Simultaneous Infection With Genetically Distinct Trypanosoma cruzi Isolates Is Associated With Distinct Myocardial Innate Immune Transcriptional Profiles During Acute Infection in Mice.

Innate immune receptors are central to host resistance against Trypanosoma cruzi infection, yet how parasite genetic diversity is associated with these responses remains poorly understood. Here, we characterized acute infection outcomes and myocardial innate immune transcriptional profiles in mice subjected to single and mixed infections with genetically distinct T. cruzi parasites (DTUs TcII, TcIII, and TcVI). Swiss mice were infected with RN2, RN23, Y, or CL-Brener individually or in combination, and parasitemia, survival, and cardiac gene expression were assessed. The Y strain, CL-Brener cl

The landscape of dementia research, diagnosis, treatment, and care in Latin America.

Latin America is undergoing rapid population aging alongside a rising burden of dementia. While the region holds substantial potential for dementia risk reduction, challenges remain, such as delayed diagnoses, limited access to specialized care and biomarker testing, persistent stigma, and deep-rooted structural inequities. To address these gaps and foster regionally informed solutions, the Alzheimer's Association convened the 2025 Alzheimer's Association International Conference (AAIC) Satellite Symposium in Lima, Peru, on May 14-15, in collaboration with the Global Brain Health Institute (GB

Chagas cardiomyopathy.

Chagas cardiomyopathy, caused by chronic infection with Trypanosoma cruzi, is a leading cause of non-ischaemic heart failure in Latin America and is increasingly recognised worldwide due to migration. Its pathogenesis is multifactorial, involving persistent parasitism, immune-mediated myocardial injury, microvascular dysfunction and progressive fibrosis, leading to a heterogeneous clinical spectrum. Four main phenotypes are recognised, including subclinical, arrhythmic, heart failure and thromboembolic, which often overlap and evolve throughout the disease course.Diagnosis relies on serologica

Hemodynamic Response During Different Modalities of Resistance Training in Patients With Cardiovascular Disease: A Systematic Review and Meta-Analysis.

PURPOSE: Resistance training (RT) can improve the quality of life and cardiovascular function in patients with cardiovascular disease (CVD). Its clinical application, however, remains limited by heterogeneous training protocols and insufficiently understood acute hemodynamic responses. This systematic review, therefore, aimed to investigate the hemodynamic response during different modalities of RT in patients with CVD. REVIEW METHODS: A systematic search of PubMed, Embase, and Cochrane Trials Register was conducted. Randomized and nonrandomized clinical trials, comparing the hemodynamic resp

CCL signaling drives T Cell-Macrophage crosstalk in the mouse colon during chronic Trypanosoma cruzi infection.

Chagas disease, caused by Trypanosoma cruzi, progresses through acute and chronic phases and is characterized by parasite persistence in multiple tissues, including the heart, gastrointestinal tract, and skeletal muscle. Digestive Chagas disease (DCD) is associated with pathological alterations in the colon during chronic infection. To characterize the immune landscape underlying this process, we used a murine model of chronic DCD and performed single-cell RNA sequencing of colonic lamina propria cells. Our analysis revealed infiltration of T cells, B cells, and macrophages, with T cells repre

Exploring T. cruzi IMPDH as a promising target through Chagas Box screening and AVN-944 inhibition.

Chagas disease, caused by Trypanosoma cruzi, remains a leading cause of heart failure in Latin America, with current treatments limited to acute-phase efficacy, significant toxicity, and emerging resistance. Inosine monophosphate dehydrogenase (IMPDH) is an essential enzyme in guanine nucleotide salvage pathway and represents a promising alternative target. Here, we combined computational screening, biochemical and cell-based phenotypic assays that support T. cruzi IMPDH (TcIMPDH) as a druggable target and identify repurposing opportunities among clinical-stage inhibitors. Using Tanimoto simil

Kinetics of Biomarkers for Therapeutic Assessment in Swiss Mice Infected with a Virulent Trypanosoma cruzi Strain.

Chagas disease (CD), caused by Trypanosoma cruzi, is a neglected tropical illness affecting 6-8 million people in Latin America. Reaching scholarly consensus on the host response to T. cruzi infection remains a significant challenge, primarily due to substantial heterogeneity in outcomes driven by both the choice of animal model and the infecting parasite's discrete typing unit (DTU). This variability complicates the evaluation and comparison of new therapeutic compounds against existing drugs, namely benznidazole and nifurtimox. This study provides a comprehensive, kinetic, multifaceted chara

Hypercoagulability and vascular proinflammatory activation promote cardiac-cerebral fibrinogenesis in a rodent model of Chagas disease.

