Astral Microtubules Are Dispensable for Pavarotti Localization During Drosophila Spermatogonial Mitoses.

We analysed here the dynamic of the kinesin-like Pavarotti (Pav) during male gametogenesis of wild-type and Sas4 mutant flies. Pav localizes to the equatorial region and the inner central spindle of late anaphase wild-type spermatogonia and displays a strong concentration at the midbody during late telophase. At metaphase of the first meiotic division, Pav shows widespread localization on the equatorial region of the spermatocytes. This unusual distribution restricts and enhances during anaphase where antiparallel cortical microtubules overlap. Additional Pav staining is also found in the inne

Cytogenomic Characterization of Murine Cell Line Sarcoma 180 = S-180.

The cell line Sarcoma 180, which is also called S-180 (or S180), was established about 110 years ago from a murine axillary sarcoma. It has been applied in >5000 studies but was never genetically characterized in detail; this study fills that gap. The cell line Sarcoma 180 was analyzed for its chromosomal constitution using molecular cytogenetic approaches, specifically murine multicolor banding (mcb). Additionally, array comparative genomic hybridization was performed to characterize copy number alterations. Sarcoma 180 has a near tetraploid karyotype without Y-chromosome material and only tw

RENEB interlaboratory comparison for biological dosimetry based on dicentric chromosome analysis and cobalt-60 exposures higher than 2.5 Gy.

In previous RENEB interlaboratory comparisons based on the manual scoring of dicentric chromosomes, a tendency for systematic overestimation for doses > 2.5 Gy was found. However, these exercises included only very few doses in the high dose range, and they were heterogeneous in terms of radiation quality and evaluation mode, and comparable only to a limited extent. Here, this presumed deviation was explored by investigating three doses > 2.5 Gy. Blood samples were irradiated (2.56, 3.41 and 4.54 Gy) using a 60Co source and sent to 14 member laboratories of the RENEB network, which performed t

Centromeres are stress-induced fragile sites.

Centromeres are unique loci on eukaryotic chromosomes and are complexed with centromere-specific histone H3 molecules (CENP-A in mammals, Cse4 in yeast). The centromere provides the binding site for the kinetochore that captures microtubules and provides the mechanical linkage required for chromosome segregation. Centromeres encounter fluctuations in force as chromosomes jockey for position on the metaphase spindle. We have developed biological assays to examine the response of centromeres to high force. Torsional stress is induced on covalently closed DNA circles from supercoiling. Plasmid-bo

Enhancing clinical decision-making for CNVs of uncertain significance in neurodevelopmental disorders: the relevance (or uselessness) of scoring and segregating.

BACKGROUND: Clinicians often deal with copy-number variants of unknown significance (CNVUS) when managing neurodevelopmental disorders (NDDs). Variant classification is often complemented with textual comments, while the American College of Medical Genetics and Genomics (ACMG)/Clinical Genome Resource (ClinGen) numerical scores are rarely reported. Our aim was to determine if the application of ACMG/ClinGen scoring and inheritance/segregation studies are relevant for the reclassification of CNVUS. METHODS: We retrieved 167 CNVUS (112 duplications, 55 heterozygous deletions) from test reports

Dose-response curve for induction of unstable chromosome aberrations by 6 MV linear accelerator photons: Analysis of intra-experimental variations.

Cytogenetic biodosimetry relies on dose-response curves (DRCs) for each type of radiation that can cause a radiation emergency. We have constructed a DRC based on the dicentric assay. Blood samples from four healthy volunteers were irradiated with acute 6 MV linac photons, 0.46-4.55 Gy; 0.68 and 1.37 Gy doses were used in the 'blind' validation study. Lymphocytes were cultured with variations in time delay in mitogenic stimulation after irradiation (2 vs. 16 h) and mitotic arrest by colchicine (3.5 vs. 16 h). Aberrations were scored in the first division metaphases, ensured by fluorescence-plu

Linear dicentric chromosomes in bacterial natural isolates reveal common constraints for replicon fusion.

UNLABELLED: Multipartite bacterial genome organization can confer advantages including coordinated gene regulation and faster genome replication but is challenging to maintain. Agrobacterium tumefaciens lineages often contain a circular chromosome (Ch1), a linear chromosome (Ch2), and multiple plasmids. We previously observed that in some stocks of the lab model strain C58, Ch1 and Ch2 were fused into a linear dicentric chromosome. Here we analyzed Agrobacterium natural isolates from the French Collection for Plant-Associated Bacteria (CFBP) and identified two strains with fused chromosomes. C

Relative biological effectiveness of 31 meV thermal neutrons in peripheral blood lymphocytes.

The reported relative biological effectiveness (RBE) for thermal neutrons has a large range (5-51, for cytogenetic endpoints), which can confound radiation protection decision-making. To determine whether thermal neutron spectra can influence RBE, the RBE of reactor-derived thermal neutrons of average energy 31 meV was evaluated in human peripheral blood lymphocytes using two classical DNA double-strand break endpoints: the dicentric chromosome assay (DCA) and the cytokinesis-block micronucleus assay. Dose-response curves for 41 to 408 mGy revealed a preference for linear regression. Maximum R

Germline mosaicism in TCF20-associated neurodevelopmental disorders: a case study and literature review.

Autosomal dominant variants in transcription factor 20 (TCF20) can result in TCF20-associated neurodevelopmental disorder (TAND), a condition characterized by developmental delay and intellectual disability, autism, dysmorphisms, dystonia, and variable other neurological features. To date, a total of 91 individuals with TAND have been reported; ~67% of cases arose de novo, while ~10% were inherited, and, intriguingly, ~8% were either confirmed or suspected to have arisen via germline mosaicism. Here, we describe two siblings with a developmental condition characterized by intellectual disabili

Automated system for establishing standard radiation dose-response curves and dose estimation for the Korean population.

