Variations in the management of canine osteoarthritis: a cross-sectional survey of veterinary practices.

INTRODUCTION: There is limited data on how veterinarians implement multimodal therapies for managing canine osteoarthritis (OA). The primary aim of this study was to describe how OA is managed by veterinarians with diverse backgrounds. The secondary aim was to report perceived side effects associated with bedinvetmab (LibrelaTM) and grapiprant. The authors hypothesized that variation exists in management patterns among veterinarians with different training backgrounds and experience levels. METHODS: A cross-sectional, observational, voluntary response survey was distributed to veterinarians a

Clinical features, diagnosis and prognosis of nivolumab-associated fulminant type 1 diabetes.

Fulminant type 1 diabetes (FT1D) is a rare immune-related adverse event associated with nivolumab, and its clinical characteristics remain unclear. This study aims to characterize the clinical features of nivolumab-associated FT1D and to generate evidence to inform its diagnosis and prevention. A systematic search of databases was conducted to identify clinical cases of nivolumab-associated FT1D reported through March 31, 2026. Clinical data for the patients were extracted and subjected to statistical analysis. A total of 59 patients were included, with a median age of 59 years (range 22, 87).

Species, cell proliferation state and oxygen partial pressure are major factors in the in vitro toxicity of repurposed potential ADPKD drugs in human and murine renal proximal tubule cells.

Unexpected nephrotoxicity during clinical stages of drug development, due to undetected toxicity in in vitro and in vivo settings, can lead to drug attrition and jeopardize drug development. The latter highlights the importance of in vitro renal toxicity testing that can safely be translated to humans. Two factors commonly overlooked in the latter are the continuous cell proliferation that occurs in regeneration processes after renal insult and the physiological O2 tension in the kidney. Autosomal Dominant Polycystic Kidney Disease is a genetic disorder characterized by the development of rena

Glucagon-stimulated copeptin response in children: a proof-of-concept study.

PURPOSE: The diagnostic workup of polyuria-polydipsia syndrome (PPS) is not well-established in children and adolescents, and several non-osmotic copeptin-stimulating agents have been investigated. Glucagon stimulation test (GST) induced a robust copeptin response in adults, but data in children are lacking. This study aimed to investigate copeptin response to GST in children tested for suspected growth hormone (GH) deficiency. METHODS: Twenty children (9 girls, age 10.3 ± 2.9 years) with no PPS underwent GST (30 µg/kg of glucagon i.m.). Plasma glucose, GH, cortisol, and copeptin were measure

The Missense Mutant ClC-5 (E211A), associated with Proton/Chloride Uncoupling Exacerbates Renal Pathology Compared to the ClC-5 null Mutant.

BACKGROUND: Dent disease type 1 is an X-linked proximal tubulopathy caused by mutations in CLCN5, which encodes the chloride/proton exchanger ClC-5. While loss-of-function mutations impair endocytosis and lead to progressive renal disease, the pathological consequences of uncoupling mutations that selectively disrupt Cl-/H+ exchange remain incompletely understood. METHODS: In this study, we examined the renal phenotype associated with the ClC-5 E211A uncoupling mutation, which abolishes proton transport while preserving chloride conductance, and compared it with that of ClC-5 null mice. Postoperative polyuria after hepatectomy associated with prolonged sodium-glucose co-transporter-2 inhibitor effect.

Sodium-glucose co-transporter-2 inhibitors are routinely withheld before surgery to reduce the risk of peri-operative metabolic complications. However, their pharmacodynamic effects may persist beyond the recommended discontinuation period. We report a 67-year-old man with type 2 diabetes who developed severe postoperative polyuria (6-7 l.day-1) with persistent glycosuria after partial hepatectomy despite discontinuation of canagliflozin 3 days before surgery. Marked transient hepatic dysfunction occurred immediately after surgery, while serum sodium remained within the normal range. Glycosuri

HNF1A-Maturity-Onset Diabetes of the Young (MODY) in a Family With a Rare Silent Variant: Clinical Challenges and Variable Expressivity.

Maturity-onset diabetes of the young (MODY) is a monogenic form of diabetes characterized by autosomal dominant inheritance, typically occurring during adolescence or early adulthood. Mutations in the hepatocyte nuclear factor 1-α (HNF1A) gene are among the most common causes of MODY and are frequently misdiagnosed as type 1 (T1D) or type 2 diabetes (T2D), leading to inappropriate management. We report the case of a preschool-aged boy who presented with polyuria and polydipsia, fasting hyperglycemia, negative pancreatic autoantibodies, and an elevated C-peptide level. A multi-generational fami

Youth with type 2-diabetes: clinical findings at onset and impact of continuous glucose monitoring (CGM) on glycemic control and disease management.

OBJECTIVES: The global obesity pandemic is associated with a higher prevalence of youth-onset type 2 diabetes (T2D). Continuous glucose monitoring (CGM) is established in type 1 diabetes, however, evidence regarding its impact in pediatric T2D is limited. This study has analyzed (a) clinical and metabolic characteristics in adolescents at T2D onset and (b) the impact of CGM on glycemic control, patient satisfaction and disease management in youths with T2D. METHODS: a) Clinical and biochemical parameters at diagnosis were assessed in adolescents with T2D. b) The impact of 8 weeks of CGM on Hb

Yellow Oleander (Thevetia peruviana) Toxicity from a Misrepresented Dietary Supplement: A Case Report.

INTRODUCTION: The "lucky nut" is the seed of the yellow oleander plant, often sold as a medicinal supplement in unregulated markets and known to contain cardiac glycosides, which may cause life-threatening bradycardias and Digoxin-like toxicity upon ingestion. Diagnosis is typically clinical, with treatment including Digoxin immune Fab with supportive intensive cardiac care. CASE REPORT: A 70-year-old male presented to the emergency department after ingesting one-fourth of a Peruvian nut he found at a local market to reduce his nocturnal polyuria. The patient brought a part of the Peruvian nu

An illustrative summary of receptor-mediated mechanisms involved in the therapeutic regulation of type 2 diabetes.

Type 2 diabetes (T2D) is a chronic metabolic disorder primarily affecting the metabolism of glucose in the body. It is characterized by polyphagia, polydipsia, polyuria, fatigue, and persistent hyperglycemia. It occurs mainly because of two major factors, namely, dysregulated insulin secretion from pancreatic β-cells and/or reduced sensitivity of insulin-responsive tissues to insulin, i.e., insulin resistance. The process of insulin secretion and insulin signaling is complex, requiring the involvement of distinct membrane receptors. Sulfonylureas are a class of antidiabetic drugs which exert t
