Brain mechanisms supporting movement-evoked chronic low back pain: an observational brain imaging study.

BACKGROUND: Chronic low back pain (cLBP) is the most prevalent chronic pain condition and the leading cause of disability worldwide. Acute movement-evoked pain strongly predicts cLBP exacerbation, reflecting maladaptive amplification of nociceptive input, yet the neural mechanisms supporting movement-evoked cLBP flares remain poorly characterised. METHODS: A total of 114 participants with cLBP performed the cLBP-evoking leg raise test during perfusion-based arterial spin labelling functional MRI. Leg raise test-based pain intensity and unpleasantness ratings were assessed with an 11-point vis

Blocking ELISA Protocol for Salmonella Infection.

Salmonella is an important zoonotic pathogen that is widely distributed in livestock, poultry, and other animals. Salmonella contamination not only affects the healthy development of the livestock industry but also poses a serious threat to public health. Therefore, establishing detection methods capable of simultaneously identifying different Salmonella serotypes is crucial. Among current detection methods for Salmonella, immunological detection methods using MAbs offer high throughput, shorter processing times, and enhanced specificity, making them useful for rapid Salmonella detection. AA in Hypertension and Preeclampsia.

Human essential hypertension is driven by a network of interacting nutritional, metabolic, inflammatory, and endothelial mechanisms in which AA and other PUFAs play a critical role. Beyond excess sodium intake, population data supports the role of inadequate calcium, potassium, and magnesium intake and low antioxidant vitamin status in shaping blood pressure regulation. These nutrients influence vascular smooth muscle tone and act as cofactors that support Δ6 and Δ5 desaturase activities, thereby governing tissue availability of AA, EPA, and DHA and downstream formation of vasodilator and anti

AA in Brain-Memory, Dementia, Depression, Anxiety, Alzheimer's Disease, Affective and Psychiatric Disorders, and Coronary Heart Disease (CHD).

The human brain is rich in AA and DHA, which are crucial for normal brain growth and development and memory. They form a major structural component of neurons of the cerebral cortex and photoreceptor cells in the retina. AA and DHA support synaptic plasticity and contribute to efficient neurotransmission. Adequate amounts of AA and DHA support neuronal growth and the formation of new synapses and thus are crucial for memory and cognitive function.Depression, anxiety, schizophrenia, and other neurological diseases occur more frequently in those with coronary heart disease (CHD) and vice versa.

Multiparametric Flow Cytometry Analysis of Naïve, Memory, and Effector T-Cells.

Polychromatic flow cytometry enables the detection and characterization of markers that are helpful in defining the phenotype of various cell subsets. Here, we described a flow cytometry-based method to characterize the phenotype of naïve, memory, and effector T cells. Being able to differentiate these cells is crucial in understanding immune response and immune profiling. Naïve T cells enable the body to fight off new, unrecognized infections and diseases, and Memory T cells are enriched for response to recall antigens. Furthermore, the antigen-experienced T-cell populations can be broadly di

Enumeration of Benign and Malignant Plasmacytoid Dendritic Cells in Peripheral Blood and Bone Marrow by Flow-Cytometric Analysis.

Dendritic cells (DCs) are powerful antigen-presenting cells involved in regulating immune response. Plasmacytoid dendritic cells (pDCs) are a subtype of DCs present in small quantities in the bone marrow, peripheral blood, and lymph nodes. They are an important component of our immune system's regulation and response to diseases. They activate T cells and play a critical role in immune tolerance. In this chapter, we review the immunophenotypic features of pDCs and provide a practical protocol for pDCs enumeration in the peripheral blood and in bone marrow samples.

Neutrophil Subset Immunophenotyping.

Neutrophils, the most abundant circulating leukocyte population, are principally involved in pathogen clearance through three major mechanisms: phagocytosis, degranulation, and release of neutrophil extracellular traps. Along with these roles, neutrophils can promote or suppress tumor growth and metastatic processes. These different immunological functions reflect a great heterogeneity of maturation and activation status, as at least two main circulating populations can be identified: low-density granulocytes and normal-density neutrophils. In this chapter, we provide a detailed protocol for m

Natural Killer Cells and Innate Lymphoid Cells Immunoprofiling.

Phenotyping of natural killer (NK) cells by flow cytometry allows a detailed analysis of their functional diversity and activation status. This multiparametric approach assesses the expression of activating or inhibitory receptors and detects markers of maturation or exhaustion. It is crucial for understanding the role of NK cells in the immune response, particularly against infections and tumor cells. This phenotyping also allows us to monitor the effects of immunomodulatory treatments and to identify prognostic or predictive biomarkers in different clinical contexts.

Immunophenotyping of Human B Lymphocytes from Peripheral Blood Mononuclear Cells.

