Population pharmacokinetics modeling of albendazole and its active metabolite, albendazole sulfoxide in mass drug administration against lymphatic filariasis.

Albendazole (ALB), a benzimidazole antiparasitic, is widely used in mass drug administration (MDA) campaigns against lymphatic filariasis (LF). It is rapidly metabolized to its active form, albendazole sulfoxide (ALB-OX), which exhibits considerable interindividual variability (IIV) in pharmacokinetics (PK). This variation may affect ALB efficacy in MDA programs. We developed a population pharmacokinetic (PopPK) model characterizing ALB and ALB-OX exposure and identifying demographic and clinical covariates contributing to IIV in patients with LF and healthy individuals. Plasma concentration-t

In vitro in-depth characterization of emodepside in combination with key anthelmintic drugs on soil-transmitted helminths.

Despite the enormous disease burden, treatments are limited for soil-transmitted helminthiasis. The handful of anthelmintic drugs suffer from variable and often insufficient efficacy against the pathogens. Resistance development, caused by high drug pressure, is an ongoing concern. New treatments are needed, and combination therapies may represent a promising therapeutic strategy. We explored emodepside as a partner in combination therapy and characterized drug-interaction effects with key anthelmintics in vitro against different soil-transmitted helminths (STHs). Drug-interaction studies were

Prevalence of Strongyloides stercoralis infection among African children: A systematic review and meta-analysis.

Strongyloides stercoralis is a neglected parasite that causes strongyloidiasis. However, the true prevalence of strongyloidiasis is underreported due to low-sensitivity diagnostic methods. This systematic review assessed the prevalence of S. stercoralis among African children and its variation by diagnostic methods, countries, and study settings. All studies published in African countries on S. stercoralis prevalence up to the year 2025 in the PubMed, Google Scholar, Scopus, and ScienceDirect databases were included. Identification, screening, checking the eligibility, and including the releva

Assessing diagnostic accuracy of onchocerciasis rapid diagnostic tests using Bayesian latent class models.

BACKGROUND: Onchocerciasis elimination programs increasingly rely on rapid diagnostic tests (RDTs) that detect antibodies to the Ov16 antigen, yet the performance of currently available rapid tests remains uncertain. The commercially available SD BIOLINE Ov16 RDT has shown inconsistent sensitivity when used on whole blood and does not consistently meet international performance thresholds for mapping or decisions to stop mass drug administration (MDA). Two novel onchocerciasis RDTs have been developed, DDTD RDT and GADx RDT, but their diagnostic accuracy has not been fully evaluated using meth

Comparison of post-mortem examination, faecal sedimentation, antibody ELISA, and real-time PCR methods in the detection of Fasciola spp. in naturally infected dairy cattle.

Fasciolosis is a food-borne disease affecting livestock worldwide. Improved diagnostic methods for detecting fasciolosis have been introduced; however, limited research has compared these methods. The current study aimed to compare the detection of Fasciola in naturally infected slaughtered dairy cattle using four methods. Blood and faecal samples were collected from slaughtered cattle in three high-throughput abattoirs in the Eastern Cape Province, South Africa, from 2020 through 2021. Post-mortem liver examination was conducted to detect and identify Fasciola spp. Blood from slaughtered carc

Fimbriarioides intermedia and F. lintoni (Cestoda: Hymenolepididae): molecular phylogeny, species status, and transmission in Arctic coastal ecosystems.

Cestodes of the genus Fimbriarioides (Hymenolepididae, Fimbriariinae) parasitise waterfowl and shorebirds in the Holarctic. Using molecular and morphological methods, we studied two Fimbriarioides spp. from sea ducks of the European North of Russia: the type species F. intermedia from eiders Somateria mollissima and S. spectabilis 1758 and a closely related species F. lintoni from a previously unrecognised host, Velvet Scoter Melanitta fusca 1758. Our analysis also involved metacestodes Fimbriarioides sp. from barnacles Semibalanus balanoides of the Barents Sea and the Sea of Okhotsk. Metacest

Frequency of Mutations Within P-Glycoprotein and Beta-Tubulin Associated With Ivermectin Treatment in Onchocerca volvulus Adult Female Worm Populations in Some Onchocerciasis Endemic Communities in Ghana.

