Impact of environmental particulates on zebrafish embryo development and gene expression.

Particulate matter (PM) poses risks to environmental and human health, yet its toxicity mechanisms in aquatic organisms remain unclear. This study investigated the effects of 2 PM types, a standard reference material (S-PM10, NIST, USA) and particulates from the Mae Moh Power Plant, Thailand (MMPS), on zebrafish embryo development and gene expression. Embryos were exposed to various concentrations, and mortality, hatching rates, and morphological abnormalities were assessed. S-PM10, with irregular morphology and broad particle size, induced developmental defects and reduced hatching. MMPS, cha

EP4/ANXA2 axis in pulmonary arterial hypertension: therapeutic implications.

BACKGROUND AND AIMS: Pulmonary arterial hypertension (PAH) is a progressive condition marked by the abnormal proliferation of pulmonary artery smooth muscle cells (PASMCs), leading to significant remodelling of the pulmonary arteries (PAs). The cyclooxygenase metabolite of arachidonic acid prostaglandin E2 and its receptor EP4 are crucial for maintaining vascular homeostasis. This study aimed to determine the role of EP4 in the pathogenesis of PAH and evaluate the potential of EP4 as a therapeutic target for PAH. METHODS: Two well-established PAH models, the monocrotaline-induced rat model an

Exploring the link between disturbed flow and endothelial cell function in an in vitro arteriovenous fistula model.

BACKGROUND: The disturbed flow contributes to juxta-anastomotic intimal hyperplasia (IH) in arteriovenous fistulas (AVFs). This study developed an in vitro method aiming to understand the hemodynamic impact on endothelial cells (ECs) in AVFs. METHODS: A tubular bifurcation AVF model was constructed, and the disturbed flow was induced near the bifurcation by pulsatile flow. Hemodynamics was simulated using computational fluid dynamics (CFD) and visualized as 2D contour plots. Human Umbilical Vein Endothelial Cells (HUVECs) were cultured on a tailored polycarbonate membrane (PCM) and placed in

Caffeic Acid Phenethyl Ester Improves Right Ventricular Function and Reduces Arrhythmogenesis by Attenuating Structural and Electromechanical Remodeling in Pulmonary Arterial Hypertensive Rats.

Right heart failure is a major cause of mortality in patients with pulmonary arterial hypertension (PAH). This study aimed to evaluate the preventive anti-remodeling effect of a natural phenolic compound, caffeic acid phenethyl ester (CAPE), on pressure-overloaded right hearts. PAH was induced in Sprague-Dawley rats by intraperitoneal injection of monocrotaline (MCT; 60 mg/kg). The rats were randomly treated with CAPE (30 mg/kg/day, i.p.) or vehicle for 28 days. Right ventricular (RV) function was assessed, and remodeling was examined using both ex vivo and in vitro analyses. Chronic CAPE trea

Phenotyping the Structure and Function of the Heart of Elite Sailors: Implications for Pre-Participation Cardiac Screening.

Background: Structural and functional adaptation of the heart to chronic exercise is dependent on multiple factors, including the volume and type of training, and has direct implications for pre-participation cardiac screening (PPCS). Sailing is a unique multi-training modality sport with limited prior description of cardiac adaptation to training. The aims of this study are (1) to describe electrocardiogram (ECG) changes in sailors, informing PPCS guidelines; (2) to assess structural and functional cardiac changes in sailors; and (3) to examine sex- or discipline-specific cardiac adaptations

The Role of the NF-κB Signaling Pathway in Atherosclerotic Plaque Rupture and Targeted Therapeutic Strategies.

Atherosclerosis (AS) is a disease characterized by chronic vascular wall inflammation and lipid deposition. Although lipid-lowering drugs such as statins have significantly reduced cardiovascular event rates, "residual inflammatory risk" remains a key factor driving disease progression and plaque rupture. As a central regulator of the inflammatory response, the nuclear factor-κappaB (NF-κB) signaling network comprises both canonical pro-inflammatory pathways and functionally more complex non-canonical pathways. Increasing evidence in recent years indicates that abnormal and sustained activatio

BACKGROUND: Cardiovascular disease is the number 1 killer in industrialized countries, with sudden cardiac death due to ventricular arrhythmias representing a major component. To reduce sudden cardiac death, accurate risk prediction and development of effective preventive treatments remain major challenges. In this study, we explored the possibility of using a population-based computational modeling approach to perform virtual clinical trials for antiarrhythmic drug discovery and drug safety testing. METHODS: We developed genetically diverse populations of 1-dimensional cardiac tissue models

Genetic determinants of heart failure susceptibility and response in the collaborative cross mouse population.

Genetic variation and lived experiences shape how our hearts respond to chronic stress and development of heart failure, manifested as compromised pumping function and abnormal hemodynamics. The hallmark of heart failure etiology is excessive stress signals followed by maladaptive structural, electrical, and functional changes to the heart muscle, also known as cardiac remodeling. The specific genetic mechanisms which underly such phenomenon, however, are still unclear, due in part to difficulties in accounting for environmental effects in human population studies. To overcome this challenge,

Echinatin Alleviates Experimental Pulmonary Hypertension by PPARγ Pathway Activation.

The peroxisome proliferator-activated receptor gamma (PPARγ) signaling pathway plays a pivotal role in regulating the proliferation, apoptosis, inflammation, and metabolism of pulmonary arterial smooth muscle cells (PASMCs), suggesting its central involvement in the pathogenesis of pulmonary hypertension (PH). Although PPARγ agonists have shown promising therapeutic effects in preclinical models, their clinical translation remains challenging. Echinatin (Ecn), a natural flavonoid compound, has been reported to exhibit anti-inflammatory, antioxidant, and anti-proliferative properties by modulat

Electrocardiographic predictors of ventricular arrhythmic events in ICD recipients: signal beyond the left ventricular ejection fraction threshold.

BACKGROUND AND OBJECTIVES: Selection for implantable cardioverter-defibrillator (ICD) therapy based on left ventricular ejection fraction (LVEF) may miss at-risk patients with LVEF > 35%. We tested whether a simple electrocardiogram (ECG) burden adds discriminatory power for appropriate ICD therapy beyond clinical factors and LVEF. METHODS: Single-center retrospective cohort of 236 ICD recipients. Six routinely reported ECG abnormalities (heart rate > 75 bpm; QRS duration > 110 ms; obtuse QRS–T angle; delayed transition zone; prolonged QTc; prolonged Tp–Te) were prespecified, summed to form an

The Aldehyde Dehydrogenase 2 rs671 Variant Enhances Platelet Activation and Arterial Thrombosis.

BACKGROUND: Acute myocardial infarction (AMI) caused by thrombosis is a major cause of mortality. A polymorphism in Aldh2 gene (rs671) is found in approximately 30% to 50% of East Asians, and it is a risk factor for AMI. This mutation impairs aldehyde dehydrogenase 2 (ALDH2) function, but the effect of ALDH2 on platelet activation and thrombosis is unknown. METHODS: Platelets were isolated from platelet-specific Aldh2 knockout (Aldh2-/-) and ALDH2E506K knock-in mice (which corresponds to human Aldh2 rs671 gene mutation), as well as from healthy human donors with the Aldh2 rs671.
