Associations between brain and blood metabolites with gastrointestinal discomfort in schizophrenia and healthy controls.

INTRODUCTION: Previous studies have reported inconsistent findings regarding the severity of gastrointestinal (GI) symptoms in schizophrenia. While abnormalities in neurometabolic pathways have been implicated in both schizophrenia and GI disorders, little is known about the biological underpinnings of GI discomfort in schizophrenia. This study aimed to investigate the relationships between brain metabolite concentrations, blood biomarkers, and self-reported GI symptom severity in patients with schizophrenia and healthy controls. METHODS: Thirty-six patients with schizophrenia in remission (S

Biallelic variants in the noncoding RNA gene RNU4-2 cause a recessive neurodevelopmental syndrome with distinct white matter changes.

Genetic variants in RNU4-2, which is transcribed into the U4 small nuclear RNA component of the major spliceosome, were recently shown to cause ReNU syndrome, a prevalent dominant neurodevelopmental disorder (NDD). These variants almost exclusively arise de novo and cluster within 18 nucleotides of RNU4-2. Here we describe a new recessive NDD associated with homozygous and compound heterozygous variants in RNU4-2. We identify 38 individuals with biallelic variants outside the 18-nucleotide ReNU syndrome region that cluster within other functionally important elements of U4: Stem II, the k-turn

Altered structural connectivity in white matter lesion-induced mild cognitive impairment: a probabilistic fiber tracking and graph theory analysis.

White matter lesions (WMLs) are a common cause of vascular cognitive impairment, yet the nature of whole-brain structural connectivity alterations in WML-related mild cognitive impairment (MCI) remains incompletely understood. In this study, an integrated approach combining whole-brain probabilistic fiber tracking with graph theory analysis was employed to investigate structural network changes in 33 patients with WML-related MCI and 37 healthy controls. The results revealed that, at the MCI stage, the brain's structural network exhibited not only reduced connectivity in specific neural circui

Relationships between sequential MRI in the neonatal period and early motor repertoire in very preterm infants.

BACKGROUND: Infants born very preterm frequently experience neurodevelopmental challenges. Early detection enables provision of targeted interventions to improve outcomes. Early neonatal and term-equivalent age (TEA) MRI and 3-5-month corrected age (CA) Prechtl Motor Optimality Score - revised (MOS-R) are useful prognostic tools, however structure-function relationships between them are not well established. METHODS: Infants born <31 weeks' gestation underwent MRI at 32-weeks postmenstrual age and TEA. Modified Kidokoro MRI scoring generated white matter, cortical grey matter, deep grey matte

White matter microstructure differences between 15q11.2 copy number variation carriers and non-carriers in mid-to-late life.

The 15q11.2 BP1-BP2 copy number variant (CNV) has been associated with neurodevelopmental and psychiatric conditions and brain grey matter structure, but its effects on white matter microstructure (WMM) in mid-to-late adulthood to assess long-term neurobiological effects remain unclear. Understanding these effects is important for evaluating long-term neurobiological impacts across the lifespan. WMM parameters were extracted from UK Biobank diffusion magnetic resonance imaging data for 15q11.2 BP1-BP2 deletion (n = 126, mean age: 66 ± 8) and duplication (n = 131, mean age: 64 ± 7) carriers, as

Transcranial acoustoelectric imaging (tABI) of seizure activity in human head model with neuronavigation.

Objective. The human brain consists of multiple interacting neuronal networks that interleave at fine spatial and temporal scales. This complexity presents a challenge for scalp EEG and other noninvasive mapping techniques to accurately identify seizures and abnormal current patterns from background activity. To address this unmet need, this study investigates transcranial acoustoelectric brain imaging (tABI) with neuronavigation as a new method for mapping EEG-derived currents through the skull.Approach.In tABI, ultrasound (US) is focused and steered in the brain as surface electrodes record a

Clinical and Genetic Characteristics of Free Sialic Acid Storage Disorder.

Free sialic acid storage disorder (FSASD) is a lysosomal storage disorder that results from biallelic pathogenic variants in the SLC17A5 gene. This gene codes for sialin, a 12-transmembrane domain protein that exports the charged sugar N-acetylneuraminic acid (Neu5Ac; sialic acid) out of the lysosome. Dysfunctional sialin causes accumulation of free sialic acid within lysosomes and a range of clinical manifestations, such as intellectual disability, facial dysmorphisms, and increased urinary excretion of free sialic acid. These findings, along with characteristic brain abnormalities on MRI, ma

Clinical Heterogeneity in a Scandinavian FMR1 Premutation Carrier Cohort and Basal Ganglia Atrophy in FXTAS.

Pathological CGG expansions in the FMR1 gene, encompassing premutations (55–200 repeats) and full range mutations (> 200 repeats), cause a spectrum of complex and incurable disorders. Premutation carriers (PMC), particularly men, face the risk of developing fragile X-associated tremor/ataxia syndrome (FXTAS). Other conditions associated with premutations in FMR1 include both Fragile X-associated primary ovarian insufficiency (FXPOI), and Fragile X-associated neuropsychiatric disorders (FXAND). Hyperintensities in the middle cerebellar peduncle (MCP), known as the MCP sign, have been considered

Diffusion abnormalities associated with brain arteriolosclerosis: An in-vivo MRI and pathology study in community-based older adults.

Brain arteriolosclerosis, a primary pathology of cerebral small vessel disease, is common in older adults and is associated with lower cognitive and motor function and higher odds of dementia. The purpose of this study was to test the hypothesis that arteriolosclerosis is associated with lower diffusion anisotropy and higher trace of the diffusion tensor in white matter, independently of other age-related neuropathologies and visible white matter hyperintensities (WMH). In-vivo diffusion MRI and detailed postmortem neuropathologic examination were combined in 154 community-based older adults p

White matter abnormalities in metabolic syndrome patients with and without mild cognitive impairment: A diffusion tensor imaging study.

