Alveolar echinococcosis drives functional reprogramming of hepatic CD8+ T cells.

BACKGROUND: Alveolar echinococcosis (AE), caused by the larval stage of Echinococcus multilocularis, exhibits infiltrative, tumor-like behavior in the liver and persists within its tolerogenic immune environment. Although T cells are central to host defense, the stage-specific remodeling of their lineage states during AE remains unclear. METHODS: A secondary AE infection model was established by portal vein injection of approximately 1,000 viable protoscoleces in C57BL/6 mice. Liver tissues collected at 3 days (3 dpi) and 3 months (3 mpi) post-infection were analyzed using single-cell RNA seq

Prevalence and molecular genotyping of cystic echinococcosis among Buffalo isolates from South-Punjab, Pakistan.

Echinococcus granulosus sensu lato (s.l.), the causative agent of cystic echinococcosis (CE) cause enormous health, production, and economic impacts on livestock and human population. Buffaloes play a central role in Pakistan's livestock economy, and often serve as key intermediate hosts in the transmission of E. granulosus s.l. However, little is known about CE in buffaloes and associated productive and financial risks in southern Punjab, Pakistan. Out of 1689 buffaloes (male-678, female-1011), 256 were found infected (95%CI = 13.45-16.87), with a 15.16% prevalence (X2 = 30.30). Samples prima

Recent advances in clinical and laboratory diagnosis of hydatid cyst: from imaging to recombinant and nanobiosensor-based approaches.

BACKGROUND: Cystic echinococcosis is a significant parasitic disease that predominantly affects the liver and lungs in humans. Its diagnosis relies mainly on imaging techniques such as ultrasound and computed tomography, supported byserological tests, including enzyme-linked immunosorbent assay and immunoblotting, which are commonly used inhuman serodiagnosis. Conventional serological assays frequently show variable sensitivity, particularly in pulmonary, inactive, or calcified cysts, and may yield false-negative or cross-reactive results. METHODS: Recent advances in recombinant antigen-based

Small particles of Echinococcus granulosus (spegs) and Echinococcus multilocularis (spems) promote follicular T helper cell expansion and are associated with IgE and IgG4 class switching in human lymph nodes.

BACKGROUND: Cystic echinococcosis (CE) and alveolar echinococcosis (AE) are zoonotic diseases caused by the larval stages of Echinococcus granulosus and E. multilocularis. Prior studies have identified small particles from the outer layer of parasitic lesions termed spegs (from E. granulosus) and spems (from E. multilocularis), which appear to interact with the human immune system, leading to lymph node enlargement. METHODS: We analyzed lymph nodes from CE (n = 3) and AE (n = 3) patients by immunohistochemical staining for spegs/spems and CD57. Automated image analysis revealed a significantl

From conventional to next-generation diagnostics in cystic echinococcosis: integrating serology, ultrasonography, and molecular tools: a review.

This review provides the relative roles of serology and ultrasonography in the diagnosis of cystic echinococcosis (CE), emphasizing their performance, limitations, and integration within current and emerging diagnostic strategies. CE is a chronic zoonotic disease caused by Echinococcus granulosus, predominantly affecting the liver and lungs in endemic livestock-raising regions. It is often a silent clinical course, variable clinical presentation, and potential for severe complications that contribute to underdiagnosis and underreporting. Ultrasonography is the first-line tool for hepatic CE, e

Hydatid disease of the ribs: a diagnostic challenge with an endless differential diagnosis - a case report.

Hydatid disease is a parasitic infection in humans caused by Echinococcusgranulosus and can lead to tissue cyst formation anywhere in the body. The most common sites for the development of parasitic cysts are the liver (75%) and lungs (15%). Skeletal involvement in echinococcal infection is relatively rare, occurring in only 1%-4% of cases. Few reports in the available medical literature describe rib hydatidosis, which clinically mimics benign or malignant cystic tumors.

Hepatic alveolar echinococcosis mimicking cholangiocarcinoma: a case study.

BACKGROUND: Hepatic alveolar echinococcosis (HAE) is associated with significant disability and mortality. After infection, there is a prolonged asymptomatic latent period, and most patients present at a late stage when they seek medical treatment. The radiological features of HAE can resemble those of liver malignancies, posing diagnostic challenges. A 52-year-old male was admitted to our hospital with discomfort and pain in the upper right abdomen persisting for more than six months without an apparent trigger. Abdominal ultrasound revealed a heterogeneous mass in the right posterior lobe of

The critical window for host defense: macrophage trogocytosis eliminates Echinococcus multilocularis at the early establishment stage.

INTRODUCTION: The ability of Echinococcus multilocularis (E.m) to establish infection depends on evading host immune clearance during the early stages. This study investigated the mechanisms by which host macrophages eliminate E.m during this critical window. METHODS: We utilized a mouse infection model with fluorescently labeled protoscoleces to observe early immune dynamics in vivo. Histopathological analysis was performed to characterize lesion phenotypes. The mechanism of parasite killing was further explored using in vitro co-culture experiments and scanning electron microscopy (SEM). R

Systemic remodeling of the multi-organ virome following Echinococcus infection in mice.

The interaction between parasitic infection and the host virome represents a frontier issue in microbial ecology, yet how Echinococcus infection affects the multi-organ virome and whether these alterations hold diagnostic or interventional potential remains poorly understood. In this study, we performed viral metagenomic sequencing on gut, liver, and lung samples from both infected and uninfected mice, integrating community structure clustering, diversity indices, and differential analyses, including STAMP and LEfSe. Our results reveal that Echinococcus infection induced significant tissue-spe
