Bi-allelic EPHA2 mutations cause dysosteosclerosis by inhibiting osteoclastogenesis and enhancing osteoblastogenesis.

Erythropoietin-producing hepatocellular (EPH) receptors, the largest receptor tyrosine kinase family, have been shown to play a crucial role in normal physiological processes and pathologies. EPHA2, encoded by EPHA2 gene, a member of the EPH receptor family, has been implicated in cataracts and various bone tumors, including osteosarcoma, chondrosarcoma, and bone metastases of breast cancer. However, no association between the EPHA2 gene and genetic skeletal disorders has been reported to date. Here, we identified a Chinese family with a diagnosis of dysosteosclerosis, presenting with signific

Perioperative Anesthetic Considerations for Temporal Bone Tumor Resection in Fibroblast Growth Factor 23 (FGF23)-Related Tumor-Induced Osteomalacia: A Case Report.

Phosphaturic mesenchymal tumors (PMTs) are uncommon, predominantly benign neoplasms frequently associated with dysregulated phosphate metabolism through the overproduction of fibroblast growth factor 23 (FGF23). Elevated levels of FGF23 lead to phosphate depletion, which may impair tissue oxygen delivery, central respiratory drive, and respiratory muscle function. Systemic manifestations may include osteomalacia and neuromuscular dysfunction, with an increased risk of respiratory and cardiovascular complications. PMTs often present with nonspecific clinical findings and may remain unrecognized

BBS10-related Bardet-Biedl syndrome with an atypical presentation of diabetes mellitus: A case report.

Bardet-Biedl syndrome (BBS) is a rare, autosomal recessive ciliopathy characterized by extensive clinical heterogeneity, which often complicates its diagnosis. The present study reported the case of a 31-year-old woman who presented with longstanding, poorly controlled type 2 diabetes mellitus and its associated complications. The patient exhibited classic features including Class I obesity, retinal dystrophy, postaxial polydactyly, renal dysfunction and hypergonadotropic hypogonadism. A definitive diagnosis was established through whole-exome sequencing, which identified compound heterozygous

A KIAA0586 c.1815G>A Founder Allele Associated with Short-Rib Polydactyly Syndrome Type IV in Romani Families.

BACKGROUND: Short-rib polydactyly syndrome type IV (Beemer-Langer syndrome) is a lethal skeletal ciliopathy that can result from biallelic pathogenic variants in KIAA0586 (TALPID3). Earlier clinical observations suggested an unusually high occurrence of this phenotype in Romani families from the Carpathian Basin. METHODS: Whole-exome sequencing was performed in affected individuals from five independent families, and the frequency of KIAA0586 c.1815G>A was assessed in a Hungarian Romani genetic database of 500 individuals. RESULTS: The affected individuals were homozygous for c.1815G>A, a sp

Bentall Procedure in a Patient with Loeys-Dietz Syndrome: A Case Report.

Loeys-Dietz syndrome (LDS) is a rare autosomal dominant connective tissue disorder characterized by aggressive aortopathy, craniofacial anomalies and systemic involvement. Rapid aortic root dilatation and valvular dysfunction mandate an early surgical intervention. We report a case of 15-year-old boy with LDS, rapidly progressive aortic root dilatation and severe aortic regurgitation (AR) who underwent successful Bentall procedure. The case highlights the need of early detection, multidisciplinary involvement and meticulous anesthetic care.

Intraoperative and radiographic indicators of extraocular muscle aberrancy in syndromic craniosynostosis.

BACKGROUND: Strabismus is common in syndromic craniosynostosis, often attributed to abnormal extraocular muscle (EOM) trajectory, and orbital anatomy. Absent EOMs have been reported, but the spectrum of primary dysgenesis remains poorly understood. The purpose of this study was to characterize the range of EOM dysgenesis in a large cohort that had undergone strabismus surgery. METHODS: The medical records of patients with syndromic craniosynostosis treated at Boston Children's Hospital (2010-2023) were reviewed retrospectively to identify all cases with neuroimaging and history of strabismus

Blood Transfusion Rates in Non-syndromic Craniosynostosis: A Proportion Meta-analysis of Open Versus Minimally Invasive Techniques.

OBJECTIVES: Premature suture fusion in non-syndromic craniosynostosis often requires surgery to permit normal brain growth. To minimize blood transfusion requirements, minimally invasive surgery (MIS) has been proposed. This study will pool the prevalence of blood transfusion in non-syndromic craniosynostosis surgeries, comparing open surgery (OS) with MISs. METHODS: A systematic review and proportion meta-analysis was conducted in accordance with Preferred Reporting Items for Systematic Reviews and Meta-Analysis guidelines and registered in PROSPERO (CRD42024604632). FAP adipogenesis drives early spinal muscle loss in dysferlinopathy and reveals a therapeutic target.

