Gene Editing of a Carcinogenic Liver Fluke Tetraspanin Impairs Parasite Surface Biogenesis and Extracellular Vesicle Uptake by Human Host Cells.

Opisthorchiasis remains a significant public health concern throughout Southeast Asia. The liver fluke Opisthorchis viverrini resides within the biliary tract, and chronic infection leads to bile duct cancer, or cholangiocarcinoma. Here, we examined the functions of liver fluke tetraspanins, 4-transmembrane domain proteins expressed on the surface of the fluke tegument, and extracellular vesicles (EVs) derived from this syncytial surface. We undertook CRISPR-Cas9 gene knockout (KO) of the O viverrini tetraspanin 2 (Ov-tsp-2) gene and found that Ov-tsp-2-KO flukes had abnormal tegument biogenes

Effect of water temperature on life history traits of Radix natalensis, the intermediate host of Fasciola gigantica.

BACKGROUND: Radix natalensis, a gastropod, serves as an intermediate host of Fasciola gigantica in tropical and subtropical regions, causing fascioliasis, a disease of public health and veterinary importance worldwide. Predicting disease burden requires understanding how climate change influences its life-history traits. This study investigated the effects of temperature on the life-history traits of R. natalensis. Snails were exposed to six temperature treatments (15.5 ℃, 19.5 ℃, 20.3 ℃ (control), 24.6 ℃, 29.8 ℃, and 35.1 ℃). For each treatment, 10 jars served as replicates, each containing t

An alien intermediate host snail, Radix rubiginosa (Gastropoda: Lymnaeidae), on Unguja Island, Zanzibar.

In June 2025, two unusual populations of lymnaeid snails were encountered within peri-urban freshwater habitats of Unguja Island, Zanzibar (United Republic of Tanzania). These were noted within a wider malacological survey for freshwater intermediate host snails across twenty-one locations. Using an integrative taxonomic approach combining conchology and anatomy (n = 21) with mitochondrial 16S rDNA (n = 8) and mitochondrial cox1 (n = 3) sequence information, these lymnaeids were identified as Radix rubiginosa (Michelin, 1831). This is a first observation of this Southeast Asian alien species i

The Liver Fluke Opisthorchis felineus Exosomal tRNA-Derived Small RNAs as Potential Mediators of Host Manipulation.

The role of extracellular vesicle non-coding RNAs in host-parasite interactions remains poorly understood, particularly for human liver flukes. Although tRNA-derived small RNAs (tsRNAs) are emerging as new regulatory molecules in parasite exosomes, they have not yet been characterized for the liver flukes. We performed small RNA sequencing to profile tsRNAs in the exosome-like vesicles derived from the liver fluke Opisthorchis felineus. Transcriptomic data from human cholangiocytes were analyzed to assess the enrichment of the predicted target genes among differentially expressed genes. We ide

Spatial analysis of sheep fascioliasis in rural communities of the Southern highlands of Peru.

Sheep fascioliasis causes considerable economic losses globally, yet risk factors for infection in Andes Highlands remain poorly characterized. We conducted a cross-sectional study to determine the risk factors of sheep fascioliasis in rural communities of the Southern highlands of Peru. Data on farm and animal characteristics, management practices, and water sources were collected. Stool samples were geographically tagged and examined microscopically for Fasciola and other parasites eggs. A total of 3,518 sheep from 268 farms were evaluated, of which 40.6% (1,429/3,518) were infected with F.

Adult Fasciola recovered from the biliary tree in a patient presenting with possible ascending cholangitis and acute acalculous cholecystitis.

BACKGROUND: Fascioliasis is a zoonotic trematode infection caused by members of the genus Fasciola (F. hepatica and F. gigantica) that localize to the hepatic biliary ducts in humans. Parasites are acquired after ingestion of freshwater plants, and fascioliasis has been reported on every inhabited continent. Typically, fascioliasis is diagnosed by serology or stool examination. This case reports the unusual finding of adult Fasciola organisms during an endoscopic retrograde cholangiopancreatography (ERCP). CASE SUMMARY: A 65-year-old woman who returned to the United States after traveling in

Paucisymptomatic opisthorchiasis in a non-endemic country: a neglected differential diagnosis?

