Comparison of Serological Detection Methods for Toxoplasma gondii Antibodies and Seroprevalence in Captive Red Pandas.

Toxoplasma gondii is a globally distributed zoonotic parasite that infects a wide range of warm-blooded animals and poses serious threats to many endangered species. Effective serological detection is critical for disease surveillance and control, yet the accuracy of commercial assays varies significantly across host species. Validated serological methods for the Chinese red panda (Ailurus styani) remain scarce, limiting effective monitoring and conservation efforts. A total of 57 serum samples were collected from captive red pandas. Three commercial serological kits-the modified agglutination

Anti-Toxoplasma activity of NBMPR is mediated through the inhibition of nucleoside and nucleobase transporters.

Toxoplasma gondii stands among nature’s most successful eukaryotic pathogens, a status achieved through refined mechanisms for intracellular survival that enable pan-vertebrate host tropism. This ecological dominance derives principally from evolutionary refinements in metabolic plasticity, a modular integration of de novo biosynthesis with high-efficiency nutrient scavenging systems. The pyrimidine biosynthesis pathway is indispensable for robust parasite proliferation. Our previous work demonstrated that exogenous uracil supplementation bypasses the DHO deficiency-induced growth restriction.

Role of wild birds in the circulation of Toxoplasma gondii in Southern Italy: molecular and epidemiological insights.

INTRODUCTION: Toxoplasmosis is caused by the protozoal parasite Toxoplasma gondii and can be acquired through the consumption of food and water contaminated with sporulated oocysts and tissue cysts. The parasite is characterized by high host flexibility, being able to infect all warm-blooded animals, including birds and mammals. Wild birds are considered important reservoirs of infectious agents, some of which possess zoonotic potential. To date, few data are available on the role of these animals in the transmission of T. gondii, especially in the Italian Peninsula. METHODS: To address this

Comparison of the efficacy of cell wall disruption methods for Saccharomyces cerevisiae INVSc1 cells expressing Toxoplasma gondii recombinant ROP6 protein.

Toxoplasma gondii infects both humans and animals, causing severe clinical manifestations. Due to the lack of a safe vaccine, the development of a safe and effective vaccine against toxoplasmosis remains a significant need. Saccharomyces cerevisiae is a well-established recombinant protein expression system with Generally Recognized as Safe status. It supports post-translational glycosylation and is employed in several licensed vaccines, including hepatitis B (Engerix-B), human papillomavirus (Gardasil), and malaria (Mosquirix). Recombinant T. gondii proteins expressed in S. cerevisiae have be

Minimum inoculum of resistance assay for evaluating for anti-toxoplasmosis compounds that target phenyl alanine tRNA synthetase.

Toxoplasma gondii is a globally important intracellular parasite, and treatment regimens are limited by the failure of drugs to target latent tissue cysts. Developing new candidates for treatment also needs to address the potential for resistance to arise. Here, we developed a Minimum Inoculum for Resistance (MIR) assay as a quantitative metric for evaluating inhibitors of T. gondii. The MIR assay, adapted from assays used in malaria drug discovery, measures the frequency for pre-existing resistance alleles by exposing different sized parasite populations to drug pressure. We profiled a series

Host-directed therapeutic strategies against apicomplexan parasites: targeting purinergic P2 receptors.

Apicomplexan parasites establish intracellular infections that profoundly alter host cell physiology and elicit complex immune responses. The long-standing coevolution between these parasites and vertebrate hosts has resulted in extensive overlap between parasite and host metabolic pathways, limiting the feasibility of conventional parasite-centered therapeutic approaches. Increasing evidence indicates that host-derived signals generated during infection play a decisive role in shaping parasite survival and dissemination. Among these signals, extracellular nucleotides released in response to c

Spinal Cord Toxoplasmosis: Mapping the Journey of a Rare Entity Through a Case Report and Review of the Literature.

Toxoplasmosis remains the most frequent cause of cerebral lesions in patients with acquired immunodeficiency syndrome (AIDS), especially in those not receiving prophylaxis. Medullary involvement, although rare, can cause irreversible neurological damage. When associated with fever in the returning traveler, the etiological diagnosis of spinal cord lesions can be challenging due to the wide range of diagnostic possibilities. We report a unique case of spinal cord toxoplasmosis associated with Salmonella non-typhi bacteremia after a trip to Cameroon, revealing an advanced human immunodeficiency

Legacy 4(1 H )-quinolone scaffolds activity against acute and chronic Toxoplasma gondii infection.

Toxoplasma gondii is a protozoan parasite capable of infecting most warm-blooded animals, including humans, and can cause severe disease in immunocompromised individuals and the developing fetus. Current treatments for toxoplasmosis are effective only against the acute stage of infection and have limited or no activity against the latent bradyzoite stage found within tissue cysts. The mitochondrion of T. gondii is a validated drug target, and the clinically used drug atovaquone acts by inhibiting the mitochondrial electron transport chain (ETC) at the coenzyme Q:cytochrome c oxidoreductase ( b

Early Warning Signs, Effects, Risk Factors, and Diagnostic Indicators of Toxoplasmosis in Pregnant Women in Africa: A Scoping Review.

Toxoplasmosis is a widely distributed zoonosis caused by the protozoan parasite Toxoplasma gondii. Infection during pregnancy is a major public health concern due to its potential impact on both maternal health and fetal development. Early detection of maternal infection is critical to prevent adverse outcomes; however, maternal signs are often subtle, non-specific or absent, complicating timely diagnosis. This scoping review aimed to map and synthesise existing evidence on early maternal signs, pregnancy and foetal outcomes, frequently assessed risk factors, and diagnostic approaches of toxop

Toxoplasma gondii hijacks host S100A6 to enhance cell migration and brain cyst formation.

Toxoplasma gondii (T. gondii) has the ability to disseminate widely in the host, including crossing the blood brain barrier (BBB) to establish persistent brain cysts. However, the molecular mechanism by which T. gondii hijacks host immune cell migration to facilitate its own dissemination remain incompletely understood. Here we show that T. gondii infection upregulates S100A6 via a calcium and P65-dependent pathway; This elevated S100A6 expression promoted its interaction with filamin A, leading to promoted cytoskeletal reorganization, enhanced cell migration and T. gondii replication. Synergy between upregulated small GTPase immunity-associated proteins and lysosomal ATP6V1D in restricting intracellular Toxoplasma growth.

Toxoplasma gondii is an obligate intracellular zoonotic protozoan parasite that can cause severe neonatal disease in congenitally acquired infections and severe opportunistic illnesses in immunocompromised individuals. Intracellularly, T. gondii evades host cell defense mechanisms by residing within a specialized non-fusogenic membranous compartment known as the parasitophorous vacuole (PV). Notwithstanding this, we have previously shown that upregulation of small GTPase immunity-associated proteins (GIMAPs) during T. gondii infection promotes fusion of lysosomes with the PV membrane (PVM), re