Cardiomyopathy and stroke are major complications of Chagas disease (CD). We investigated if hypercoagulability, endothelial cell (EC) dysfunction, and blood stasis cause cardiac-cerebral fibrinogenesis in CD. C57BL/6 mice infected with Trypanosoma cruzi (Tc) were evaluated at acute (AT), indeterminate (IT), and chronic (CT) stages of CD. Mice were given anti-parasite, bicistronic immunogens (BCV or BCVR) to determine the role of Tc in fibrinogenesis. We monitored (1) platelet and coagulation cascade activation; (2) cardiac-cerebral blood vessels injury; (3) proinflammatory/prothrombotic pheno

Accuracy of inflammatory biomarkers in assessing outcomes in Chagas disease: a scoping review.

Chagas disease (CD) is a global health issue with acute and chronic forms that cause severe complications, especially affecting the heart. Inflammatory biomarkers have been proposed as potential predictors of adverse outcomes in CD, however, their clinical utility remains uncertain. We conducted a scoping review of five databases, including observational studies evaluating the diagnostic and/or prognostic accuracy of inflammatory biomarkers in CD. Two reviewers independently screened and extracted data. Of 239 articles screened, 5 met the inclusion criteria. Adiponectin was associated with an

Evaluation of the autonomic nervous system in autochthonous patients from Amazon with acute Chagas disease treated with benznidazole.

Chagas disease, a neglected tropical disease, is endemic in the Amazon region, where oral transmission predominates. Autonomic nervous system (ANS) impairment is a recognized pathophysiological mechanism contributing to disease progression, including Chagas cardiomyopathy. This study aimed to assess ANS function in patients with acute Chagas disease from the Amazon, evaluating responses pre- and post-benznidazole treatment. We included 28 acute-phase patients and 20 healthy controls. Participants underwent comprehensive cardiac evaluations, including 12-lead ECG, echocardiogram, 24-hour Holter

Double negative T cells (CD4-/CD8-) are associated with Trypanosoma cruzi persistence in the mouse colon during chronic Chagas disease.

INTRODUCTION: Chagas disease (CD), caused by the blood borne parasite Trypanosoma cruzi, affects 6-8 million people primarily in Latin America. Chronic CD leads to progressive damage in major organs, particularly the heart and gastrointestinal tract, with significant morbidity and mortality. Despite the gut serving as a key parasite reservoir, the immune response within this tissue during chronic infection remains poorly understood. This study investigates the role of double-negative (DN; CD3+/CD4-/CD8-) T cells in gut immunity during T. cruzi infection. Understanding their function could prov

Androgen-mediated immunosuppression impairs the effectiveness of the immune response during acute Trypanosoma cruzi infection.

Despite advances in our understanding host immune responses to Trypanosoma cruzi, Chagas disease affects an estimated 7 million people globally and continues to pose a substantial public health concern. Gonadal hormones, such as testosterone (TS), are known to modulate immunity and may affect the severity of infections, including those caused by T. cruzi. In this study, we investigated the effects of testosterone on acute experimental T. cruzi infection in male Wistar Hannover rats. Animals were placed into four groups: orchiectomized (OCX), infected, testosterone supplemented, and control. Te

Treatment with low-dose bicistronic immunotherapy mitigates parasite persistence and improves serological and cardiac outcomes in adult mice infected with Trypanosoma cruzi: comparative analysis with benznidazole.

Chagas disease (CD), caused by Trypanosoma cruzi (Tc), remains difficult to treat in adults, as benznidazole (BZN) has limited curative efficacy and notable adverse effects. This study evaluated a newly-designed immunotherapy without (BCV) and with RIG-I adjuvant (BCVR). Adult C57BL/6 mice were infected and treated with BCV/BCVR (0.5 mg/kg, two doses, 21-day interval) or BZN (50 mg/kg for 6-weeks). Outcomes included parasite burden, biomarkers of inflammation, fibrosis, and heart disease, and left ventricular (LV) function. Infected/untreated mice developed high acute parasite burden, and pers

CD73 activity controls cytotoxic CD4 T-cell response driving myocardial pathology in chronic Chagas disease.

Chagas disease, caused by Trypanosoma cruzi, is the major cause of infectious cardiopathology worldwide. Although cytotoxic CD4 T-cells (CD4 CTLs) have recently been recognized as crucial effectors in infections and inflammation, the mechanisms that control their differentiation and impact on pathological outcomes remain largely undefined. Here, we demonstrate that the ectonucleotidase CD73, which generates adenosine from extracellular AMP, acts as a key immunoregulator of CD4 CTL response during T. cruzi infection. Using murine models, we found that infection induced a robust expansion of CD4
