Protein-truncating variants in APBA1 in minors with severe, early-onset obesity: implications for genetic testing.

BACKGROUND: Severe, early-onset obesity is a monogenic disorder in a subset of patients. Recently, rare protein-truncating variants (PTVs) in BSN and APBA1 have been implicated as monogenic causes of obesity. Truncating variants in these two genes have been associated with adult-onset monogenic obesity. Here, we aimed to investigate the occurrence of rare PTVs in APBA1 and BSN in minors with severe obesity manifesting in early childhood or adolescence. METHODS: Previously obtained exome data of 209 children and adolescents (0–18 years) with severe obesity manifesting in early childhood or adol

Auditory Processing Disorder in childhood: a critical appraisal of diagnostic validity, functional assessment, and interdisciplinary practice.

Auditory Processing Disorder (APD) refers to listening difficulties in individuals with normal hearing thresholds but impaired processing of auditory information. Despite decades of research, its diagnostic validity in childhood remains highly debated. This narrative review critically examines the conceptual foundations, methodological limitations, and clinical implications of APD, with the goal of promoting more functional and interdisciplinary approaches to auditory assessment. National and international literature, including professional guidelines, systematic reviews, and observational stu

Proprioception and Sensorimotor Regulation Across the Day-Night Cycle in Developmental Dyslexia: Toward an Embodied Perspective.

Background: Sensorimotor differences have frequently been reported in children with developmental dyslexia, but are often considered secondary or comorbid to phonological deficits. Within an embodied cognition perspective, reading acquisition emerges from dynamic interactions between bodily regulation, multisensory integration, and learning-related neural plasticity. Proprioception contributes to spatial orientation, motor coordination, and perceptual stabilization, while sleep-dependent processes play a critical role in the consolidation and automatization of cognitive and motor skills. Defining dyslexia: 2025 revision.

In October 2025, the Board of Directors of the International Dyslexia Association approved a revised definition of dyslexia. This paper introduces this 2025 definition, outlines the iterative and data-driven process used in its development, and clarifies the intended purposes and boundaries of the definition for different stakeholders. The paper also reviews the current scientific evidence relevant to each component of the definition and provides the rationale for revisions to the 2002 IDA definition. In addition, it offers a set of high-level, practice-oriented guidelines intended to support

Differential profiles of reading development in neurodevelopmental disorders under an adapted tier 2 response to Intervention protocol.

INTRODUCTION: Reading proficiency is a foundational skill. Failure to achieve reading competence constitutes a global educational and public health challenge. This burden is especially high among children with neurodevelopmental conditions. Despite the availability of evidence-based frameworks, such as Response to Intervention (RTI), a lack of scalable, context-sensitive models remains for supporting struggling readers in low-resource environments. Building on this context, this study describes the Less Intensive Response to Intervention Tier 2 (LIRTI2). We hypothesize that the LIRTI2 will imp

Combined accelerated theta burst stimulation and reading instruction for the treatment of persistent developmental dyslexia: methodology and preliminary findings.

Developmental dyslexia (DD) is a neurodevelopmental disorder that results in poor reading proficiency due to atypical neurobiological attributes. Despite many opportunities to learn, the most treatment resistant cases of DD can still have severe reading problems that persist into adulthood. The current manuscript presents methodology and preliminary results for a study which aims to assess the impact of a combined accelerated theta burst stimulation (aTBS) and individualized reading instruction paradigm to improve reading outcomes in young adults with persistent DD. This multiple baseline, sin

Associations between dyslexia and mental health in primary school students: the roles of parenting style, and character strengths.

OBJECTIVE: Dyslexia, a neurodevelopmental disorder that impairs reading, poses risks to students' academic and psychological well-being. This study examined the associations between dyslexia and mental health outcomes (anxiety and depression) in primary school students, focusing on the moderating roles of parenting style, and character strengths. METHOD: A cross-sectional survey was conducted among 2,322 primary school students in Yinchuan city, China. Dyslexia risk was identified through multi-step screening. Binary logistic regression was used to test main effects and interaction effects (d

Development and validation of red flags for early detection of dyslexia in Sinhala (REaDDS tool) for dyslexia screening.

