Rate and Type of Surgery and Prevalence of Malignancy in Patients With Ollier's Disease and Maffucci Syndrome.

Patients with Ollier's Disease (OD) and Maffucci Syndrome (MS) are at risk for fractures, deformities, and limb-length discrepancies (LLDs). This study aims to determine the rate and types of surgery performed on these patients and evaluate the prevalence of developing a malignancy at a prior surgical site. A survey link was posted to a private online social media support group page specific to OD and MS patients. A total of 125 surveys were obtained from five continents. Two surveys were incomplete, and 123 surveys were evaluated with 109 OD and 14 MS patients. 89% had at least one surgery at

Sost deficiency restores calvarial bone thickness without recovery of sutural growth in Runx2 missense mutant mice.

Runt-related transcription factor 2 (Runx2) is a key transcription factor essential for osteogenesis, and RUNX2 haploinsufficiency causes cleidocranial dysplasia, whereas sclerostin encoded by Sost negatively regulates bone mass by antagonizing Wnt/β-catenin signaling. Although the individual roles of Runx2 and sclerostin in skeletal biology have been extensively studied, their combined effects on calvarial development have not been directly examined in vivo. Here, we investigated whether Sost deficiency affects the cranial phenotype associated with Runx2 haploinsufficiency. Using skeletal pre

Multidisciplinary approach and clinical management of McCune-Albright syndrome.

McCune-Albright syndrome (MAS) is a rare, sporadic mosaic disorder classically characterized by fibrous dysplasia (FD) of bone, café-au-lait macules, and endocrine hyperfunction. The estimated prevalence ranges from 1 in 100,000 to 1 in 1,000,000 individuals, with no clear differences across ethnic groups. The central pathogenic mechanism involves postzygotic somatic gain-of-function mutations in the GNAS gene, which encodes the α subunit of the stimulatory G protein (Gsα). The currently identified pathogenic variants are concentrated at codons R201 (e.g., R201H and R201C) and Q227. This revie

Exploring bone formation mechanism and pattern during RANKL inhibition in a fibrous dysplasia mouse model.

Fibrous dysplasia (FD) of bone is a fibro-osseous disorder caused by GNAS mutations with defective osteogenic differentiation and increased bone remodeling activity. Inhibition of RANKL leads to the replacement of FD lesions with bone. However, the mechanism and pattern of deposition of the newly formed bone remain unclear. Here, we perform morphological and molecular analyses on EF1α-GsαR201C (FD) mice receiving an anti-mouse RANKL antibody. We show that, although the treatment reduces the expression of osteogenic genes, osteoblastic cells continue to produce bone matrix within FD lesions. Ho

A Case of Short-Rib Thoracic Dysplasia: Diagnostic Clues in Early Pregnancy and Genetic Analysis.

To investigate the diagnostic clues and genetic etiology of short-rib thoracic dysplasia (SRTD) in early pregnancy, we report a case of a fetus suspected of having SRTD based on ultrasound findings at 13 weeks of gestation. Prenatal genetic testing revealed a pathogenic variant in the DYNC2H1 gene, and post-abortion examination of the specimen along with X-ray findings confirmed the diagnosis as short-rib thoracic dysplasia 3 (SRTD3; OMIM #613091). Such cases are rarely reported in the early gestational period. The detection and subsequent confirmation in this case at 13 weeks demonstrate that

Deferoxamine alleviates T-2 toxin-induced articular chondrocyte ferroptosis via the Nrf2/xCT/GPX4 axis: Implications for Kashin-Beck disease pathology.

Kashin-Beck disease (KBD) is a debilitating endemic osteochondropathy. Environmental toxins, particularly T-2 toxin, and selenium deficiency are established etiological factors. This study aimed to investigate whether T-2 toxin induces ferroptosis in articular chondrocytes and explore the therapeutic potential and mechanism of the iron chelator, Deferoxamine (DFO). A total of 32 three-week-old male Sprague-Dawley rats were randomly allocated into four groups: Control, T-2 toxin (100 ng/g·BW/d), T-2 toxin + low DFO (10 mg/kg), and T-2 toxin + high DFO (100 mg/kg). After a 4-week intervention, k

Acrodysostosis type 1: mechanisms explaining PRKAR1A mutation mediated dysregulation of cAMP-PKA signalling.

Acrodysostosis type 1 (ACRDYS1) is a rare multisystem developmental disorder affecting skeletal growth, endocrine function, neurodevelopment, metabolism, and tooth formation. It is caused by heterozygous mutations in PRKAR1A, which encodes the type Iα regulatory subunit (RIα) of protein kinase A (PKA), a central mediator of cyclic AMP (cAMP)-dependent signalling. Although ACRDYS1 belongs to the broader family of Gsα-cAMP-PKA-related disorders, its underlying mechanism is distinct. Disease-associated PRKAR1A mutations cluster within regions of RIα that bind cAMP and undergo conformational rearr

Genetic Evidence of a Recent Decline and Crossbreed Distribution of TRPV4 c.1024G>T Variant in Domestic Cats.

The folded ear phenotype of Scottish Fold cats is associated with an autosomal dominant TRPV4 variant (c.1024G>T) linked to osteochondrodysplasia. Although genetic testing has been implemented to guide breeding, empirical evidence of its impact on allele frequency remains limited, and crossbreed investigations are lacking. Here, we evaluated longitudinal changes in TRPV4 c.1024G>T allele frequencies in Scottish Folds and surveyed the variant in 8610 cats from 14 breeds in Japan. Overall, between 2017 and 2024, the proportion of homozygous cats significantly declined (from 14.2% to 1.9%, p < 0.

Ulnar lengthening for forearm deformities in hereditary multiple exostoses: a systematic review and meta-analysis (2015-2025).

BACKGROUND: Hereditary multiple exostoses (HME) is an autosomal dominant skeletal disorder frequently associated with progressive forearm deformities, including ulnar shortening, radial bowing, and radial head dislocation. Ulnar lengthening using distraction osteogenesis has become the principal surgical strategy; however, outcomes, techniques, and complication profiles vary across studies. This systematic review and meta-analysis synthesize contemporary evidence on radiographic correction, functional outcomes, and complications following ulnar lengthening for HME-related forearm deformities.
