Psilocybin-containing mushroom-assisted psychotherapy is associated with sustained psychological and somatic improvements: two case studies from Jamaica.

We report two case studies that examine the therapeutic potential of structured psilocybin-containing mushroom biomass regimens administered within a clinical framework, evaluating their impact on mental health outcomes as well as associated somatic symptoms. Two patients underwent the De La Haye Psilocybin Treatment Protocol (DPTP), which integrates micro- and high-doses (combined psilocybin and psilocin content of 1-2 mg versus 40-60 mg) with psychohistoriographic psychotherapy. Follow-up included an integration session and subsequent office visits, including the completion of the Revised My

Congenital anosmia.

INTRODUCTION: Congenital anosmia (CA) is a rare condition among all olfactory disorders (OD). It is estimated that one in 10,000 persons is born with anosmia, and the diagnosis may be related to various syndromes, especially Kallmann (hypogonadotropic hypogonadism). METHODS: Patients with CA were identified in the REDCap database at the University Clinic for Flavour, Balance, and Sleep; Department of Otorhinolaryngology, Goedstrup Hospital, Denmark. In addition to demographics and clinical findings, the database recorded patient-reported outcome measures, such as the Sino-Nasal-Outcome-Test 2

Genome-wide machine learning analysis of anosmia and ageusia with COVID-19.

The COVID-19 pandemic has caused substantial worldwide disruptions in health, economy, and society, manifesting symptoms such as loss of smell (anosmia) and loss of taste (ageusia), that can result in prolonged sensory impairment. Establishing the host genetic etiology of anosmia and ageusia in COVID-19 will aid in the overall understanding of the sensorineural aspect of the disease and contribute to possible treatments or cures. By using human genome sequencing data from the University of Iowa (UI) COVID-19 cohort (N = 187) and the National Institute of Health All of Us (AoU) Research Program

Patient Perspectives on a Hypothetical Olfactory Implant: A French Survey.

INTRODUCTION: Smell loss severely impacts quality of life, and current treatment options remain limited. Emerging concepts such as olfactory implants, combining an external electronic nose and an intracranial neurostimulator, raise important questions regarding patient perceptions, expectations, and acceptability. METHODS: The French patient organization <ext-link ext-link-type="uri" xlink:href="https://Anosmie.org" xmlns:xlink="http://www.w3.org/1999/xlink">Anosmie.org</ext-link> conducted an online survey among members experiencing smell loss. Long-Term Olfactory Dysfunction in COVID-19 Patients: A Systematic Review.

Olfactory dysfunction (OD) emerged early in the COVID-19 pandemic as a prevalent and often persistent symptom. While most individuals recover within weeks, a significant proportion continue to suffer from long-term impairments, including both quantitative and qualitative sensory deficits. Our review aimed to summarize current evidence on long-term post-COVID-19 OD with a duration of at least three months, including prevalence, recovery trajectory, and prognostic factors. The PubMed and Scopus databases were searched for relevant studies up to August 2024 following Preferred Reporting Items for

SARS-CoV-2 disrupts the integrity of a human blood-brain barrier model in the absence of infection.

Neurological symptoms are recognized in patients with COVID-19, and include anosmia, ageusia, cognitive impairments as well as more severe complications including encephalitis and ischemic strokes. Furthermore, persistent cognitive impairment including 'brain fog' is recognised as part of the long COVID syndrome. While SARS-CoV-2 has been shown to infect cells of the central nervous system (CNS) in vitro and in vivo, it is unclear whether direct infection of the CNS or indirect mechanisms including activation of the coagulation cascade or immune activation are the key drivers of COVID-19 neuro

Partial Recovery of Anosmia Since Childhood With Acquired Parosmia: Mechanistic Insights and Therapeutic Implications-A Case Report.

BACKGROUND: Isolated congenital anosmia (ICA) traditionally represents an irreversible sensory disorder affecting 1 in 5,000-10,000 individuals. This case demonstrates rare partial olfactory recovery in childhood-onset anosmia following corticosteroid therapy, providing novel insights into olfactory neuroplasticity and therapeutic potential. CASE PRESENTATION: A 32-year-old woman with lifelong anosmia, diagnosed with ICA at age 20 using Sniffin' Sticks testing (2013), experienced first olfactory perceptions in 2014 following one week of oral corticosteroids and nine months of topical mometaso

Limited observed efficacy of olfactory training in severe long-term post-COVID-19 syndrome: a prospective cohort study.

PURPOSE: Persistent chemosensory dysfunction is a debilitating symptom of Post-COVID-19 Syndrome (PCS). While olfactory training (OT) is effective in early post-viral smell loss, its efficacy in chronic, multisystemic PCS remains unclear. This study aimed to characterize a severely affected cohort and to evaluate changes in olfactory function during a structured OT intervention. METHODS: In this prospective study, 29 patients with persistent PCS lasting > 12 weeks (mean duration approximately 30 months) were recruited from a specialized post-COVID outpatient clinic. All patients underwent str

Olfactory Threshold Can Be Reliably Tested With Sniffin' Sticks in Non-Clinical Settings: Associations Shown in a Diverse, General Population.

OBJECTIVE: The olfactory threshold part of the Sniffin' Sticks test is widely used to assess chemosensory function, yet most studies are conducted in controlled laboratory or clinical environments with selective samples. This study examined odour threshold performance with the Sniffin' Sticks threshold test in a large, heterogenous sample, testing whether established associations generalise outside the lab. METHOD: A total of 798 museum visitors aged 18-88 years completed the threshold test and rated their olfactory and gustatory ability, nasal patency, and sensitivity. Participants were clas

Genetic context unmasks olfactory loss as a marker for dyskinesia risk via levodopa exposure trajectories in Parkinson's disease.

BACKGROUND: Olfactory impairment is common in Parkinson's disease (PD), but its prognostic value for levodopa-induced dyskinesia (LID) remains uncertain. We examined whether genetic background modifies the olfaction-LID association and how levodopa exposure influences this relationship. METHODS: We analyzed longitudinal PPMI data from 1254 participants with early PD (921 sporadic; 333 genetic carriers, including GBA, LRRK2, and other variants). Baseline olfaction was classified as normosmia, hyposmia, or anosmia using the University of Pennsylvania Smell Identification Test. The outcome was t

Multifaceted signaling of the cell surface receptor neuropilin-1 (NRP1): pathophysiological roles and therapeutic potential.

Neuropilin-1 (NRP1) is a developmentally conserved cell surface receptor that functionally acts to bind various ligands like class 3 semaphorins and several members of the VEGF family to engage in different signaling pathways that control cell migration and motility. Due to its widespread expression, NRP1 is implicated in immune function, axonal guidance, viral entry, tumor progression and angiogenesis. NRP1 is dysregulated across multiple cancer types and promotes tumor progression in a context-dependent manner in tumor cells and the tumor microenvironment, including endothelial, stromal, and

Navigating the efficacy, safety and outcome of intranasal platelet-rich plasma for persistent olfactory loss: a systematic review and meta-analysis.

BACKGROUND: Persistent olfactory dysfunction (OD) poses a significant clinical challenge with limited therapeutic options. Intranasal platelet-rich plasma (PRP), an autologous source of regenerative growth factors, presents a biologically plausible intervention, yet its efficacy across various OD etiologies remains synthetically underexplored. METHODS: This systematic review and meta-analysis adhered to PRISMA guidelines. A comprehensive search of PubMed/MEDLINE, Web of Science, and Cochrane Library (2010-2025) identified studies assessing intranasal PRP for persistent OD (> 6 months). Random
