Mosaic loss of the Y chromosome (LOY) is a somatic phenomenon that occurs in males as they age (Jacobs, 1963; Forsberg, 2017; Danielsson, 2020) and is associated with an elevated risk of developing numerous diseases, including Alzheimer's Disease (AD) (Dumanski, 2016; Palmer, et al., 2022; Garcia-Gonzalez, 2023), cancer (Holmes, 1985; Loftfield, 2018), diabetes (Loftfield, 2018), age related macular degeneration (Grassmann, 2019), and autoimmune disease (Persani, 2012; Lleo, 2013). The induced pluripotent cell lines (iPSCs) UMi050-A and UMi050-B were derived from a Hispanic male with loss of t

The human Y chromosome (ChrY), which confers male sex determination, contains a relatively small number of protein-coding genes compared to other chromosomes; consequently, its functional impact on adult physiology was once severely unappreciated. While the repetitive structure of the ChrY once impeded sequencing, technological advances have now made it possible to identify its contents. Despite the historical view of ChrY as a virtual wasteland, we now know that it encodes a variety of genes which are hugely consequential to both human health and disease. The extreme downregulation of ChrY ge

Males and females show significant differences in body structure, typical behavior, average life expectancy, and disease susceptibility. This review article introduces the current understanding and recent findings on the factors that lead to sexual dimorphism. First, recent studies have shown that sex chromosomes underlie male- and female-specific phenotypes through various mechanisms. For example, X chromosome inactivation exerts both positive and negative effects on female health, independent of sex hormone actions. Furthermore, differences in the frequency and clinical consequences of the m

Individuals with Turner syndrome (TS) phenotypes may exhibit short stature, ovarian dysfunction, and neurocognitive disorders. Their genomes can include ring chromosomes formed from the X chromosome (RCX). Here, we present cytogenomic and clinical findings from seventeen individuals with TS who bore mosaic forms of RCX and frequently presented with short stature and concern for TS. The subjects were retrospectively included and tested at a single-site cytogenetics laboratory for over 42 years. Here, we illustrate each subject's comprehensive cytogenetic workup and phenotypes. The cohort shows

Prenatal cell-free DNA (cfDNA) screening and carrier screening (CS) may have incidental findings that have implications for maternal health outside the scope of the test. We investigated outcome information for individuals with both a DMD full gene deletion/duplication on CS and a suspected maternal X chromosome abnormality on SNP-based prenatal cfDNA screening. This retrospective analysis included de-identified data from pregnant individuals referred for CS and prenatal cfDNA screening at a single reference laboratory (9/2019-12/2021). Maternal karyotype and/or chromosomal microarray analysis

BACKGROUND: Chromosomal abnormalities are a significant cause of miscarriages. Carriers of balanced chromosome rearrangement are often at risk of recurrent pregnancy loss (RPL), as they are more likely to produce gametes with unbalanced chromosome rearrangements.
OBJECTIVE: This study evaluated the chromosomal abnormalities detected in couples with history of primary recurrent pregnancy loss.
DESIGN: Retrospective, cross-sectional study.
SETTING: Single center, tertiary healthcare center in Turkey.
PATIENTS AND METHODS: This study reviewed conventional cytogenetic/molecular cytogenetic ana

Background/Objectives: Ring chromosomes (RCs) can be rare or common depending on the chromosome involved. With interest in the historical RCs identified by our laboratory, we curated instances to provide further information to this research field. Methodology: We carried out a retrospective, single-center study of constitutional RCs identified starting in the late 1980s. Details for 40 RCs with a modal number of 46 chromosomes are featured here. Results: Mosaic and non-mosaic RCs are identified, with a preponderance of pediatric-aged females at first ascertainment. We corroborated an enrichmen

RESEARCH QUESTION: Can algorithms be developed to assess maternal contamination in the embryo spent culture medium without parental samples and correct copy number variations (CNV)?
DESIGN: An algorithm was developed to confirm the proportion of maternal contamination based on the mosaic ratios of X and Y chromosomes and an algorithm to correct copy number variation (CNV) in embryos with maternal contamination. Performance of algorithms was tested in 317 culture medium samples. A prospective clinical study was then conducted to validate the accuracy.
RESULTS: Among 55 male embryos with mater

BACKGROUND: Organochlorine (OC) insecticides are a class of environmentally persistent chemicals linked to risk of several cancers, including non-Hodgkin lymphoma and prostate cancer. In vitro and animal studies suggest some OCs may be genotoxic, but evidence in humans is limited. Mosaic loss of the Y chromosome (mLOY) is a marker of genotoxicity and genomic instability that has been associated with certain cancers and may reflect intermediate effects of pesticide exposure. We examined associations between OC use and mLOY in circulating blood of male farmers.
METHODS: This investigation inclu

