Transcriptomic and proteomic analysis of osteogenesis imperfecta disease-specific induced pluripotent stem cells.

BACKGROUND: Osteogenesis imperfecta (OI) is a rare genetic bone disorder for which treatment options remain limited. Disease-specific induced pluripotent stem cells (iPSCs) offer a valuable model to investigate its underlying pathological mechanisms. METHODS: Peripheral blood cells from two OI patients were reprogrammed into iPSCs. Transcriptomic sequencing and proteomic analysis were employed to systematically identify differentially expressed genes (DEGs) and proteins (DEPs) between OI-iPSCs and normal iPSCs. Bioinformatics approaches were subsequently applied for enrichment analysis and pr

Functional validation using a zebrafish model reclassifies the rare COL1A2 p.Gly418Ala variant as a cause of Osteogenesis Imperfecta.

Osteogenesis imperfecta (OI) is a genetically heterogeneous skeletal dysplasia mainly caused by mutations in the COL1A1 and COL1A2 genes, which encode type I collagen. Here, using whole-exome sequencing(WES), we identified a rare heterozygous missense mutation (c.1253G > C, p.Gly418Ala) in COL1A2 in a Chinese family with recurrent prenatal skeletal dysplasia. This variant is located within the highly conserved Gly-X-Y repeat of the collagen triple-helical domain and predicted to be deleterious by bioinformatics programs. However, according to the American College of Medical Genetics and Genomi

The pathogenesis and intersecting mechanisms of monogenic bone mass disorders.

Monogenic disorders of bone mass arise from variants in single genes disrupting the intricate processes of bone formation and resorption. Genes underlying bone mass disorders are involved in various intersecting molecular mechanisms in cells of the mesenchymal and haematopoietic lineages. Signalling pathways, including WNT-β-catenin, TGFβ-BMP and NF-κB, have central roles in bone metabolism, and variants in their regulators or effectors can have opposing effects on bone mass. Defects in osteoblast differentiation, collagen biosynthesis and extracellular matrix mineralization primarily drive th

Hematopoietic Stem Cell Transplantation in Infantile Osteopetrosis: Lessons from a Resource-Limited Setting.

Infantile Osteopetrosis is a rare inherited bone disorder characterized by a reduced or complete lack of osteoclast function, leading to defective bone resorption and increased bone density. The defective osteoclast differentiation or function is of hemopoietic origin, thus making Hematopoietic stem cell transplantation (HSCT) the only curative treatment option for this condition. This retrospective study analysed the data of 10 cases of osteopetrosis, undergoing fully HLA-matched HSCT, at AFBMTC from April 2016 to December 2025. Among the cohort, 6(60%) were male with a mean age of 16.09 ± 14

Re: "A novel splice-altering frameshift variant in the COL1A1 gene underlies osteogenesis imperfecta type I: molecular characterization of a four-generation Chinese pedigree and literature review".

A paper entitled "A novel splice-altering frameshift variant in the COL1A1 gene underlies osteogenesis imperfecta type I: molecular characterization of a four-generation Chinese pedigree and literature review" was recently published in this journal. Unfortunately, it contains a number of errors that the authors should correct. The most serious errors are incorrect sequence variant descriptions and inappropriate use of AI-assisted protein structure prediction.

Cleidocranial dysplasia in a Moroccan patient: a case report.

Cleidocranial dysplasia is a rare autosomal dominant disorder characterized by skeletal and dental anomalies, often enabling early recognition by dental practitioners. A 14-year-old Moroccan girl presented to the Pediatric Dentistry Department of Ibn Rochd University Hospital with aesthetic concerns. Clinical and radiographic examinations revealed multiple general, craniofacial, and dental abnormalities that suggested an underlying syndrome. Further investigations supported the diagnosis of cleidocranial dysplasia. The patient received appropriate dental care and was referred to the genetics d

A New Wnt1 Mutant Rat Model of Osteogenesis Imperfecta and Its Application in AAV9-Mediated Gene Therapy.

Osteogenesis imperfecta (OI) is a genetically and clinically heterogeneous bone disorder, with more than 20 genes contributing to OI development. Previously, we identified WNT1 c.620G > A (p.Arg207His) mutation among Chinese patients with autosomal recessive OI (AR-OI). This study aims at investigating the causative role of WNT1 deficiency in OI and evaluate whether AAV-based gene therapy could ameliorate bone abnormalities. We generated and analyzed the Wnt1R207H/R207H rat model. The AAV9-Wnt1 virus was delivered via direct intraosseous injection into the femoral marrow cavity of the OI rats

Balancing osteogenesis and adipogenesis in osteogenesis imperfecta: PLIN2 and E2F2 as key targets for mesenchymal stem cell therapy.

