A radiological case series of three siblings with osteogenesis imperfecta and shared paternal inheritance.

Источник пометил публикацию как отозванную

Osteogenesis imperfecta (OI), or brittle bone disease, is a genetically inherited connective tissue syndrome that manifests through autosomal dominant or recessive patterns. We present an unusual case involving three siblings with the same father but different mothers. The patients had extremely short stature, recurrent fractures, and varying degrees of severity of OI, in addition to distinctive features such as blue sclera and dentinogenesis imperfecta. The radiographic skeletal surveys revealed the characteristic features of OI in all three siblings. The patients exhibited Wormian bones, mul

Severe Aortic Regurgitation in Osteogenesis Imperfecta Complicated by Mediastinal Hematoma.

BACKGROUND: Osteogenesis imperfecta (OI), a heritable type I collagen disorder, can cause aortic regurgitation (AR) and increase perioperative bleeding risk, even with normal routine coagulation tests. CASE SUMMARY: A man in his 40s presented with AR due to right coronary cusp prolapse from cusp bending. Blue sclera and childhood fractures suggested OI. After an attempted repair, surgical aortic valve replacement was performed; a large postoperative mediastinal hematoma with anemia was managed conservatively without reoperation. DISCUSSION: A prior report identified cusp bending in OI mainly

Corrective osteotomy for 111.4-degree tibial procurvatum in osteogenesis imperfecta type III patient: a rare case report with multistage interventions.

Osteogenesis imperfecta (OI) is a rare genetic disorder marked by bone fragility and progressive deformity. Type III OI represents a severe form characterized by recurrent fractures, bowing, and significant functional impairment. We report a 17-year-old with type III OI and an extreme 111.4° tibial procurvatum deformity alongside recurrent femoral fractures. His history included multiple intramedullary nailing procedures and revision surgeries. The most recent treatment involved a two-level tibial osteotomy with subsequent cast immobilization, resulting in marked improvement. The Lower Extremi

Co-Occurrence of Osteogenesis Imperfecta Type III and Chronic Abruption-Oligohydramnios Sequence: A Case Report Suggesting a Possible Role of Type I Collagen Fragility.

Pregnancy complicated by severe osteogenesis imperfecta (OI) is rare, and chronic abruption-oligohydramnios sequence (CAOS) is an uncommon obstetric disorder characterized by persistent bleeding and oligohydramnios without membrane rupture. To our knowledge, this is the first report describing the coexistence of type III OI and CAOS. A 24-year-old woman with OI developed recurrent bleeding early in pregnancy. Genetic testing revealed a novel COL1A1 splice-site variant (c.1876-2A>C). Ultrasonography showed a circumferential subchorionic hematoma and progressive fluid loss, leading to CAOS diagn

Report of the favorable pregnancy outcomes in an FKBP10-related Bruck syndrome case and a narrative review of pregnancy in severe osteogenesis imperfecta.

BACKGROUND: Osteogenesis imperfecta (OI) is a genetic syndrome that affects skeletal and connective tissues. OI type XI, also known as Bruck syndrome, is an extremely rare autosomal recessive disorder characterized by symptoms of severe OI, along with additional features. Despite the fact that patients with severe OI can survive into adulthood and have preserved fertility, only a few case reports have been published about pregnancy complications in Bruck syndrome. CASE REPORT AND NARRATIVE REVIEW: We report a case of maternal FKBP10-related Bruck syndrome, along with details of antenatal care,

Simultaneous Bilateral Tibial Tubercle Avulsion Fractures Complicated by Compartment Syndrome in an Adolescent With Osteogenesis Imperfecta: A Case Report.

BACKGROUND: Osteogenesis imperfecta (OI) is a rare collagen disorder causing brittle bones. Tibial tubercle fractures are uncommon in children, comprising < 2.7% of pediatric fractures, and bilateral injuries in OI are exceptionally rare. No prior reports detail bilateral tibial tubercle fractures with concurrent bilateral compartment syndrome. We present the first known case of this presentation in an 11-year-old boy. CASE PRESENTATION: An 11-year-old male with OI fell on his knees, presenting with severe bilateral leg pain. Imaging revealed bilateral Ogden III tibial tubercle fractures. Due

Lung scRNA-seq reveals chronic inflammation and emphysemous phenotype in mice with osteogenesis imperfecta.

Osteogenesis imperfecta (OI), or brittle bone disease, is a rare congenital disorder characterized by bone fragility and increased fracture incidence mainly due to mutations in type I collagen or genes associated with collagen synthesis. Genetic and allelic heterogeneity underlie the phenotypic spectrum of OI yet all forms commonly feature early mortality stemming from pulmonary complications, the molecular cause for which has not been resolved. Using single-cell RNA sequencing (scRNAseq), we identified novel molecular and cellular mechanisms underlying the lung abnormalities observed in our C

Evaluation of Fracture and Osteotomy Union in the Setting of Osteogenesis Imperfecta: Multicenter Reliability of the Modified Radiographic Union Score for Tibial Fractures (RUST).

BACKGROUND: A critical component of patient care for osteogenesis imperfecta patients is understanding the development of union following fracture and osteotomy surgery. Studies evaluating the criteria of bony union in this population have varied. An objective standardized method for fracture healing was developed as the radiographic union score for tibial fractures (RUST), and subsequently revised to the modified RUST (mRUST). The RUST score has been shown to have excellent interobserver and intraobserver reliability in a single-center investigation in an OI population. The purpose of the stu

Identifying Risk Factors for Revision Surgery After Fassier-Duval Rodding in Osteogenesis Imperfecta Patients: A Large Single-Center Cohort Review.

