Pathogenic variants in the potassium channel tetramerization domain-containing 1 (KCTD1) gene cause scalp-ear-nipple syndrome (SENS), a rare autosomal dominant developmental disorder. Although the recurrent p.Gly62Asp variant has been previously reported, its genotype-phenotype correlation and structural pathogenic mechanism remain poorly understood. Here, we describe a 10-year-old boy with classical SENS who additionally presented with severe craniofacial abnormalities, including marked hypertelorism, short palpebral fissures, depressed nasal bridge, and nasal bone hypoplasia, together with m

Aplasia cutis congenita (ACC) is a rare congenital malformation characterized by the localized absence of skin and occasionally subcutaneous and bone tissues. Its etiology is multifactorial, and management ranges from conservative wound care to surgical intervention. We report the case of a male newborn presenting with a 4-5 cm ulcerated lesion at the vertex of the scalp, with complete absence of bone tissue and dural exposure. Physical examination revealed syndromic features (facial asymmetry, hypertelorism, low-set ears, and cutaneous syndactyly) and an atrial septal defect. A strong autosom

ObjectiveWhile median cleft lip (MCL) remains rare, improvements in prenatal and postnatal care have improved survival past the neonatal period and thus have increased consideration of surgical repair. MCL is also anatomically distinct from unilateral and bilateral cleft lips and is associated with underlying genetically based syndromes. There is a lack of aggregated data on MCL, and this study provides an overview of the literature, characterizes associated phenotypes, and synthesizes surgical and follow-up management.DesignFollowing Preferred Reporting Items for Systematic Reviews and Meta-A

INTRODUCTION: Gorlin syndrome (GS) or basal cell nevus syndrome type 1 (BCNS1) is a rare genetic disease belonging to the spectrum of genodermatoses. Here, we present a case with BCNS1 that was caused by haploinsufficiency of PTCH1 gene, which is associated with BCNS1.
METHODS: A diagnosis for BCNS1 was clinically set for a 15-year-old female patient who presented with four nevoid skin lesions, four radiolucent osteolytic lesions of the jaws, macrocephaly, unilateral congenital cataracts, strabismus, hypertelorism, and calcification of the falx cerebri. The patient's parents bore no clinical

Craniofrontonasal dysplasia (CFND) is a rare X-linked craniofacial malformation that manifests with midline defects such as hypertelorism, coronal craniosynostosis, and extracranial skeletal anomalies. The standard timeline for surgical management typically includes early cranial vault remodeling and later orbital hypertelorism correction, most commonly through orbital box osteotomies at mixed dentition. For infants presenting with unicoronal craniosynostosis (UCS), fronto-orbital advancement (FOAR) is often used to correct asymmetry and allow for cranial expansion. However, in patients with k

BACKGROUND: Frontonasal dysplasia (FND) is a rare craniofacial anomaly characterized by hypertelorism and nasal deformities. Traditional intracranial approaches for correction, such as box osteotomy or facial bipartition, are effective but associated with high morbidity. This study presents an extracranial umbrella graft technique for nasal reconstruction in patients with FND.
METHODS: Between 2014 and 2024, 55 patients with FND were retrospectively evaluated, and 14 met the criteria for extracranial nasal reconstruction. Inclusion criteria were interorbital distance (IOD) <40 mm and the pres

3MC syndrome is a rare congenital malformation disorder caused by biallelic pathogenic variants in COLEC10, COLEC11, and MASP1. It is characterized by distinctive craniofacial anomalies, growth retardation, developmental delay, and variable systemic findings. Here, we report seven previously unreported patients with 3MC syndrome from five unrelated families. The cohort included five females and two males, aged 1-10 years. All patients exhibited characteristic craniofacial features, including hypertelorism, blepharoptosis, highly arched eyebrows, and epicanthus inversus. Cleft lip and/or palate

BACKGROUND: Heterozygous variants in the ZEB2 gene are known to cause Mowat-Wilson syndrome (MWS). The classical clinical spectrum of MWS includes characteristic facial features, intellectual disability, epilepsy, Hirschsprung disease (HSCR), and various congenital malformations. Reported pathogenic variants have predominantly been truncating variants or missense variants involving the C-terminal zinc-finger domain. To date, no disease-causing missense variant affecting the N-terminal zinc-finger domain has been documented.

Nevoid basal cell carcinoma syndrome (NBCCS) is a rare autosomal dominant hereditary condition associated with developmental abnormalities and tumourigenesis with a prevalence rate of 1:60,000. The objective of this study was to identify NBCCS in patients diagnosed with odontogenic keratocysts (OKC) at Nair Hospital Dental College in Mumbai, India, and to compare the findings with those from Indian and international studies. Archived institutional data on OKC cases from the years 2009-2024 were analysed for NBCCS based on the criteria established by Kimonis et al. in 1997. Of the 231 cases of

Genetic variations in MID1 were initially identified as the cause of X-linked Opitz G/BBB syndrome, which is characterized by hypertelorism, hypospadias, and a high incidence of intellectual disability. However, increasing evidence has shown that MID1 mutations are associated with a broader spectrum of phenotypes, and the genotype-phenotype correlation remains unclear. In this study, the authors report a 4-year-old boy presenting with congenital unilateral nostril hypoplasia, preaxial polydactyly, hemispheric brain asymmetry, and moderate intellectual disability. Whole-exome sequencing identif