BACKGROUND: Ivermectin is the drug of choice for the control of onchocerciasis because of its potent embryostatic and microfilaricidal effect. However, with long periods of mass drug administration, the effect of ivermectin on the Onchocerca volvulus worms is waning. These suboptimal responses have been linked to genes such as p-glycoprotein (P-gp) and beta-tubulin (B-tub). The purpose of this study was to define the proportion of adult female O. volvulus from some endemic communities in Ghana that have mutations in these genes. METHODS: O. volvulus adult female worms were obtained from 33 pa

Comparative Effect of Water-Diluted Stool on the Growth and Development of Rhabditis sp., Strongyloides stercoralis, and Hookworm.

BACKGROUND: Raw vegetables in southern Thailand frequently carry Rhabditis sp. Epidemiological shifts (2008-2023) show an increase in Rhabditis sp. colonization alongside declining hookworm and Strongyloides stercoralis infections, suggesting a potential association with climate variations. To evaluate moisture effects under controlled conditions, we compared nematode egg and first-stage larval development using an agar plate culture (APC) control and a 1:20 water-diluted stool suspension (SS) simulating wet environments. METHODS: Stool samples positive for Rhabditis sp. from 18 humans (12 co

Transient receptor potential channel activation targets multiple stages of the schistosome parasite.

Schistosomiasis, a neglected tropical disease caused by infection with parasitic blood flukes of the genus Schistosoma, affects more than 250 million people worldwide. Currently, patients rely on a single medicine, praziquantel (PZQ), discovered half a century ago. To combat drug resistance and reach elimination of the schistosome parasite, alternative drugs to PZQ are required that treat and prevent disease. Here, we describe a drug candidate, M5493, with potent activity against multiple parasite life cycle stages. The target is a parasite transient receptor potential ion channel, distinct fr

Long-term dynamics of gastrointestinal parasite egg shedding in a semi-feral Misaki horse population in Japan.

This study investigated long-term gastrointestinal parasite egg shedding in a semi-feral Misaki horse population using 3200 fecal samples collected from 158 individually identified horses during five fiscal years (FY2019-FY2023; April 2019-March 2024). Strongyle-type eggs were detected in 91.7% of samples (2935/3200), with a mean count of 1352 eggs per gram (EPG). Repeated-measures analyses showed marked variation associated with fiscal year, season, age, and management history. Strongyle-type EPG values were generally higher in spring and summer than in autumn and winter, with the month-level

Economic evaluation of primary health care integration and targeted mass drug administration in counties in western Kenya.

INTRODUCTION: The 2025 World Health Organization (WHO) Global Report on Neglected Tropical Diseases (NTDs) highlights major progress made worldwide in controlling and eliminating these diseases. Sustaining NTD control requires cost-efficient strategies integrated into routine health systems. We evaluated the cost-effectiveness of integrating soil-transmitted helminths (STH) and schistosomiasis (SCH) treatment into routine Primary Health care (PHC) services compared with the campaign-based approach across four counties and selected sub-counties, wards, and primary health facilities in western K

Highly efficient RPA product degradation and expanded PAM recognition enable RPA-Cas3 detection of Clonorchis sinensis.

Clonorchis sinensis is a major foodborne pathogen threatening aquatic food safety. Microscopy, the diagnostic standard, is laborious and often lacks sensitivity for low-intensity infections. Molecular strategies coupling isothermal amplification with CRISPR/Cas systems have attracted interest, but their reliability is often undermined by competition between amplification and detection and intrinsic limitations of Cas effectors, notably PAM dependence of Cas12a and nonspecific activation of Cas13a. To address these limitations, we used the type I-A CRISPR-Cas3 system (TsiCas3) from T. siculi. B

Climate Change and Socioeconomic Vulnerability: The Carpathian Basin as a Potential Hotspot in the Dissemination of Dirofilaria repens in Europe.

Dirofilaria repens is a zoonotic parasite expanding across Europe due to climate change. We hypothesised that in this process, the Carpathian Basin has a facilitating effect. Using 426 georeferenced European cases, the probability of infection occurrence was determined in relation to climatic factors, surface water availability, regional social deprivation, and stray dog population density. To analyse the potential impacts of ecological and social factors (deprivation and stray dog population density), the MaxEnt algorithm, and spatial Empirical Bayes smoothing and bivariate local indicators o

Helminths of the invasive Podarcis muralis (Laurenti, 1768) and native P. tauricus (Pallas, 1814) wall lizards from two localities in southern Ukraine.