BACKGROUND: The presence of metabolic syndrome (MetS) constitutes a significant risk factor for the development of cognitive dysfunction and dementia. MetS can result in damage to the integrity and microstructure of white matter (WM). There may exist certain distinctions in WM between MetS patients with mild cognitive impairment (MCI) and MetS patients with preserved cognitive function. Using diffusion tensor imaging (DTI), we aim to investigate the alterations of the WM in MetS patients with and without MCI. METHODS: 36 MetS patients with MCI (MetS-MCI group), 30 age-, gender-, and education

Identification of a diagnosis-selective neurobiological substrate for bipolar disorder, major depressive disorder, and schizophrenia: a meta-analysis of 57,717 subjects.

BACKGROUND: Neuroimaging studies have consistently revealed neuroanatomical abnormalities in individuals with bipolar disorder (BD), major depressive disorder (MDD), and schizophrenia (SZ). However, it remains unknown whether and to what extent disorder-selective gray matter variations occur in these prominent psychiatric disorders. This study conducted a meta-analysis of 25 years of published voxel-based morphometry (VBM) research to assess the presence of selective and robust neuroanatomical substrates of gray matter variation in BD, MDD, and SZ. METHODS: Peer-reviewed experiments encompass

Mediodorsal and pulvinar thalamus reductions across the psychosis spectrum.

BACKGROUND: Abnormalities in thalamic structure and function are a consistent feature of psychosis spectrum disorders including schizophrenia (SZ), schizoaffective (SAD), and bipolar disorder with psychosis (BDP), yet the extent to which specific thalamic nuclei differ across diagnoses, and how these differences relate to cognition and psychopathology, remains unclear. We examined these questions, focusing on two nuclei - the mediodorsal (MD) and pulvinar (Pu), among individuals with psychosis (n = 2160) in the Bipolar-Schizophrenia Network on Intermediate Phenotypes consortium. METHODS: Whol

Multi-reader evaluation of deep learning-based auto-segmentation of eloquent brain arteriovenous malformation on MRA and white matter tractography in stereotactic radiosurgery.

OBJECTIVE: To minimize the radiation injury for white matter (WM) pathways during brain arteriovenous malformation (bAVM) stereotactic radiosurgery (SRS), the WM tractography is integrated into treatment planning to identify WM pathways and restrict receiving dose. Manual segmentation of eloquent bAVM adjacent to WM pathways is time-consuming and prone to substantial inter-practitioner variability due to intricate entanglement within eloquent brain areas. The objective of this study is to develop and evaluate a deep learning (DL) system for the segmentation of eloquent bAVM in a clinical setti

Polygenic contributions to brain cortical measures, subcortical volumes and hippocampal subfields in first episode psychosis.

BACKGROUND: Psychosis is characterized by both genetic and neurostructural abnormalities. However, the mechanisms linking genetic risk to brain structural alterations remain unclear. This study investigates associations between polygenic risk scores (PRS) and brain structural measures in individuals experiencing a first-episode psychosis (FEP) and healthy controls (HC). METHODS: A total of 241 participants (130 FEP, 111 HC, mean age = 24.8 years, 34.9% females) underwent structural magnetic resonance imaging (MRI) and genotyping. Intermediate structural covariance network abnormalities in argyrophilic grain disease between Alzheimer's disease and healthy controls.

BackgroundArgyrophilic grain disease (AGD) is a common tauopathy in the elderly, but its neuroimaging features remain less well characterized compared with Alzheimer's disease (AD). Notably, structural covariance network (SCN) analysis has not previously been applied to AGD.ObjectiveThis study aimed to investigate SCN alterations in pathologically confirmed AGD and AD and to characterize disease-specific patterns of network disruption.MethodsWe examined 12 AGD, 13 AD, and 18 healthy controls (HC). Individualized structural covariance matrices were constructed from regional gray matter volumes,

Beyond β-amyloid: transthyretin cerebral amyloid angiopathy.

Cerebral amyloid angiopathy (CAA) is most commonly associated with β-amyloid deposition, but other amyloid proteins may accumulate in leptomeningeal and superficial cortical vessels. We report a man first evaluated neurologically at 44 years of age after approximately one year of recurrent upper-limb and perioral sensory symptoms with speech disturbance, retrospectively considered transient focal neurological episodes (TFNEs). During follow-up, he developed bilateral vitritis and progressive behavioral, psychiatric, and cognitive impairment. Brain computed tomography showed diffuse linear and

Postmortem MRI reveals distinct structural features in sudden unexpected death in epilepsy.

Magnetic resonance imaging (MRI) abnormalities have been reported in individuals who later die from sudden unexpected death in epilepsy (SUDEP), but their specificity and predictive value remain uncertain. Postmortem MRI (PM-MRI) offers a unique opportunity to distinguish structural features associated with SUDEP from changes related to epilepsy, comorbid illness, or the postmortem interval. We performed PM-MRI in nine individuals: five with suspected SUDEP, two with epilepsy who died from non-seizure-related causes, and two without epilepsy who died from sudden cardiac death. Early deviations from normative brain morphology and cortical microstructure in schizophrenia spectrum disorders.

In schizophrenia spectrum disorders (SSD), structural alterations of the gray matter (GM) and white matter (WM) have been widely described. However, the complex interplay between early disease-related changes and ongoing brain maturation challenges our ability to identify early biomarkers. In this study, we investigated structural abnormalities and their association with symptoms in a drug-naïve or minimally medicated sample comprising 113 patients with SSD and 112 neurotypical controls. Specifically, we derived centile scores using normative modelling from cortical thickness (CT), and subcort
