221 small-for-dates (SFD) and 244 average-for-dates (AFD) children were personally examined at birth, and seen thereafter at regular intervals up to 4 years, when a thorough assessment was made. There was an excess of SFD children with major congenital abnormalities; but no difference between the groups in the number of children who were handicapped without congenital abnormality. We describe a 15-year-old boy with an ectodermal dysplasia syndrome (EDS) associated with unilateral adrenal cyst. The EDS combination of aplasia cutis verticis, hypohidrosis, nipple/breast hypoplasia, onychodysplasia, and delayed dental eruption with minor tooth anomalies has not been previously reported as an autosomal dominant trait. The association with adrenal cyst may alert other units to review their experience to determine if such combination is more than a coincidence.

Muscinous breast carcinoma occurred in a 67-year-old true hermaphrodite who had been reared as a male. The peripheral blood karyotype was 46, XX. A left ovary and right ovotestis were intra-abdominal. Other tumors included an ovarian thecoma and an intestinal leiomyoma. The chromosomal constitution of 8 human breast tumor cell lines has been analyzed by conventional staining and G-banding methods. The stem line number was established in each case. In all cell lines, a large number of marker chromosomes have been identified. The cytogenetic findings by G- and C-banding in a primary breast carcinoma are reported. The tumor is characterized by a high modal number of chromosomes (79) and 14 markers of ten different origins. Chromosome #1 is more frequently involved in structural and numerical aberrations than is any other chromosome; nine copies of 1q were present in each cell examined. This observation supports the hypothesis that duplication of chromosome 1q may play an important role in the progression of malignant tumors.

In a family with a high incidence of cancer, the proband manifested early-onset colon cancer at age 39 years. His mother, her twin sister, and their daughters had ovarian carcinoma. In the proband's sibship, six of eight had cancer of differing anatomic sites. The susceptibility to ovarian carcinoma appeared to have been transmitted through the men; one was cancer free while two had cancer. Thus, eight close relatives had ovarian carcinoma. Examinations of 51 blood relatives failed to show cutaneous stigmata or congenital abnormalities of known hereditary cancer or precancer syndromes.
