Fasciolosis, a disease induced by liver fluke infection, poses a major threat to the livestock industry. The emergence of drug resistance and the lack of commercial vaccines necessitate the development of novel control strategies. Studies on helminth immunoregulatory molecules have become a key research focus. Bone morphogenetic proteins (BMPs), ligand members of the TGF-β superfamily, have been identified in Fasciola gigantica and designated FgBMPs, and are hypothesized to regulate host immune responses. To explore the effects of FgBMP on hosts, the mechanistic actions of FgBMP-1 on buffalo p

The study was conducted from October 2024 to June 2025 at the Kombolcha ELFORA abattoir and Processing Plant to assess the prevalence and economic impact of bovine Fasciolosis in cattle sourced from three agroecological zones in northern Ethiopia. From the 382 examined bovine samples, 184 faecal samples (48.17%) and 195 livers (51.05%) tested positive for Fasciolosis. Infection rates were highest in cattle with poor body condition (51.55% in faecal samples and 54.64% in livers), followed by medium (47.62% and 50%) and good body condition groups (20% and 25%, respectively), with significant dif

BACKGROUND: The earliest interactions between Fasciola hepatica newly excysted juveniles (FhNEJ) and the intestinal epithelium remain poorly understood, partly due to the limited availability of physiologically relevant in vitro models. Here, we establish a human duodenum organoid-derived epithelial monolayer (ODM) co-culture system to investigate the early host-parasite interface, providing a controlled experimental platform for analysing interactions between FhNEJ and the intestinal epithelium.
METHODS: ODMs representing the human duodenum were co-cultured with either live FhNEJ or a tegume

Fasciolosis is a food-borne disease affecting livestock worldwide. Improved diagnostic methods for detecting fasciolosis have been introduced; however, limited research has compared these methods. The current study aimed to compare the detection of Fasciola in naturally infected slaughtered dairy cattle using four methods. Blood and faecal samples were collected from slaughtered cattle in three high-throughput abattoirs in the Eastern Cape Province, South Africa, from 2020 through 2021. Post-mortem liver examination was conducted to detect and identify Fasciola spp. Blood from slaughtered carc

Fascioliasis, a zoonotic parasitic disease caused by the liver flukes Fasciola hepatica and Fasciola gigantica, poses a significant threat to livestock and human health globally. Climate change is altering the transmission dynamics of this disease; however, the direction and magnitude of this change remain uncertain due to a lack of mechanistically grounded predictions at continental scale. In this study, we developed a novel risk index to assess how climate change may affect the potential for fascioliasis transmission across Africa and Europe. The index integrates the following three componen

Fasciolosis caused by Fasciola hepatica is an economically important parasitic disease of livestock and a major zoonotic disease in humans. Vaccination is a promising strategy to reduce the impact of fasciolosis and dependence on fasciolicides. Saposin-like protein 2 (FhSAP2) has previously shown high immunogenicity and reductions in fluke burden in experimental animal models; however, its efficacy has not been evaluated in sheep. Twenty-one female sheep were randomly assigned to three groups (n = 7): vaccinated, adjuvant control, and infection control. The vaccinated group received 400 μg of

INTRODUCTION: The liver fluke, Fasciola hepatica, is considered one of the principal issues affecting the health and productivity of the UK sheep population. Diagnostic testing is advised to ensure appropriate use of treatment. A new lateral flow test to detect antibodies to liver fluke has recently been developed for use by farmers. Point of care rapid diagnostic tests are rare in agriculture so this represents a novel innovation in this sector.
METHODS: Prototype test kits and accompanying guidance were taken to six farms for initial usability evaluation, before commercial launch of the tes

In a dairy herd in the state of Goiás, Central-West Brazil, a cow died following an acute illness characterized by jaundice and hemoglobinuria. Necropsy revealed jaundice, dark-red urine and dark-red discoloration of the kidneys, and a diffusely copper-colored liver with two extensive, firm lesions surrounded by hyperemic halos in the right and left lobes. Histopathological analysis showed multifocal coagulative necrosis in the liver, accompanied by abundant intralesional bacillary structures and multifocal thrombosis. Renal lesions included tubular cell degeneration and necrosis, intracellula

