The toxicology of orally administered Δ9-tetrahydrocannabinol in sheep.

INTRODUCTION: As part of a larger study to assess the potential value of industrial hemp biomass (the low Δ9-tetrahydrocannabinol (Δ9-THC) variety of Cannabis sativa L.) as a feed for ruminants, a pharmacokinetics study of Δ9-THC, one of the main cannabinoids of interest, was undertaken. In undertaking this study, the toxicology of orally administered Δ9-THC was incidentally investigated and was discussed in the current paper. METHODS: Eight Merino ewes were administered with two per os (PO) doses of 88.5 mg Δ9-THC/kg bodyweight (BW) 12 h apart. Blood samples were taken periodically to determ

Nausea and vomiting as adverse events of oliceridine: a systematic review and meta-analysis of randomized controlled trials.

BACKGROUND: Postoperative nausea and vomiting (PONV) continue to be some of the most common and troublesome complications following anesthesia. Oliceridine, a G protein-biased µ-opioid receptor agonist, has the potential to provide effective pain relief while reducing the incidence of opioid-associated side effects. OBJECTIVES: The aim of this study is to evaluate the effectiveness and safety of oliceridine for preventing PONV compared with morphine and a placebo. METHODS: A comprehensive search was performed across PubMed/MEDLINE, Embase (Ovid), and the Cochrane Central Register of Controll

Gabapentin-Induced Hypoglycemia in a Patient With Pre-Diabetes: A Case Report and Review of the Literature.

Gabapentin, a γ-aminobutyric acid (GABA) analogue, is widely prescribed for neuropathic pain and partial seizures. While common adverse effects include somnolence and dizziness, hypoglycemia is rarely reported. A 76-year-old woman with hypertension, dyslipidemia, hypothyroidism, and pre-diabetes (HbA1c 6%) developed recurrent symptomatic hypoglycemia shortly after starting gabapentin 300 mg once daily for neuropathic pain. Within two days, she experienced dizziness, sweating, and fatigue with capillary glucose readings as low as 2.4 mmol/L, confirmed on laboratory chemistry. Investigations rev

Effect of Concomitant Medications on Adverse Event Frequency, Severity, and Time to Resolution in a Cenobamate Open-Label Trial.

BACKGROUND AND PURPOSE: Polytherapy in epilepsy is associated with a higher risk of adverse events, poorer adherence, and impaired quality of life. Thus, reducing concomitant antiseizure medications (ASMs) and/or using a newly added ASM such as cenobamate with only 1 other ASM (i.e., bitherapy) may improve tolerability. This post hoc analysis evaluated the impact of the number and type of concomitant ASMs on treatment-emergent adverse events (TEAEs) that occurred in the cenobamate C021 open-label clinical trial. METHODS: Adjunctive cenobamate was initiated in adults (aged 18-70 years) with un

Mixed Presentation of Heroin-Associated Spongiform Leukoencephalopathy and CHANTER Syndrome After Fentanyl Use: A Case Report.

BACKGROUND: Toxic leukoencephalopathy is a heterogenous syndrome characterized by destruction of brain white matter. Inhalational heroin has been linked to a specific set of neuroimaging findings termed heroin-associated spongiform leukoencephalopathy (HSLE). Clinical findings in HSLE generally develop and progress through an initial, intermediate, and terminal stage. Fentanyl has been linked to a separate set of neuroimaging findings and neurological symptoms labeled Cerebellar Hippocampal And Basal Ganglia Transient Edema with Restriction (CHANTER) syndrome, in which patients often present w

Flunarizine and cinnarizine in the prophylaxis of vestibular migraine and migraine-associated menière's disease: An 8-month retrospective study.

BACKGROUND: Vestibular migraine (VM) and migraine-associated Menière's disease (MAMD) are common causes of recurrent vertigo, often resulting in substantial disability and impaired quality of life. Preventive therapy is challenging, and calcium channel antagonists such as flunarizine and cinnarizine have been proposed as effective options, though comparative evidence remains limited. METHODS: This was a retrospective cohort study including 402 consecutive patients diagnosed with VM (n = 298) or MAMD (n = 104) at two vestibular specialized centers. Patients received either flunarizine (10 mg/d

Vitrectomy Status Alters Early Intravitreal Drug Distribution: An Ex Vivo PET-CT Study.

IMPORTANCE: A significant need exists for new antipsychotic medications with different mechanisms of action, greater efficacy, and better tolerability than existing agents. Xanomeline is a dual M1/M4 preferring muscarinic receptor agonist with no direct D2 dopamine receptor blocking activity. KarXT combines xanomeline with the peripheral muscarinic receptor antagonist trospium chloride with the goal of reducing adverse events due to xanomeline-related peripheral muscarinic receptor activation. In prior trials, xanomeline-trospium chloride was effective in reducing symptoms of psychosis and gen

Efficacy and safety of tradipitant in motion sickness: a systematic review and meta-analysis.

BACKGROUND: Motion sickness affects up to 30% of the general population, yet the three pharmacological agents currently approved in the United States, dimenhydrinate, meclizine, and scopolamine carry significant sedative and cognitive liabilities and offer incomplete protection against vomiting. Tradipitant, a selective neurokinin-1 receptor antagonist, has been evaluated across three randomized controlled trials for motion sickness prevention, but no pooled quantitative synthesis of this evidence exists. This study aimed to evaluate the efficacy and safety of tradipitant versus placebo in adu

An inherited SLC25A12-related recessive form of congenital porencephaly in Limousin cattle.

BACKGROUND: Congenital abnormalities in cattle, which include lesions in the central nervous system, usually occur sporadically. Porencephaly, a condition characterised by the presence of cystic fluid-filled cavities within brain tissue, is often virus-induced, but rare inherited forms have been identified in other species. Thirty-four calves affected by porencephaly were reported in Limousin cattle in Great Britain and 16 underwent clinicopathological investigation. We aimed to: (1) characterize the disorder phenotype, (2) investigate its possible genetic cause, and (3) determine the frequenc

Timing and Causes of Systemic Readmissions After Hip Fracture Surgery: An 8-Year Retrospective Cohort Study.

BACKGROUND: Systemic readmissions after hip fracture surgery impose a substantial clinical and economic burden. Hip fracture represents a sentinel orthopedic injury with prolonged postoperative vulnerability extending beyond the index admission. Evidence from the Middle East remains scarce, particularly regarding the incidence, causes, and timing of systemic readmissions in multiethnic populations. This study aimed to determine the one-year incidence, etiologies, and temporal distribution of systemic readmissions following proximal hip fracture surgery, and to explore whether commonly document