Dysferlinopathy (Limb Girdle Muscular Dystrophy R2/LGMD2B) causes progressive weakness of limb-girdle muscles. Although gait abnormalities are common, the contribution of spinal muscles to disease symptoms remains poorly understood. Using cross-sectional MRI in patients and longitudinal analyses in a dysferlinopathic mouse model, we identify early-onset, progressive adipogenic degeneration of spinal muscles as a previously unrecognized hallmark of dysferlinopathy. Multiple spinal muscles, including the iliocostalis, longissimus, and spinalis, are affected, with degeneration preceding pathology

Witteveen-Kolk Syndrome: Two Patients With Craniofacial Features and Novel SIN3A Variants.

Witteveen-Kolk syndrome (WKS) is a rare autosomal dominant neurodevelopmental disorder caused by pathogenic SIN3A variants, with diagnosis complicated by variable physical findings. We describe two unrelated patients with de novo SIN3A variants identified through trio whole exome sequencing, expanding the phenotype of WKS. Patient 1, a 16-year-old female, presented with bilateral cleft lip and palate, retained primary teeth, and horseshoe kidney: findings not previously reported in WKS. Patient 2, a 5-year-old male, exhibited macrocephaly and obesity, also novel to the WKS phenotype. These fin

PEEK Pectus Bar: Evaluation of a New Material for Pectus Excavatum Repair in Children and Teenagers.

The Nuss procedure for Pectus Excavatum repair has relied on metallic bars for the past 3 decades. The technical efforts have focused on the improvement of stability and the reduction of the associated risks. However, some challenges still remain, like metal ion release, thoracic wall mechanic behavior, and the need for implant remotion. In other areas of surgery, polyetheretherketone (PEEK), a polymer that allows 3D printing, has shown mechanical properties closer to human bone and excellent biocompatibility, becoming an alternative material for medical implants. Herein, we present the mechan

Foot Progression Angle Modulates Knee Loading During Walking in Individuals with Flexible Flatfoot.

PURPOSE: This study evaluated tibiofemoral loading and medial meniscal stress distribution in individuals with flexible flatfoot (FFF) during walking under different foot progression angle (FPA) conditions. METHODS: This study analyzed the gait of 28 FFF patients (16 males, 12 females) under three FPA conditions (neutral, toe-in, toe-out). Kinematic (Vicon) and kinetic (Kistler) data were used to estimate tibiofemoral forces in OpenSim. Subsequently, joint angles and muscle forces at peak tibiofemoral forces were used to drive a finite element (FE) model of the knee, enabling the comparison o

Correlation Between Radiographic Parameters and Clinical Range of Motion in Slipped Capital Femoral Epiphysis.

BACKGROUND: Surgical procedures that improve motion for slipped capital femoral epiphysis (SCFE) deformity are typically guided by radiographic imaging. However, the specific aspects of the deformity that have the greatest impact on hip functional biomechanics remain unclear. This study investigated the relationship between radiographic parameters and clinical ROM measurements in patients with SCFE to better elucidate which elements of SCFE pathoanatomy have the greatest effect on joint mobility. METHODS: Patients who underwent in situ pinning for stable SCFE at a single institution between 2

The association between collagenopathies and the risk of spontaneous preterm birth: A systematic review with selective meta-analysis.

INTRODUCTION: Spontaneous preterm birth (sPTB) remains a leading cause of neonatal morbidity and mortality. Inherited collagenopathies, including Ehlers-Danlos syndrome (EDS) and Marfan syndrome (MFS), may predispose to sPTB through impaired connective tissue integrity; however, the available evidence has not been systematically synthesized according to collagenopathy subtype. We aimed to evaluate the association between inherited collagenopathies and sPTB through a systematic review, undertaking quantitative synthesis only where clinically appropriate and methodologically comparable. Morphological and behavioural predictors of motor laterality in domestic horses: A cross-sectional field study.

Motor laterality in horses affects performance, training, and welfare. A simple morphological marker for laterality would help riders, trainers, and veterinarians identify likely locomotor asymmetries early. This study tested whether observable morphological and behavioural characteristics were associated with rider-perceived motor laterality in domestic horses, using a cross-sectional observational field study design conducted in the Blida and Algiers regions of Algeria. The study included 100 regularly ridden domestic horses. Mane orientation, facial whorl characteristics, standing curvature