BACKGROUND: Osteogenesis imperfecta (OI) necessitates innovative mesenchymal stem cell (MSC) therapies targeting key molecular pathways to enhance targeted and combination treatments and improve bone health. OBJECTIVES: To investigate the therapeutic mechanisms of various interventions for OI by analyzing relevant datasets, with a focus on lipid metabolism-related genes, particularly PLIN2, in order to determine whether they influence the balance between osteoblast and adipocyte differentiation. MATERIAL AND METHODS: This study analyzed datasets from the Gene Expression Omnibus (GEO; GSE1575

Non-COL1A1/2 genetic burden and osteoporosis-overlap in patients referred with osteogenesis imperfecta phenotype.

PURPOSE: Patients referred with an osteogenesis imperfecta (OI) phenotype exhibit marked genetic heterogeneity, and a substantial proportion may harbor variants associated with early-onset osteoporosis (EOO) or OI-mimicking disorders rather than classical collagenopathies. This study aimed to define the genetic spectrum and non-COL1A1/2 variant burden in a cohort clinically diagnosed with OI. METHODS: Ninety-eight unrelated patients referred with a clinical diagnosis of OI were analyzed using a targeted NGS panel on the DNBSEQ-400 platform and, when indicated, whole-exome sequencing. Clinical Variability Within the PLOD2-Associated Phenotypic Continuum: Three Novel Variants in Four Patients from a Descriptive Case Series.

Background/Objectives: Bruck syndrome type 2 (BS2) is an ultra-rare autosomal recessive disorder within the osteogenesis imperfecta (OI) spectrum caused by biallelic pathogenic variants in PLOD2, which encodes lysyl hydroxylase 2 (LH2), an enzyme essential for bone-specific collagen cross-linking. Marked clinical heterogeneity complicates diagnosis, particularly in patients with atypical or incomplete presentations. We aimed to further delineate the clinical and molecular spectrum of PLOD2-associated disease. Methods: In this descriptive case series, we performed clinical, radiological, and mo

Bone Healing and Skeletal Stability Following Orthognathic Surgery in Fibrous Dysplasia.

Fibrous dysplasia represents a challenge for orthognathic surgery due to the altered anatomy that generates significant asymmetries. In addition, skeletal instability and uncertainty about postoperative bone healing due to the lack of stability of the osteosynthetic material generate great concern in these cases. The authors present the case of a patient who underwent corrective orthognathic surgery. Clinical and radiographic follow-up lasted 2 years. The results showed adequate healing and maintained skeletal stability. This suggests that this procedure may be a safe and effective option for

The Application of Cell and Gene-Modified Cell Therapy in the Treatment of Osteopetrosis.

Bone tissue represents a dynamic tissue that undergoes continuous renewal and remodeling through the coupled actions of osteoblast-driven bone formation and osteoclast-mediated bone resorption. Proper coupling between these two processes is essential for maintaining bone homeostasis, whereas its disruption leads to skeletal pathology. Excessive osteoclast activity underlies disorders such as osteoporosis and Paget's disease, while osteoclast deficiency, either quantitative or functional, results in osteopetrosis. Osteopetrosis comprises a group of genetically heterogeneous and clinically varia

Malignant transformation of a solitary osteochondroma to chondrosarcoma in a cat.

An 8-year-old male neutered Domestic Shorthair cat presented due to left forelimb lameness. Clinical examination revealed a firm, non-mobile protuberance on the craniomedial aspect of the left distal radius. Histological and radiographic findings were consistent with a solitary osteochondroma. Reoccurrence of clinical signs and acute increase in the mass size occurred 1.5 years after diagnosis. A left forequarter amputation was performed due to concerns over malignant transformation. Histology was consistent with chondrosarcoma. Feline osteochondromas are rare, benign osseous neoplasms: they a

Polyostotic Fibrous Dysplasia Mimicking Widespread Skeletal Metastases on 68 Ga-PSMA PET/CT and Demonstrating a Superscan Pattern on Bone Scintigraphy.

Fibrous dysplasia (FD) may demonstrate increased 68 Ga-PSMA uptake and mimic skeletal metastases in prostate cancer. We present a 63-year-old man with newly diagnosed prostate cancer who underwent 68 Ga-PSMA PET/CT for staging. Imaging revealed widespread skeletal uptake involving multiple regions, raising suspicion for diffuse metastatic disease. Bone scintigraphy demonstrated a superscan pattern. However, CT showed characteristic benign features, including ground-glass appearance, bone expansion, and preserved cortical integrity. Histopathologic examination confirmed fibrous dysplasia. This