Biological dosimetry is crucial for estimating the doses from biological samples and guiding medical interventions for accidental radiation exposure. This study aimed to derive rapid and precise dose estimates using a dicentric chromosome assay. To address the challenges of manual scoring of dicentric chromosomes, we upgraded an automatic system aimed at enhancing the precision of dicentric chromosome detection while reducing the need for human intervention. We collected blood from 30 individuals aged 20-67 years to create 30 dose-response curves aiming to investigate the differences in respon

Case report: The smallest 9p21.3 microdeletion involving CDKN2A but not CDKN2B causes multiple plexiform neurofibromas.

Chromosome 9p21.3 is a locus associated with a rare autosomal dominant cancer predisposition syndrome characterized by early-onset melanoma and a broad spectrum of neural system tumors. Two major tumor-suppressor genes, cyclin-dependent kinase inhibitor 2A and 2B (CDKN2A and CDKN2B), as well as a large non-coding RNA ANRIL, are often co-deleted in the core region. Herein, we report a pregnant woman who had developed more than 20 plexiform neurofibromas since the age of 13 and experienced 11 times of surgical resections. No melanoma or other tumors were found. A germline 9p21.3 deletion involvi

Cytogenomic characterization of karyotypes with additional autosomal material.

Chromosomal rearrangements involving additional material in individuals with phenotypic alterations usually result in partial trisomy, often accompanied by partial monosomy. To characterize chromosomal rearrangements and analyze genomic characteristics in the breakpoint regions in 31 patients with additional material on an autosomal chromosome. Different tests were performed to characterize these patients, including karyotyping, chromosomal microarray analysis (CMA), and fluorescent in situ hybridization (FISH). In silico analyses evaluated A/B chromosomal compartments, segmental duplications,

Novel MSH6 exon 5-6 skipping variant in a Taiwanese family with Lynch syndrome: implications for genetic testing and cancer management.

Lynch syndrome is an autosomal dominant disorder predisposing individuals to colorectal and other cancers, primarily caused by variants in mismatch repair genes. This study describes a novel MSH6 variant affecting transcript structure in a Taiwanese family meeting the Amsterdam II criteria for Lynch syndrome. A 67-year-old male presented with jejunal adenocarcinoma and a strong family history of colorectal cancer. Immunohistochemistry revealed loss of MSH6 expression, while next-generation sequencing performed on tumor tissue failed to detect any MSH6 variants. Comprehensive genetic analysis,

Acute Myeloid Leukemia with MYC Amplification on a Ring Chromosome 8.

MYC amplification and overexpression are uncommon in acute myeloid leukemia (AML). An 82-year-old man developed leukocytosis during monoclonal gammopathy of renal significance. A chromosomal analysis revealed 46,XY,+r(8)[20]. Amplified MYC signals were detected on chromosome 8. The patient was diagnosed with AML and administered venetoclax and azacitidine. After the third course, clones with ring chromosome 8 had decreased in number, but clones unrelated to t(8;21)(q22;q22) had subsequently emerged. After the sixth course, the white blood cell count had markedly increased, and a chromosome ana

A zebrafish model of crim1 loss of function has small and misshapen lenses with dysregulated clic4 and fgf1b expression.

INTRODUCTION: Heterozygous deletions predicting haploinsufficiency for the Cysteine Rich Motor Neuron 1 (CRIM1) gene have been identified in two families with macrophthalmia, colobomatous, with microcornea (MACOM), an autosomal dominant trait. Crim1 encodes a type I transmembrane protein that is expressed at the cell membrane of lens epithelial and fiber cells at the stage of lens pit formation. Decreased Crim1 expression in the mouse reduced the number of lens epithelial cells and caused defective adhesion between lens epithelial cells and between the epithelial and fiber cells. METHODS: We

Horizontal transfer of nuclear DNA in transmissible cancer.

Horizontal transfer of nuclear DNA between cells of host and cancer is a potential source of adaptive variation in cancer cells. An understanding of the frequency and significance of this process in naturally occurring tumors is, however, lacking. We screened for this phenomenon in the transmissible cancers of dogs and Tasmanian devils and found an instance in the canine transmissible venereal tumor (CTVT). This involved introduction of a 15-megabase dicentric genetic element, composed of 11 fragments of six chromosomes, to a CTVT sublineage occurring in Asia around 2,000 y ago. The element fo

Establishment and verification of the dose-effect curve of dicentric chromosomes by semi-automatic analysis of human peripheral blood irradiated by X-ray in vitro.

OBJECTIVE: To establish and validate a dose-response curve for dicentric chromosomes (DC) induced by X-rays in human peripheral blood in vitro using semi-automated scoring. METHODS: Peripheral blood samples were collected from three healthy individuals and exposed to X-ray doses of 0, 0.25, 0.5, 0.75, 1, 2, 3, 4, and 5 Gy at a dose rate of 1.158 Gy/min. Dicentric chromosomes in metaphase were scored both full- and semi-automatically, and a dose-response curve was generated with CABAS software based on dicentric yields. Dose estimations were then performed for 12 biodosimetry standard samples

Establishment and validation of a DIP panel for forensic ancestry inference and personal identification.

BACKGROUND: Biallelic Deletion/Insertion polymorphisms (DIPs), known for their significant diversity across various populations, serve as valuable markers for forensic ancestry inference and personal identification. In this study, we utilized DIPs to provide a potentially powerful forensic examination tool specifically tailored for East Asian populations. Our focus on ancestry allows us to delve deeper into the genetic signatures that characterize this diverse group, offering enhanced resolution in forensic analyses. METHODS: A total of 56 autosomal DIPs, 3 Y-chromosome DIPs, and the Amelogen