BACKGROUND: The Fassier-Duval (FD) telescopic rod is the standard of care for treating long bone fractures and deformities in children with osteogenesis imperfecta (OI). Despite its advantages, revision surgery remains common. This study aimed to analyze the medium-to long-term outcomes of FD rodding and identify risk factors for revision surgery. METHODS: A retrospective review was conducted on all OI patients who received lower limb FD rods between 2009 and 2020. Data on patient demographics, OI type, surgical details, and follow-up were collected. The primary outcome was revision surgery.

OMICS Profiling Identifies Signatures of Senescence in Osteogenesis Imperfecta Osteoblasts Counteracted by 4-PBA.

Mutations in collagen I are the most common cause of osteogenesis imperfecta (OI), leading to delayed protein folding and structurally abnormal molecules. While some aberrant collagen is secreted into the extracellular matrix (ECM), impairing bone quality, a significant fraction is retained intracellularly, disrupting osteoblast homeostasis. 4-phenylbutyrate (4-PBA) has been shown to improve osteoblast function and ECM composition in OI models. To investigate the intracellular consequences of mutant collagen retention and the mechanisms of 4-PBA, we analysed the secretome and transcriptome of

Does osteogenesis imperfecta predispose infants to metaphyseal fractures? A systematic review.

OBJECTIVE: The notion that osteogenesis imperfecta (OI) predisposes children to metaphyseal fractures is controversial. This review aimed to ascertain the prevalence of metaphyseal fractures in children with OI less than 2 years of age. MATERIALS AND METHODS: Medline, CINAHL and Web of Science were searched for studies reporting metaphyseal fractures in children with OI less than 2 years of age. Articles not retrievable, not published in English, or where 'metaphyseal' fracture was not defined/illustrated, were excluded. Article quality was independently assessed using the Critical Appraisal

Severe osteogenesis imperfecta due to homozygous glycine substitutions in COL1A1 and COL1A2.

BACKGROUND: Osteogenesis imperfecta (OI) is a rare monogenic bone fragility disorder most often caused by heterozygous pathogenic variants in COL1A1 and COL1A2, typically showing autosomal dominant inheritance. Biallelic pathogenic variants in these genes are rare and associated with Ehlers-Danlos-like features or severe OI forms. METHODS: Within a collaborative study on pediatric bone fragility in Egyptian patients, we performed whole-exome sequencing and segregation analysis in families with suspected OI. Furthermore, we systematically analyzed triple-helical-domain glycine substitutions fo

Clinical and Genetic Characterization of Osteogenesis Imperfecta in Japanese Patients: Outcomes of Sequential Bisphosphonate Therapy.

Although novel drug development and clinical trials are ongoing, bisphosphonates remain the standard treatment of osteogenesis imperfecta (OI). This study aimed to evaluate clinical effects of bisphosphonate therapy across clinical subtypes and genetic variants of OI, and to explore treatment outcomes of sequential bisphosphonate therapy in patients with severe disease. This retrospective analysis was conducted using clinical data from patients with genetically confirmed OI. Bone mineral density (BMD), trabecular bone score (TBS), height, and fracture incidence were analyzed in patients underg

Molecular and clinical findings in Osteogenesis Imperfecta: A cohort study from a single tertiary center.

OBJECTIVE: Osteogenesis imperfecta (OI) is a clinically and genetically heterogeneous skeletal fragility disorder. Although COL1A1/COL1A2 variants account for most cases, non-COL1A genes and overlapping monogenic bone fragility conditions complicate diagnosis and genotype-phenotype interpretation in routine practice. We aimed to describe the clinical spectrum and molecular findings of patients evaluated for suspected or confirmed OI at a tertiary referral center, and to assess phenotype patterns across genotype-informed groups. METHODS: We conducted a single-center retrospective cohort study

Transcriptomic and proteomic analysis of osteogenesis imperfecta disease-specific induced pluripotent stem cells.

BACKGROUND: Osteogenesis imperfecta (OI) is a rare genetic bone disorder for which treatment options remain limited. Disease-specific induced pluripotent stem cells (iPSCs) offer a valuable model to investigate its underlying pathological mechanisms. METHODS: Peripheral blood cells from two OI patients were reprogrammed into iPSCs. Transcriptomic sequencing and proteomic analysis were employed to systematically identify differentially expressed genes (DEGs) and proteins (DEPs) between OI-iPSCs and normal iPSCs. Bioinformatics approaches were subsequently applied for enrichment analysis and pr

Functional validation using a zebrafish model reclassifies the rare COL1A2 p.Gly418Ala variant as a cause of Osteogenesis Imperfecta.

Osteogenesis imperfecta (OI) is a genetically heterogeneous skeletal dysplasia mainly caused by mutations in the COL1A1 and COL1A2 genes, which encode type I collagen. Here, using whole-exome sequencing(WES), we identified a rare heterozygous missense mutation (c.1253G > C, p.Gly418Ala) in COL1A2 in a Chinese family with recurrent prenatal skeletal dysplasia. This variant is located within the highly conserved Gly-X-Y repeat of the collagen triple-helical domain and predicted to be deleterious by bioinformatics programs. However, according to the American College of Medical Genetics and Genomi