INTRODUCTION: Gorlin-Goltz syndrome is a rare tumor-predisposing condition caused by genetic variants in the PTCH1 and SUFU pathways. The list of genetic variants associated with Gorlin-Goltz syndrome is not yet exhaustive. In this study, we present a unique case with typical features and a genetic variant previously classified as a variant of uncertain significance.
CASE PRESENTATION: In this case, genetic testing was performed due to fetal hydronephrosis and omphalocele. Additional features included macrocephaly with hypertelorism and agenesis of the corpus callosum. Genetic testing reveale

The authors present a novel single-stage extracranial transnasal approach for repairing frontoethmoidal encephalomeningocele in a 1-year-old girl, eliminating the need for a coronal incision. Through an inverted-Y nasal incision, the procedure encompassed herniated tissue excision, duraplasty, medial orbital osteotomies for medial hypertelorism correction, cranial base reconstruction with hydroxyapatite and poly-L-lactic acid mesh, costochondral graft nasal dorsum reconstruction, and Y-to-V flap closure. Postoperative recovery was uneventful, with follow-up demonstrating improved orbital align

BACKGROUND: Juvenile-onset open-angle glaucoma (JOAG) is a heterogeneous early-onset glaucoma subtype. Axenfeld-Rieger syndrome (ARS) is an autosomal dominant disorder caused by FOXC1 variants, which may present with severe early-onset glaucoma and can be clinically mistaken for JOAG. This study aimed to determine the genetic cause and clarify the clinical diagnosis in a Chinese family initially diagnosed with JOAG.
METHODS: A 15-year-old male proband and his family members received detailed ophthalmic and systemic evaluations. Whole-exome sequencing was performed in the proband, and candidat

BACKGROUND: There are few studies on the long-term effects of hypertelorism correction on occlusion status. The objective of this study is to identify and assess long-term changes in occlusion status following hypertelorism correction.
METHODS: An observational retrospective study was performed on consecutive patients who underwent hypertelorism correction via facial bipartition or box osteotomy between 2008 and 2019. Patients with Apert, Crouzon, or Pfeiffer syndromes, and patients who failed to complete at least 5 years of follow-up, were excluded from this study. Twenty-two patients who un

Simpson-Golabi-Behmel syndrome (SGBS) is a rare, X-linked overgrowth disorder caused by pathogenic variants in the GPC3 gene, which encodes glypican-3. It is characterized by prenatal and postnatal overgrowth, craniofacial dysmorphism, supernumerary nipples, skeletal anomalies, and variable neurodevelopmental delay. Because of overlapping features such as macroglossia and coarse facial appearance, SGBS may be mistaken for acromegaly. We report a 33-year-old man evaluated for apparent acromegaloid facies. He had tall stature (196.5 cm; >97th percentile), macroglossia, mandibular prognathism, hy

Orbital hypertelorism is a rare and surgically demanding craniofacial anomaly requiring highly accurate three-dimensional repositioning of the orbital complexes. This retrospective case series reports the use of virtual surgical planning with patient-specific cutting guides for orbital box osteotomy in five pediatric patients (mean age at surgery 5.8 years; range 5-8 years) treated at a tertiary craniofacial center between 2021 and 2024. Osteotomy and ostectomy lines were planned on CT data and transferred intraoperatively using customized fronto-orbital and infraorbital guides, followed by me

INTRODUCTION: Craniofrontonasal syndrome displays an unusual X-linked dominant inheritance pattern due to pathogenic variants in EFNB1, which encodes a membrane-bound ligand for the ephrin receptor. Females exhibit a more severe phenotype than males and can have craniosynostosis, hypertelorism, craniofacial asymmetry, thoracic skeleton abnormalities, and an assortment of other features. Males typically only develop a milder hypertelorism, or they have no discernable features at all.
METHODS: Exome sequencing was used to identify a likely pathogenic EFNB1 c.129-2A>G splice site variant in an i

The cohesin complex is an evolutionarily conserved multi-subunit protein assembly essential for sister chromatid cohesion, meiotic recombination, DNA double-strand break repair, and transcriptional regulation. Pathogenic variants in its subunits are implicated in a spectrum of reproductive and developmental disorders, including non-obstructive azoospermia, premature ovarian insufficiency, reproductive aging, aneuploidy, Cornelia de Lange syndrome, Roberts syndrome, cancer, and neuropsychiatric disease. Consequently, identifying cohesin mutations is a priority for precision diagnostics and pers

Orbital hypertelorism is observed in a wide spectrum of congenital disorders, such as in case of frontonasoethmoidal tumors or encephaloceles, craniofrontonasal dysplasia, craniofacial clefts, syndromic craniosynostosis, Rasopathies and various malformative and multisystemic congenital syndromes. Yet its underlying pathophysiological mechanisms remain incompletely understood. Our systematic literature review aimed to delineate and classify the different conditions associated with true orbital hypertelorism based on both genetic bases and physiopathological mechanisms of orbital hypertelorism.
