Trigonocephaly-associated hypotelorism: an assessment of orbital shapes and dimensions.

Trigonocephaly occurs due to the premature fusion of the metopic suture. The aim of this study was to document the orbital morphometry of patients with trigonocephaly compared to controls, and to classify the different morphological appearances of the orbits in trigonocephaly. Preoperative two-dimensional computed tomography scans of 15 patients with non-syndromic isolated trigonocephaly (13 male, two female; median age 11 months) and eight controls (four male, four female; median age 22 months) were digitized and analysed. Orbital height, width, and surface area (left and right) and inter-orb

Navigating challenges: a child with Apert syndrome and global developmental delay from a low-income family.

Apert syndrome is a rare congenital disorder characterised by craniosynostosis, syndactyly and distinct facial dysmorphisms. We found a child with Apert syndrome from an economically disadvantaged family residing in an urban slum during a routine home visit by the All India Institute of Medical Sciences Bhubaneswar Extended Health Clinic. The child presented with global developmental delay, unvaccinated status and severe malnutrition, manifesting as underweight and stunted growth. Despite being eligible for free healthcare services under the Rashtriya Bal Swasthya Karyakram, the child remained

Fzd7 Restrains Pink1-Dependent Mitophagy in Suture Stem Cells to Maintain Cranial Suture Patency.

Craniosynostosis results from premature fusion of the cranial sutures, yet the contribution of suture mesenchymal stem cell (SuSC) dysfunction to this process remains incompletely understood. Here, we combined single-cell RNA-seq with 2-µm Visium HD spatial transcriptomics to define stage-specific changes in Prrx1+ SuSCs in the Fgfr2C342Y/+ mouse model. We observed early downregulation of Frizzled-7 (Fzd7), increased mitophagy-associated signatures and readouts with elevated Pink1 expression, and premature osteogenic activation within the SuSC niche. In vitro, loss of Fzd7 increased mitophagy

Subarachnoid space in infancy and early childhood: a systematic review of measurements.

BACKGROUND: Benign enlargement of the subarachnoid space (BESS) is a condition characterized by widened subarachnoid spaces, typically associated with macrocephaly or increasing head circumference during infancy or early childhood. Often described as idiopathic and self-limiting, BESS has also been associated with developmental delays and spontaneous subdural collections. Yet, complete interrogation of these associations is limited by lack of normative subarachnoid space characterization and standardized diagnostic criteria. OBJECTIVE: This project aimed to review and synthesize literature on

Novel IL11RA Compound Heterozygous Variants in a Chinese Pediatric Patient With Pancraniosynostosis.

Craniosynostosis, marked by premature fusion of the cranial sutures, leads to abnormal head shapes and possible neurological complications. This report describes a 4-year-and-2-month-old boy diagnosed with pancraniosynostosis who presented with progressive forehead protrusion 11 months after initial cranial suture reconstruction. Whole exome sequencing identified 2 novel compound heterozygous variants in the IL11RA gene (c.673C>T, P. Arg225Trp inherited from the mother and c.728C>G, P. Pro243Arg inherited from the father), both classified as variants of uncertain significance. The patient unde

Longitudinal Neurodevelopmental and Adaptive Functioning in Infants and Toddlers With Multiple Suture or Syndromic Craniosynostosis.

Given the low incidence of multisuture and syndromic craniosynostosis (CS), information about development over time is limited for these patients. This longitudinal prospective study included neurodevelopmental evaluations with the Bayley Scales of Infant and Toddler Development-Third Edition (Bayley-III) at 1 to 4 time points: preoperatively, 6 months and 18 months postoperatively, and at age 36 months. Caregivers also reported on general adaptive functioning, developmental milestones, and therapies received. Scores were described at each time point and compared with test norms. Pearson corre

A Case of FBN1-Related Early Onset Marfan Syndrome with Multi-Suture Craniosynostosis.

Early-onset Marfan Syndrome (eoMFS) is caused by pathogenic variants in the fibrillin-1 (FBN1) gene. Typical clinical findings include a prematurely aged appearance, severe atrioventricular valve dysfunction (mitral/tricuspid valve insufficiency), and skeletal findings-primarily arachnodactyly, multiple joint contractures, and pectus deformity. Craniosynostosis has been reported in rare cases of eoMFS, but not in association with the c.3037G>A, p.Gly1013Arg variant in FBN1. Here we present a case of a 2-month-old male with bilateral lambdoidal and sagittal craniosynostosis diagnosed with eoMFS

Evaluating the utility of a novel 3D mobile imaging application in the surgical management of craniosynostosis.

