Schistosome-derived microRNAs: promise, pitfalls and priorities for molecular diagnosis.

BACKGROUND: Schistosomiasis remains a major neglected tropical disease with persistent transmission despite decades of control programs. Recent discoveries of schistosome-derived microRNAs (miRNAs) have introduced new opportunities for precise, non-invasive molecular diagnosis. These miRNAs, often encapsulated in extracellular vesicles (EVs) or associated with Argonaute proteins, are stable in host biofluids and reflect active infection, worm burden, and pathological status. Therefore, this commentary aims to summarize current advances in schistosome-derived miRNAs for molecular diagnosis, dis

Schistosoma infection as a cause of myeloradiculopathy: case report from Mozambique and systematic review.

BACKGROUND: Schistosomal myelopathy (SM) is a rare and devastating neurological manifestation of schistosomiasis, predominantly affecting individuals in endemic regions. SM may present a wide range of positive and negative manifestations such as paresthesia, myalgia, low back pain, paraplegia and urinary dysfunction. Limited awareness and lack of access to supportive microbiological and imaging diagnostics means may delay clinical recognition. METHODS: Case report of SM and a systematic review of the literature on its epidemiological patterns and clinico-radiological characteristics, includin

Prevalence and associated factors of non-variceal upper gastrointestinal bleeding among cirrhotic patients with upper gastrointestinal bleeding on follow-up at a Tertiary Hospital in Ethiopia.

BACKGROUND: Liver cirrhosis is a major cause of morbidity and mortality worldwide. Non-variceal upper gastrointestinal bleeding (NVUIGB) accounts for 24-42% of bleeding episodes in cirrhotic patients and carries a mortality rate of 15-30%. Understanding its prevalence and associated factors is critical for prevention and improved outcomes. This study assessed the prevalence and predictors of NVUGIB among cirrhotic patients with upper gastrointestinal bleeding at a tertiary hospital in Addis Ababa, Ethiopia. METHODS: A hospital based cross-sectional study was conducted from December 2020 to De

A male-pheromone-elevated transcription factor ZNF362.1 in female schistosomes determines sexual maturation.

Egg production by female schistosomes drives both transmission and pathology of schistosomiasis, affecting over 200 million people. Female maturation relies on the male-derived pheromone β-alanyl-tryptamine (BATT), but underlying molecular mechanisms are unclear. We identified the BATT-responsive transcription factor gene znf362 as a key regulator of female reproductive development. Functional studies showed that znf362.1, but not znf362.2, is essential for BATT-induced ovary and vitellaria maturation. Single-cell transcriptomics and in situ hybridization revealed up-regulation of znf362.1 in

Fine-scale mapping of Oncomelania hupensis habitats in eastern China using multi-season Sentinel-2 imagery and semi-supervised deep learning.

Accurate mapping of Oncomelania hupensis, the intermediate host of Schistosoma japonicum, is challenging due to sparse field data and dynamic seasonal flooding. We developed SnailMatch, a semi‑supervised deep learning framework combining multi‑seasonal Sentinel‑2 imagery to delineate snail habitats in Guichi, China. The model used 229 field-confirmed snail presence sites, 229 systematically sampled absence sites, and 5,759 unlabeled locations. Twelve cloud-free Sentinel-2 scenes covering all seasons of 2023 are processed through an attention-based Multi-Seasonal Fusion module that captures sub

Olfactory gene profiling in ticks: A scoping review with a functional perspective.

Olfaction is important in ticks' host-seeking and mating behaviors, as it enables them to detect volatile chemical compounds. This scoping review aims to systematically map and synthesize the existing literature on olfactory gene families in ticks and to examine the application of functional genomics or behavioral approaches. We conducted a thorough search in five databases for publications up to 19 September 2025. This scoping review was implemented according to the Systematic Reviews and Meta-Analyses extension for Scoping Reviews (PRISMA-ScR) guidelines. A total of 15 studies were included

Expanding the frontiers of schistosomiasis treatment: The promise of nanoparticle-based therapies.

Schistosomiasis is one of the most overlooked parasitic diseases worldwide. The new roadmap for neglected tropical diseases (2021-2030), adopted by the World Health Assembly, sets global goals for combating these diseases. eliminating schistosomiasis. Current treatment options for schistosomiasis, such as Praziquantel, are limited. Nanoparticle-based treatments and vaccines have shown promise in overcoming the limitations of Praziquantel therapy by offering targeted and sustained drug delivery. We advocate prioritizing and enhancing funding and collaboration for nanoparticle research targeting

TRPtracker: a community database for monitoring praziquantel sensitivity at TRPMPZQ variants.

The anthelmintic praziquantel (PZQ) has been used for decades as the clinical therapy for schistosomiasis, and remains the only available drug. As a cheap and effective drug therapy for all human disease-causing Schistosoma species, usage of PZQ underpins mass drug administration strategies aimed at eliminating schistosomiasis as a public health problem by 2030. Concern over the potential emergence of resistance to PZQ is therefore warranted, as it would constitute a major threat to this approach. In terms of molecular adaptations conferring PZQ resistance, variation in the sequence and/or exp

YAP1 in control: how RNA networks and protein modifications shape its function and therapeutic targetability.