Sinhala is an alpha-syllabic language. However, majority of dyslexia screening tools are developed in English. Despite growing evidence on language skills associated with dyslexia, a linguistically and culturally appropriate standardized tool is not available for Sinhala-speaking children. This study aimed to develop and validate a screening instrument to identify early signs of dyslexia, focusing on language-based skills in children aged 4-6 years. The Red Flags for Early Detection of Dyslexia in Sinhala (REaDDS) tool was developed through two phases. Study 01 encompassed tool development, De

Deficits in orthographic-phonological automatic integration for chinese adults with reading fluency difficulty: Evidence from ERP.

This study used Event-Related Potential (ERP) technology to investigate the neural mechanisms underlying orthographic-phonological automatic integration in Chinese adults with Reading Fluency Difficulty (RFD). Twenty-two fluent Chinese readers (mean age = 22.41, 9 males) and 19 individuals with RFD (mean age = 21.37, 5 males) participated in this study. The Mismatch Negativity (MMN) component, elicited through a cross-channel passive oddball paradigm, served as a neural index to examine orthographic-phonological automatic integration. The study found that adults with RFD in Chinese exhibited s

A discovery and replication study of dyslexia does not reveal reproducible gray matter volume differences.

Two decades of research have provided evidence for gray matter volume (GMV) differences in developmental dyslexia (or reading disability, RD) in the left perisylvian cortex. However, there are concerns about result inconsistencies, likely attributable to small sample sizes, lenient statistical thresholds, and insufficient accounting for demographic variables and global GMV (Ramus et al., 2018). To address these concerns, we conducted a Discovery and Replication Study (N=262) using data from the Adolescent Brain Cognitive Development Study. We found GMV differences between the RD and Control Gr

Accommodation or Adaptation? How L2 Reading Proficiency Affects Reading Strategies in EFL Learners with and without Dyslexia.

This study investigates how reading proficiency in an orthographically opaque foreign language (L2 English) modulates word recognition and decoding strategies of Italian adolescents with and without developmental dyslexia (DD), whose native language orthography is highly transparent. The size of the visual and phonological units that are processed while reading is modulated by the orthographic depth of the language being read. In the case of early biliterate readers, reading strategies are characterized by cross-linguistic transfer. It is an open question whether this is also the case for late

Development and validation of the dyslexia assessment and intervention scale to measure dyslexia knowledge.

Practitioner knowledge about dyslexia is related to improved educational practices, but few measures of dyslexia knowledge have been validated with rigorous processes. The current study developed the Dyslexia Assessment and Intervention Scale (DAIS) with the three-phase process outlined by Boateng et al. (Frontiers in Public Health 6:149, 2018) to assess the knowledge of participants of a university-based certificate program. The three-phased development process resulted in a 21-item survey that adequately represented standards. The items were tested with 221 respondents and resulted in accept

Morphological knowledge and spelling in French-speaking adults with developmental dyslexia.

Spelling errors are among the most persistent symptoms of dyslexia in adults. This study investigated the influence of morphological information on spelling accuracy in French-speaking adults with developmental dyslexia. Specifically, we aimed to (1) compare university-level adults with dyslexia and age-matched controls in spelling morphologically complex words, and (2) examine the role of morphological awareness in spelling among individuals with dyslexia. In total, 53 adults with dyslexia and 55 typical adult readers completed a spelling task with five types of words: words with silent final

Different minds, shared struggles: Personality and self-beliefs across neurodivergent conditions.

Neurodivergent individuals differ from their neurotypical peers in cognitive, emotional, and behavioral characteristics. However, research has yet to provide a systematic comparison of personality and self-beliefs across multiple formally diagnosed and self-identified neurodivergent conditions. This study examines whether self-identified neurodivergence explains additional variance beyond formal diagnosis in the Big Five personality dimensions, positive as well as negative self-beliefs across seven conditions (autism, ADHD, ADD, DCD, dyslexia, dyscalculia, and giftedness). Using two waves of d

Uncovering subgroup heterogeneity in dyslexia intervention outcomes using explainable machine learning.

Identifying heterogeneity within literacy intervention outcomes can inform more targeted strategies for dyslexia remediation. Based on prior work that used machine learning to predict literacy intervention responders and non-responders at baseline, this study uncovered 2 subgroups of responders and 4 subgroups of non-responders based on feature importance profiles of 132 students who were diagnosed with dyslexia. Their average age was 8.2 years at baseline (98.08 months). They received literacy intervention for an average of 33.62 months. Intervention subgroup membership predicted long-term li