PURPOSE: Craniosynostosis is one of the most common craniofacial defects. It is typically treated with surgical intervention to correct abnormal head shape. Currently, there is no standard method for evaluating treatment effectiveness. We aim to assess the utility of MirrorMe3D, a novel three-dimensional mobile imaging application, in monitoring the head shape changes of two patients with craniosynostosis. METHODS: We implemented the application in the surgical management of a 22-month-old with mild metopic craniosynostosis and a 19-month-old with severe trigonocephaly. Using the application,

Third Patient With Biallelic Variants in SMAD6 With an Overlapping Phenotype: Developmental Delays, Dysmorphic Features, and Cardiovascular Abnormalities.

SMAD6 encodes an inhibitory SMAD protein that modulates BMP and TGF-β signaling. Heterozygous pathogenic variants in SMAD6 have been primarily associated with aortic valve disease, radioulnar synostosis, and nonsyndromic sagittal and metopic synostosis. However, only two syndromic patients with biallelic variants have been reported in the literature. We report a 4-year-old girl with neurodevelopmental delays, dysmorphic features, complex congenital heart disease, renal asymmetry, and arterial tortuosity. Whole exome sequencing showed two homozygous SMAD6 variants of uncertain significance: c.1

A Case of Malan Syndrome With Pulmonary Artery Dilatation due to a Novel Frameshift Variant in Exon 2 of the NFIX Gene.

Malan syndrome is a rare overgrowth disorder caused by deletions or pathogenic variants in the NFIX gene. Here, we present the case of a Japanese male infant with Malan syndrome, which was caused by a novel frameshift variant resulting from an eight-base insertion in Exon 2 of the NFIX gene. Echocardiography revealed dilatation of the pulmonary artery without hemodynamic abnormalities. Malan syndrome shares many clinical features with Sotos syndrome, including overgrowth, macrocephaly, developmental delay, and intellectual disability. Therefore, molecular genetic testing is required for diagno

Presence of Emissary Veins in the Glabellar Region in Metopic Craniosynostosis.

Emissary veins connect intra- and extracranial venous circulation by bridging the dural venous sinuses to the external veins of the skull. Their variable presence and location may have important implications in perioperative blood loss. Clinical observations and previous studies suggest that emissary veins are more frequently present in the glabellar region in patients with metopic synostosis and may serve as a source of bleeding during cranioplasty. This study investigated the presence of emissary veins in the glabellar region in patients with metopic synostosis. We retrospectively analyzed c

Male-Biased Social Deficits in Chd8+/R2219* Mouse Model of Autism Linked to Hippocampal Abnormalities.

Autism spectrum disorder (ASD) is a neurodevelopmental condition with a 4:1 male bias, yet the biological basis for this sex difference remains poorly understood. Heterozygous mutations in the chromatin remodeler CHD8 confer high risk for ASD, with phenotypes influenced by sex and genetic background. Here, we generated Chd8+/R2219* mice carrying a CRISPR-Cas9-generated mutation orthologous to a human variant. These mice recapitulated core ASD features, including macrocephaly and autistic-like behaviors. Notably, social deficits showed a male preponderance, directly mirroring the human sex bias

Dissolving Doubts: Resorbable Fixation in Posterior Vault Distraction Osteogenesis.

Posterior cranial vault distraction osteogenesis (PVDO) traditionally requires titanium screw fixation of metallic distractor footplates. Cho and colleagues recently described an off-label application of the Synthes RapidSorb Injectable Polymer System for footplate fixation in PVDO, reporting reduced distractor removal operative time in an initial single-institution series. This study expands that cohort and further evaluates operative outcomes across a multisurgeon institutional experience. Retrospective review identified consecutive PVDO patients treated between 2013 and 2025. The most recen

Familial p.(Ala73Thr) Variant in GNB2 Associated With Mild Neurodevelopmental Features and Pilocytic Astrocytoma.

Pathogenic variants in GNB2 have been associated with a neurodevelopmental disorder that includes global developmental delays and intellectual disability, hypotonia, increased risk for seizures, heart and renal anomalies, and characteristic facial features. Thus far, all reported patients with GNB2-related disorder have had de novo missense variants. Many of these single nucleotide substitutions have been recurrent. Exome sequencing identified a heterozygous GNB2 variant, c.217G>A, predicting p.(Ala73Thr) (NM_005273.3) in a family, including two brothers with developmental delays and their mor

Comparison of Craniofacial Morphology Following Single- Versus Multi-Segment Fronto-Orbital Advancement for Metopic Synostosis.

ObjectiveMetopic synostosis (MS) is commonly corrected using multi-segment fronto-orbital advancement (FOA), including the frontal bone and supraorbital bandeau. Although this technique reliably expands the anterior vault, it is associated with contour irregularities and bandeau step-offs. The single-segment FOA, utilizing a unified parietal bone flap, was developed with the goal of restoring a seamless frontal contour. This study compares morphologic outcomes between single-segment and multi-segment FOA in MS.DesignA retrospective review of 3-dimensional imaging of patients with MS who underw
