Toxoplasma gondii is a globally important zoonotic parasite, with domestic cats serving as the definitive host by shedding oocysts into the environment, where they undergo sporulation, a process that increases the resistance of the oocyst wall and facilitates long-term environmental persistence. In densely populated urban environments, free-roaming cat populations may contribute to environmental contamination and pose a potential public health risk. Here, we assessed the presence of T. gondii DNA in fecal samples from stray cats residing in temple communities across Bangkok, Thailand, using mo

Toxoplasma gondii establishes infection in hosts by deploying effector proteins that reprogram cytokine-driven immunity. While protozoan parasite-encoded macrophage migration inhibitory factor (MIF) homologs exemplify cytokine mimicry, whether T. gondii targets additional host cytokine axes remains unclear. Here, we identify a T. gondii 3-ketoacyl-CoA reductase (KCR) as a noncanonical cytokine modulator that engages the granulocyte-macrophage colony-stimulating factor (CSF2) receptor alpha chain (CSF2Rα). In HEK293T cells, forward and reverse co-immunoprecipitation (Co-IP) assays demonstrated

Toxoplasma gondii, an apicomplexan parasite, relies heavily on epigenetic regulation of gene expression, which is controlled by chromatin-modifying enzymes and histone acetylation, to invade and establish infection in the host. Bromodomain proteins are important epigenetic regulators in parasites, acting as "readers" of histone lysine acetylation to control gene expression. While many bromodomain proteins are unique to parasites, only a few have been explored for therapeutic applications. In this study, we characterized the parasite-specific bromodomain protein TgBDP4 as a key epigenetic reade

Microglia represent one of the mechanisms by which the parasite enters in the brain and contributes to inflammatory responses during infection. P2X7 receptor, NLRP3 inflammasome, and other inflammatory markers are elevated in toxoplasmosis, while resveratrol has demonstrated anti-inflammatory and antiparasitic potential. This study investigated resveratrol on the P2X7/NLRP3 pathway (in vitro) and explored potential interactions with ROP18 and AMA1 proteins of Toxoplasma gondii (in silico). BV-2 microglial cells were exposed to different RH strain parasite-to-cell ratios and infection times to

Toxoplasmosis is a disease of worldwide distribution, causing high morbidity and mortality in humans, as well as heavily impacting animal health and the economy. Toxoplasma gondii, the causative agent, is an intracellular parasite with a complex life cycle whose completion entails asexual, pre-sexual, and sexual stage conversions. Pre-sexual and sexual differentiation take place only within the intestinal epithelium of felines. Recently, several transcriptional factors and epigenetic components crucial to trigger parasite stage transitions within the cat have been identified, allowing, through

BACKGROUND: Toxoplasmosis is a globally prevalent zoonosis caused by Toxoplasma gondii (T. gondii), a parasite that infects nearly all warm-blooded animals, including humans, cats, and pigs. Beyond incurring substantial economic losses to the swine industry, T. gondii infection poses a severe threat to public health, as pigs, which are key intermediate hosts for T. gondii transmission, serve as a major source of human infection via the food chain. Thus, developing specific, sensitive, rapid, and easy-to-perform detection methods for porcine T. gondii is of paramount importance for the preventi

Toxoplasma gondii, the causative agent of toxoplasmosis, a disease widely distributed, is an intracellular parasite that invades host cells of different tissues using specialized endocytic activity. Recent studies suggest that tunneling nanotubes (TNTs), thin cell-surface projections, may participate in the parasite-host cell interaction. Here we report results that suggest the involvement of host-cell TNTs in the adhesion of T. gondii tachyzoites to epithelial LLC-MK2 cells. Microscopy analysis showed that incubating cells in a medium containing 0.45 M sucrose induces reversible assembly of T

Toxoplasma gondii is an obligate intracellular protozoan parasite that causes toxoplasmosis, posing a significant threat to human health and livestock production worldwide. Although monovalent DNA or mRNA vaccines often confer only partial protection, whether these platforms can be effectively integrated into a heterologous prime-boost regimen against T. gondii remains to be fully elucidated. Here, we constructed GRA35-encoding DNA and mRNA vaccines and evaluated their immunogenicity and protective efficacy, administered either alone or in heterologous prime-boost combinations, in C57BL/6 and

Toxoplasma gondii is an obligate intracellular parasite of the phylum Apicomplexa and the causative agent of toxoplasmosis, a disease with a worldwide distribution that causes serious consequences in immunocompromised patients and during pregnancy. Current pharmacological treatments have significant limitations, including their toxicity, lack of efficacy against the chronic phase of the parasite, and low selectivity, highlighting the need to develop new therapeutic targets. One of the most promising targets is the fatty acid synthesis pathway II (FASII), a metabolic pathway located in the para

Toxoplasma gondii is an infectious disease that infects nearly one third of the world's population and endangers the health of immunocompromised people, pregnant women, and livestock. There are no existing drugs able to treat the infection and prevent tissue cysts from developing. Therefore, the creation of a safe vaccine should be considered as a matter of great importance. While many of the vaccines have not proven successful in the past, recently created live-attenuated vaccines (LAVs) using CRISPR-Cas9 gene editing technology were able to outperform the previous ones. The current review ai

Hemophagocytic lymphohistiocytosis (HLH) is a serious medical disorder characterized by uncontrolled activation of the immune system, leading to a cytokine storm. It can be triggered by infection in patients with genetic predisposition, immunosuppression, or malignancy. Disseminated Toxoplasma gondii infection is a rare but recognized trigger in solid organ transplant recipients, particularly in the presence of donor-recipient serologic mismatch. HLH presents with non-specific clinical, systemic, and laboratory abnormalities. We present a case of a 42-year-old man, with a history of kidney tra

Toxoplasma gondii is a globally important intracellular parasite, and treatment regimens are limited by the failure of drugs to target latent tissue cysts. Developing new candidates for treatment also needs to address the potential for resistance to arise. Herein, we developed a minimum inoculum for resistance assay as a semiquantitative metric for evaluating inhibitors of T. gondii. The resistance assay, adapted from malaria, measures the frequency of pre-existing resistance alleles by exposing different-sized parasite populations to drug pressure. We profiled a series of bicyclic pyrrolidone

This study employed a fused hybrid strategy for the structural modification of diosgenin. Diosgenin-cinnamate hybrids (1-13) were synthesised via Steglich esterification and evaluated for anti-Toxoplasma gondii activity and IEC-6 cytotoxicity. Compounds 1-7 exhibited IC50 values of 334-891 µM against the parasite and 261-979 µM against host cells, whereas 8-13 and diosgenin were inactive (IC50 >1000 µM). Compared to spiramycin (SI = 0.97), compounds 1, 3, 5, 6 and 7 showed selectivity indices (SI) of 1.07-1.19, with the para-nitro derivative 5 having the highest SI (1.19) and the lowest host c