CASE PRESENTATION: We report a case of opisthorchiasis in a 71-year-old woman with intermittent upper abdominal pain, whose cause had remained unclear for a long time. She grew up in Kazakhstan and moved to Germany more than 30 years ago. After positive serological testing for Opisthorchis during a recent visit to her home country, she presented at our clinic for further evaluation. Stool examination confirmed the presence of Opisthorchis eggs. The symptoms quickly resolved after treatment with praziquantel. Abdominal sonography revealed adenomyomatosis of the gallbladder fundus and multiple g

From Allozymes to Genomics: Reframing the Systematics and Population Structure of Opisthorchis viverrini and Its Bithynia Hosts.

The carcinogenic liver fluke Opisthorchis viverrini underlies one of the world's heaviest burdens of bile duct cancer, yet for decades it was treated as a single, genetically uniform parasite whose transmission was shaped mainly by environment and human behavior. However, advances in molecular biology have fundamentally reshaped this conceptual model. Evidence accumulated over the past three decades demonstrates that O. viverrini comprises geographically structured populations linked to hydrological connectivity, host distribution, and long-term evolutionary processes across interconnected riv

Projected climate change effects on suitable habitat for Pseudosuccinea columella and Radix natalensis in uMgungundlovu, South Africa.

INTRODUCTION: Climate change impacts oceanic and atmospheric circulation patterns, precipitation, air and sea surface temperatures leading to altered intermediate host vector distribution, reproduction, and maturation. With temperatures predicted to rise by 1.5°C or more, understanding the influence of environmental characteristics and climatic conditions on distribution of intermediate host vectors, occurrence and habitat suitability is vital in vector borne diseases and their control policies. Hence, the study focused on the predicted changes in habitat suitability of Pseudosuccinea columell

Genetic diversity and host-symbiont cophylogenetic analysis of Neorickettsia endobacteria of Fasciola hepatica.

Neorickettsia are obligate intracellular bacteria that persist as endosymbionts of digenean trematodes and include several vertebrate pathogens. However, many digenean-associated lineages remain poorly characterized, including Neorickettsia associated with the liver fluke Fasciola hepatica (nFh). Here, we present a large-scale population genomic analysis of nFh to assess its prevalence, genetic diversity, evolutionary history, and transmission dynamics. We screened whole-genome sequencing data from 502 F. hepatica samples collected across Peru, the UK, Uruguay, and the USA, detecting Neoricket

The Impact of "The Magic Glasses Opisthorchiasis" on Schoolchildren's Knowledge, Attitudes and Practices Surrounding Opisthorchis viverrini in the Lower Mekong Basin, a Cluster-Randomised Controlled Trial.

Opisthorchis viverrini (OV) is a liver fluke endemic to the Lower Mekong Basin. Infections often begin in childhood and are causally linked to cholangiocarcinoma, an often-fatal bile duct cancer. Anthelmintic treatment is the primary control strategy, but infection can recur. Therefore, additional strategies are needed. This study assessed the impact of "The Magic Glasses Opisthorchiasis" (MGO), a cartoon-based intervention, on schoolchildren's OV-related knowledge, attitudes and practices (KAP). A cluster (school)-randomised controlled trial was conducted in Cambodia, Laos and Thailand. Clust

The hidden passengers: On the role of exotic crayfish in the spread of symbiotic and pathogenic organisms in Northern Iberian Peninsula.

The spread of invasive crayfish species poses a growing threat to freshwater ecosystems and public health, not only through direct ecological impacts but also by facilitating the transmission of symbionts and potential zoonotic pathogens. This study characterizes the symbiont diversity associated with the crayfish species that occur in northern Spain. In addition, the first comprehensive and comparative compilation of reported symbiont and parasite species of crayfish species reported in Iberia is provided. Paragonimiasis Following Ingestion of Frozen Freshwater Crab: Two Case Reports.

Paragonimiasis is a zoonotic disease caused by trematodes belonging to the genus Paragonimus. It is acquired by ingesting freshwater crustaceans or raw game meat. Here, we report two patients (a couple) who developed paragonimiasis and pulmonary lesions with peripheral eosinophilia after sharing and consuming previously frozen freshwater crabs (Eriocheir japonica) together at the same restaurant in Tokyo. One patient developed peripheral airspace consolidation, which was initially misdiagnosed as chronic eosinophilic pneumonia, whereas the other exhibited eosinophilic pleural effusion. Antipar