Flow cytometry has made it possible to identify human B cell subsets based on surface and intracellular antigens, a process known as immunophenotyping. CD19 and CD20 are commonly expressed in all B cells; the presence or absence of CD27 separates memory B cells from naïve cells, while IgD and IgM are used to distinguish naïve from transitional B cells. Markers such as CD38, CD27, and CD24 are helpful to identify plasmablasts and immature B cells. CD21 points to mature B cells, CD10 is on immature B cells, CD138 marks plasma cells, and CD5 is high in regulatory B cells. Memory B cells have less

Immunophenotyping Studies in Nonhuman Primate Models.

Nonhuman primate (NHP) models are important in biomedical research due to their close evolutionary relationship with humans, which provides a high degree of anatomical, genetic, and physiological similarity. This close resemblance is particularly valuable for studying complex biological processes and diseases that are difficult to model in other animals. NHPs have hematopoietic and immune systems that closely mirror those of humans, making them crucial for researching hematopoietic stem cell biology, immunology, infectious diseases, and vaccine development, offering insights that are more like

Immunophenotypic Assessment of Natural Killer Cell Degranulation and Monocyte Function from Cryopreserved Peripheral Blood Mononuclear Cells.

Natural killer (NK) cells and monocytes are central components of the innate immune system, playing critical roles in host defense, immune surveillance, and inflammation. Functional alterations in these cell populations are implicated in cancer, infectious diseases, cardiovascular disease, and responses to environmental and psychosocial stressors. Accurate measurement of NK cell and monocyte distribution and function is therefore essential for both basic and translational immunology. In large-scale or longitudinal studies, cryopreserved peripheral blood mononuclear cells (PBMCs) are commonly u

Immunophenotyping of Acute Myeloid Leukemia.

Immunophenotyping by multiparameter flow cytometry is a rapid and efficient technique to simultaneously assess and correlate multiple individual cell properties like size and internal complexity along with antigen expression in a population of cells. This method is utilized for rapid characterization of the blasts and classification of acute myeloid leukemia (AML) in both the peripheral blood (PB) and bone marrow (BM). This technique is not only useful in the initial diagnosis but also in monitoring and determining the prognosis of the disease through minimal residual disease (MRD) testing. Th

Flow Cytometric Immunophenotyping of Acute Lymphoblastic Leukemia.

Immunophenotyping by flow cytometry is an important component in the diagnostic evaluation of patients with acute lymphoblastic leukemia. This technique further permits the detection of minimal residual disease after therapy, a robust prognostic factor that may guide individualized treatment. We describe here laboratory methods for both the initial characterization of lymphoblasts at diagnosis and the detection of rare leukemic lymphoblasts after treatment. In addition to antibody combinations suitable for diagnosis and detection of minimal residual disease, we describe procedures for peripher

High Immunophenotyping of Human PBMCs Using Lyophilized Antibodies.

Immunophenotyping of human peripheral blood mononuclear cells (PBMCs) is a critical technique for understanding immune responses in both health and disease. Traditionally, this process relies on the use of liquid antibody cocktails, which are time-consuming to prepare, prone to laboratory error, and have limited shelf lives. Recent advancements in lyophilization of antibodies and other reagents have addressed these challenges by enhancing reagent stability, extending shelf life, and simplifying transport and storage. Dried antibody cocktails have been shown to perform comparably to liquid cock

High-Dimensional Immunophenotyping of Human Leukocytes by Spectral Cytometry.

Spectral cytometry has rapidly evolved over the past two decades. The development of complete spectral analysis using the flow cytometer as a spectrometer has been actualized due to advances in optics, detectors, and electronics that have made single-cell flow spectroscopy a reality. Polychromatic flow cytometry is a complex process, including the design of antibody panels and instrument compensation. Spectral cytometry has contributed significantly to effectively dealing with the complexity of polychromatic flow cytometry. It has substantial potential to improve primary and clinical research.

Immunophenotyping of Human Regulatory T-Cells from Peripheral Blood Mononuclear Cells.

Regulatory T cells, or Tregs, are a unique type of CD4⁺ T cell that act as the immune system's brakes-they stop it from turning against the body's own tissues. For about twenty years now, researchers have been digging into what sets Tregs apart from other T cells, both in their composition and function (1). Markers like CD25 (IL-2Rα), Foxp3, and CTLA-4 usually show up when Tregs are active, but these markers aren't always reliable-they can shift depending on where the cells are and what they are doing. Foxp3 is a transcription factor that Tregs need to develop and function. Mutations on Foxp3

Global, regional, and national burden of exposure to chewing tobacco, 1990-2023: a systematic analysis of the Global Burden of Disease Study 2023.

BACKGROUND: Exposure to chewing tobacco is a known health risk factor but has received less attention than smoking in global tobacco control efforts. As the evidence base linking chewing tobacco use to poor health grows, it is essential to closely track the changing use of chewing tobacco distinct from other forms of smokeless tobacco, as well as the estimated burden associated with its use. These data are essential to inform policy makers who seek to identify and avert the health burden associated with these dangerous products. METHODS: We estimated the prevalence of chewing tobacco use and
