerative mucosal lesions. Diagnosis was established through mucosal biopsies, revealing intracellular Leishmania amastigotes