YAP1 (Yes-associated protein 1), a central downstream effector of the Hippo signaling pathway, is tightly regulated through coordinated post-transcriptional and post-translational mechanisms. At the post-transcriptional level, expression stability and translational output of YAP1 are governed by competitive crosstalk among non-coding RNAs, modulation by RNA-binding proteins, and diverse mRNA modification processes. At the post-translational level, protein stability and subcellular distribution are finely controlled by an integrated modification landscape, including ubiquitination, acetylation,

Wiring love governs female reproductive fate in schistosomes.

Male regulation of female reproductive development in schistosomes depends on an intact reproductive neural network. You et al. identify a limbic system-associated membrane protein as essential for maintaining this network, enabling the efficient release of the pheromone β-alanyl-tryptamine dipeptide, which promotes female maturation.

Potential antidepressant properties of aminophylline in male mice exposed to chronic restraint stress.

BACKGROUND: Aminophylline, a bronchodilator used for treating airway obstruction, has been predicted through in silico models to exert antidepressant effects. However, there are still no studies that have validated these claims in a biological system. In this paper, we evaluated the antidepressant effects of aminophylline in mice subjected to chronic restraint stress (CRS). METHODS: CRS was conducted for a duration of 15 days, with 4-hour daily stress exposure. Aminophylline (5 mg/kg, 10 mg/kg, and 20 mg/kg) and fluoxetine (10 mg/kg, positive control) were administered daily via intraperitone

Endocytosed lipids induce cell aggregation via filopodia retraction in a close relative of animals.

Capsaspora owczarzaki is a protist that may both reveal aspects of animal evolution and curtail the spread of schistosomiasis. Capsaspora exhibits a regulated aggregative behavior reminiscent of cellular aggregation in some animals-a process that might have contributed to the origin of animals. This aggregative behavior may also be vital for Capsaspora to colonize the intermediate host of parasitic schistosomes and potentially prevent the spread of schistosomiasis. Both applications demand elucidation of the mechanisms underlying Capsaspora aggregation. Toward this goal, we evaluated the chemi

PfVPS4, an ESCRT AAA-ATPase, is essential for asexual proliferation and gametocyte sexual conversion in Plasmodium falciparum.

BACKGROUND: Malaria, caused by Plasmodium spp., remains a major global health threat. Among them, Plasmodium falciparum is the most pathogenic, and its asexual intraerythrocytic proliferation is the pathological basis. This process has enormous biosynthetic demands and highly relies on the coordinated function of the endomembrane and vesicular transport systems. The transition from asexual proliferation to sexual differentiation similarly involves remodeling of internal membrane complexes, membrane reshaping, and precise protein sorting. In eukaryotic cells, the Endosomal Sorting Complexes Req

Dual-Physical-Field Nanocatalysis: Injectable Hydrogel Enables Piezo-Photothermal Synergy for Breast Cancer Therapy.

Piezocatalytic therapy (PCT) harnesses mechanical energy to generate tumor-lethal reactive oxygen species (ROS), but its efficacy is limited by rapid electron-hole recombination and poor intratumoral retention. To overcome these limitations, we engineered heterostructured BiOCl@CuO nanosheets embedded in an injectable, conductive, thermosensitive hydrogel composed of Pluronic F127 (F127) and reduced graphene oxide (rGO), with rGO specifically enhancing the hydrogel's mechanical strength and conductivity, for the combinational treatment of breast cancer. Under ultrasound (US) stimulation, BiOCl

Access to praziquantel in Italy: persistent gaps and ongoing responses amid a Europe-wide shortage.

In Italy, praziquantel (PZQ) is not registered for human use and can be imported from European countries where it is authorized, according to a specific ministerial decree. Recent Europe-wide shortages, however, have made this route impractical, resulting in the unavailability of this essential medicine. This editorial summarizes historical gaps in supply, the actions undertaken, and the interim solutions implemented to restore PZQ access in Italy. The Italian experience highlights broader challenges related to essential medicines and underscores the need for coordinated national and European

Cecal schistosomiasis with perforation: case report and literature review.

Schistosomal cecum involvement is an extremely rare condition in endemic areas like Sudan. We report a 17-year-old male presenting with acute abdomen diagnosed preoperatively as perforated appendicitis. Emergency surgery revealed purulent peritonitis, a cecal mass, and a large cecal perforation and right hemicolectomy was done. Histopathology revealed viable schistosomal ova and granulomas in the cecum. Post-operative management included anthelmintic therapy. This case presented the third case of large bowel schistosomiasis in Sudan, emphasizing the need for a high index of suspicion for paras

Comparative mitogenomics of freshwater snails of the genus Biomphalaria (Gastropoda: Planorbidae) and new insights into mitochondrial genome evolution in Hygrophila.

BACKGROUND: Freshwater snails of the genus Biomphalaria include several species of major public health importance as intermediate hosts of schistosomiasis and represent a key group for evolutionary and taxonomic research within Hygrophila. However, complete mitochondrial genomes are available for only a limited number of Biomphalaria species, restricting comparative assessments of mitogenome evolution and the ability of mitogenomic data to resolve contentious relationships within Planorbidae and across Hygrophila. Here, we generate and analyse six complete mitochondrial genomes from four Neotr