BACKGROUND: Klinefelter syndrome (KS) is the most common genetic cause of male infertility in humans. Mosaic KS is a subtype of KS. Due to its low prevalence, the lack of typical clinical signs, and the limited professional awareness of the syndrome, many cases of mosaic KS remain undiagnosed.
OBJECTIVE: To investigate the prevalence of rare mosaic KS, karyotype characteristics, reproductive hormone levels, and sperm quality in southern China, and to enrich our knowledge of patients with mosaic KS.
METHODS: The chromosome results of 8,110 cases of infertile male patients attending the Reprod

BACKGROUND: Isocentromere Y chromosome [idic(Y)] is a type of Y chromosome structural aberration, and patients have clinical manifestations such as Turner syndrome, unclear genitals, and male infertility.
METHODS: The patient underwent non-invasive prenatal genetic testing (NIPT), but failed three consecutive tests due to low fetal fraction (FF). Therefore, interventional amniocentesis was performed. G banding chromosome karyotyping analysis, copy number variation sequencing (CNV-seq), and fluorescence in situ hybridization (FISH) were used for prenatal diagnosis of fetal chromosomes.

OBJECTIVE: Herein, we report the first case of 46,XX/47,XXY mosaicism with a partially deleted Y chromosome.
CASE REPORT: Chorionic villus sampling (CVS; G-banding) was performed due to increased nuchal translucency; the results showed a 46,XX karyotype. However, ultrasonography revealed male-type external genitalia. The stored CVS nuclear plate was reanalyzed to investigate the cause of the fetal sex discordance. The result showed a mosaic of a small supernumerary marker chromosome (sSMC) derived from Y chromosome. The infant was delivered vaginally at 39 weeks. Peripheral blood G-banding an

STUDY QUESTION: Is there an increased risk of reproductive or colon cancer in women with Turner syndrome and their family members?
SUMMARY ANSWER: Our data suggest that there is an increased risk for sigmoid colon cancer in women with Turner syndrome and an increased prostate cancer risk in second- and third-degree male relatives.
WHAT IS KNOWN ALREADY: Turner syndrome has been associated with lower risk of breast cancer, but increased risk of gonadoblastoma and colon cancer in some, but not all studies. There is also evidence for a genetic predisposition to sex chromosome aneuploidy, which

Klinefelter syndrome (KS) is a chromosome disorder characterized by small firm testes, gynecomastia, hypogonadism, and abnormally elevated concentrations of follicle-stimulating hormone (FSH). Most KS patients show a classic 47,XXY karyotype, while about 20% have other numeric sex chromosome abnormalities, including mosaicisms. 47,XXY/46,XX mosaicism is extremely rare, and has been reported in just 10 individuals with features suggestive of KS. None of these had any spermatozoa in their ejaculate or testicular samples. We describe the first case of sperm retrieval in a 19-year-old male patient

This study was designed to identify outcome data for non-invasive prenatal testing (NIPT) results suggestive of an atypical sex chromosome abnormality of fetal/placental origin. A single-center descriptive case series was performed between January 1, 2022 and August 1, 2024, which identified 16 cases, 11 of which completed diagnostic testing. Of those 11 cases, only 2 were found to have detectable chromosomal abnormalities of the fetus (monosomy X and mosaic monosomy X). The majority of the 9 cases without detectable fetal chromosome abnormalities cannot be assessed for the presence of confine

Hereditary hypophosphatemic rickets, most commonly caused by X-linked dominant PHEX variants, leads to hypophosphatemia and bone mineralization defects. We identified a novel mosaic nonsense variant in the PHEX gene on the X chromosome by next-generation sequencing-c.1971C > A, p.(Tyr657X)--in a man with clinical features of hypophosphatemic rickets. As the variant was only found in 67% of DNA reads, we considered the possibility of sex chromosome aneuploidy (e.g. a 48,XXXY sex chromosome complement with an unaffected X chromosome i.e. variant on 2 of 3 X chromosomes producing a variant allele

OBJECTIVE: Prenatal single-nucleotide polymorphism (SNP)-based cell-free DNA (cfDNA) screening can identify genome-wide paternal uniparental disomy (GW-UPDpat), including cases with complete hydatidiform mole with a coexisting fetus (CHMCF), those with placental mesenchymal dysplasia (PMD) and those with a mosaic/chimeric GW-UPDpat syndrome. Our objective was to review laboratory data and pregnancy outcome for SNP-based cfDNA screening tests with results compatible with GW-UPDpat and a normal cell line.
METHODS: This was a retrospective study of all cfDNA screening results from a single comme

Monochorionic twins are typically monozygotic with identical fetal sex. We present a monochorionic, diamniotic twin pair of a triplet gestation with discordant fetal sex also affected with twin anemia polycythemia sequence (TAPS). The pregnancy was conceived with ovulation induction, and the initial ultrasound at 8 weeks revealed a dichorionic triamniotic triplet gestation. Discordant sex between the monochorionic pair was noted on ultrasound at 20 weeks, with Fetus 1 appearing phenotypically female and Fetus 2 appearing phenotypically male. At 26 weeks, the middle cerebral artery peak systoli