Triclabendazole (TCLZ) is a halogenated benzimidazole anthelmintic widely used in humans and animals to treat fascioliasis which acts by disrupting microtubule-dependent processes in the species of Fasciola. This study aims to characterize TCLZ's degradation profile under different degradation conditions and assess the toxicological potential of its degradation products (DPs). TCLZ was subjected to forced degradation as required by the ICH Q1A(R2) and Q1B guidelines, including oxidation (H2O2), photolysis (UV and visible light), thermal, and hydrolysis (acidic, basic, and neutral) conditions.

Hepatic fascioliasis is a foodborne zoonosis that can mimic hepatobiliary malignancy and occasionally requires surgery when diagnosis is uncertain or when a focal complicated lesion is not amenable to endoscopic or medical management. We report 2 cases managed at a military-associated surgical center in Ukraine involving civilian employees of the Armed Forces of Ukraine working in rear-area, non-combat facilities. Case 1: a 38-year-old man with more than 1 year of right upper quadrant pain and a subcapsular tract-like lesion in hepatic segment VI. Eosinophils and cholestatic enzymes were not e

The introduction of exotic species has major impacts on biodiversity, ecosystems, the economy, and public health. A less explored issue is the silent co-invasion of parasites associated with introduced species, often overlooked due to their low detectability. Among these, helminths (e.g., platyhelminths and nematodes) introduced with terrestrial and freshwater gastropods are especially important, given the adaptability and ease of transport and their capacity to act as intermediate hosts or vectors of many parasites capable of infecting humans. Using the Iberian Peninsula as an study example,

Fascioliasis, a globally prevalent zoonosis, severely threatens public health and livestock security. Current diagnostic approaches, hindered by the need for sophisticated instrumentation and specialized expertise, are inadequate for on-site surveillance in resource-constrained settings. This study developed a rapid, visual detection assay for Fasciola hepatica via recombinase-aided amplification (RAA) integrated with CRISPR/Cas12b, addressing critical equipment and operational constraints. Targeting a specific mitochondrial DNA fragment of F. hepatica, recombinant plasmid standards were const

BACKGROUND: Fascioliasis, caused by Fasciola hepatica and Fasciola gigantica, is a global veterinary problem in livestock and an emerging zoonotic disease in various countries. Here we present prevalence estimates of Fasciola spp. in all host and environmental compartments involved in the life cycle and identified risk factors associated with Fasciola transmission in a rural community in north-central Vietnam.
METHODOLOGY/PRINCIPAL FINDINGS: We conducted a cross-sectional survey in a community in Nghe An province where fascioliasis is reported to be endemic and inhabitants commonly consume ra

BACKGROUND: Radix natalensis, a gastropod, serves as an intermediate host of Fasciola gigantica in tropical and subtropical regions, causing fascioliasis, a disease of public health and veterinary importance worldwide. Predicting disease burden requires understanding how climate change influences its life-history traits. This study investigated the effects of temperature on the life-history traits of R. natalensis. Snails were exposed to six temperature treatments (15.5 ℃, 19.5 ℃, 20.3 ℃ (control), 24.6 ℃, 29.8 ℃, and 35.1 ℃). For each treatment, 10 jars served as replicates, each containing t

In June 2025, two unusual populations of lymnaeid snails were encountered within peri-urban freshwater habitats of Unguja Island, Zanzibar (United Republic of Tanzania). These were noted within a wider malacological survey for freshwater intermediate host snails across twenty-one locations. Using an integrative taxonomic approach combining conchology and anatomy (n = 21) with mitochondrial 16S rDNA (n = 8) and mitochondrial cox1 (n = 3) sequence information, these lymnaeids were identified as Radix rubiginosa (Michelin, 1831). This is a first observation of this Southeast Asian alien species i