Helminth communities of the invasive Podarcis muralis and the native P. tauricus were investigated in southern Ukraine based on newly collected material and historical data. A total of nine P. muralis and 20 P. tauricus specimens were examined. Two helminth species were recorded in each lizard host. In P. muralis, the nematode Spauligodon saxicolae and cystacanths of the acanthocephalan Centrorhynchus globocaudatus were found, while S. saxicolae and the cestode Oochoristica tuberculata were found in P. tauricus. Historical data from 116 P. tauricus collected between 1951 and 1990 revealed a to

Exploring Blastocystis: hidden complexity in a common intestinal microbe.

Blastocystis is an anaerobic stramenopile that colonizes the large intestine of humans and many animals worldwide. Small-subunit rRNA-based typing reveals deep genetic diversity: at least 44 genetic variants, namely, subtypes (STs), are recognized, with ST1-ST4 accounting for most human carriage. Its medical relevance remains contested. Blastocystis carriage is frequently asymptomatic, and while associations with non-specific gastrointestinal symptoms (e.g. diarrhoea, abdominal pain and bloating), irritable bowel syndrome and extraintestinal manifestations such as urticaria have been proposed.

Giardia duodenalis in domestic cats from the US and Canada: Prevalence and zoonotic potential.

Giardia duodenalis is a flagellated commensal protozoan that can infect both humans and animals. Infection can result in clinical signs such as diarrhea or, more often, the infection can be subclinical. Within the G. duodenalis species, there are several major genetic assemblages, A - H. Certain assemblages are species specific (C and D in dogs, F in cats) while assemblages A and B can infect humans as well as other mammals. This retrospective study used data collected from a commercial reference laboratory (Antech Diagnostics, Mars Petcare Science & Diagnostics). Real-time PCR data using feca

Role of zinc oxide nanoparticles in the parasitism and abomasal pathophysiology of lambs artificially infected with Haemonchus contortus.

Haemonchus contortus is a major gastrointestinal nematode affecting sheep and causing substantial economic losses. Nutritional strategies like zinc supplementation may enhance host resilience and immune responses. This study evaluated the effects of zinc oxide microparticles (ZnO-MC) and nanoparticles (ZnO-NP) on parasitological, histopathological, and mucosal responses in infected lambs. Fifty-nine Santa Ines lambs were allocated into three groups: unsupplemented control, ZnO-MC, or ZnO-NP. Infected animals received 1200 H. contortus L3 weekly for 24 weeks. Parasitological parameters, weight

The development and validation of long-read ITS-1/5.8S/ITS-2 nemabiome metabarcoding for ovine gastrointestinal nematodes using Oxford Nanopore Technologies (ONT) sequencing.

Internal Transcribed Spacer ITS-2 rRNA nemabiome metabarcoding is increasingly used to characterize gastrointestinal nematode (GIN) communities. While powerful, current approaches have some limitations in their flexibility and applicability to smaller-scale studies and diagnostic use. The short read lengths provided by the Illumina platform may lack discriminatory power for some closely related species and also pose a challenge for new marker selection and primer design. To address these challenges, we have developed ITS-1/5.8S/ITS-2 rRNA Oxford Nanopore Technologies (ONT) long-read metabarcod

Clinical and epidemiological characteristics of patients hospitalized for toxocariasis in Japan: A nationwide analysis using the diagnosis procedure combination database.

Toxocariasis is a zoonotic helminth infection caused by Toxocara canis or Toxocara cati. While it has traditionally been regarded as a pediatric soil-transmitted disease, recent reports from Japan suggest that an increasing number of adult-onset cases are potentially associated with foodborne transmission. Ocular involvement in adults can also result in severe visual impairment. However, nationwide data describing the epidemiological and clinical characteristics of hospitalized patients in Japan are limited. We conducted a nationwide retrospective study using the Japanese Diagnosis Procedure C

Trophic niche segregation in small pelagic fishes suggests mesopelagic pearlsides as the main transmission pathway of Anisakis larvae in a Patagonian food web.

Many parasites are transmitted through trophic interactions, yet the specific pathways connecting intermediate and definitive hosts often remain poorly understood in marine food webs. In the Southwestern Atlantic, the Argentine anchovy (Engraulis anchoita) has been considered a potential vector of larval Anisakis to higher trophic levels, but low infection levels in San Matías Gulf suggest alternative transmission routes in this gulf. We investigated the role of pearlsides (Maurolicus spp.), abundant mesopelagic fishes in the gulf, in the transmission of anisakid nematodes. Examination of 229
