BACKGROUND: In Ghana and many other sub-Saharan African countries, onchocerciasis and malaria are common infectious diseases that affect people of all ages. Their geographic overlap in specific areas could influence the diagnosis and management of both diseases. This study investigated the occurrence and patterns of malaria among individuals of various age groups diagnosed with onchocerciasis in two districts: Nkwanta North District in the Oti region and Adaklu District in the Volta region of Ghana. METHODS: A retrospective case-control study of 345 participants was conducted using archived bl

Our prospective cross-sectional survey of dogs from Western Fiji aimed to evaluate the fitness of common diagnostic test modalities to rule-in Dirofilaria immitis infection in patients with suggestive clinical history or signs. In the absence of a perfect reference standard, we used latent class modelling to evaluate the relative diagnostic accuracy of two point-of-care (POC) detection modalities - the modified Knott's test (MKT) to detect circulating microfilaria and four antigen lateral flow immunoassays (LFI): Anigen Rapid Canine HW Ag Test® (Bionote Co.), SNAP® Heartworm RT Test (IDEXX Lab

Morbidity Management and Disability Prevention (MMDP) remain an under-addressed component of lymphatic filariasis elimination. Despite the simplicity of the hygiene-based regimen, adherence to these practices remains low due to multiple barriers, exacerbating chronic suffering. As the primary role remains vested in the patients, this study explores their perspectives and barriers to home-based care to inform strategies to improve outcomes. This study employed a mixed-methods approach using an explanatory sequential design. The quantitative phase involved a cross-sectional survey of 321 adult p

Heartworm disease (Dirofilaria immitis) is a globally distributed vector-borne zoonosis affecting canids and felids, which can be fatal by compromising the cardiorespiratory system. This study analyzes the annual and monthly infection risk in Colombia by integrating ecological niche modeling for Culex quinquefasciatus-the most important and widely distributed vector in the country-with the temperature-dependent number of D. immitis generations within the vector. Results identified the human footprint (78 %) as the primary determinant of vector distribution, surpassing climatic variables and co

BACKGROUND: Onchocerciasis is a filarial neglected tropical disease targeted by the World Health Organization for elimination (interruption) of transmission (EOT), principally by mass drug administration (MDA) with ivermectin. Variable effectiveness and success of MDA, among other factors, has led to a markedly heterogeneous contemporary spatial landscape of endemicity and transmission, with some foci having achieved or nearing EOT, while in others, transmission persists despite decades of MDA or has only recently been identified. Communities reaching EOT or free from infection are thus vulner

BACKGROUND: The lymphatic filariasis (LF) elimination program in Nepal was launched in 2003, three years after the implementation of a global program to eliminate lymphatic filariasis globally. Based on antigenic prevalence below the cut-off value, i.e., ≤ 2% shown by the transmission assessment survey (TASI) report of 2017-2018, the program was stopped in Nepal. The report indicated that antigen-positive children were clustered in the community and considered hotspots. In order to find the status of lymphatic filariasis transmission at the community level, the present study was designed in th

BACKGROUND: Loa (L.) loa hypermicrofilaraemia (≥ 8,000 mf/mL) increases the risk of severe adverse events during mass ivermectin administration for onchocerciasis control. Albendazole has been proposed as a potential alternative for reducing microfilaraemia prior to ivermectin administration.
METHODOLOGY AND PRINCIPAL FINDINGS: This prospective study was conducted in northern Gabon from November 2021 to April 2022. Individuals infected with L. loa were screened and allocated to three groups: two treatment arms receiving 400 mg or 800 mg of albendazole daily for 30 days among hypermicrofilarae

Heartworms (Dirofilaria immitis) are parasitic nematodes that cause significant cardiopulmonary-associated morbidity and mortality in canids worldwide. The global spread of heartworms is believed to have occurred alongside the dispersal of modern domesticated dogs over the past few hundred years. However, this conclusion has been based on limited sampling, both geographically and numerically. To test this theory more rigorously, we analyse the whole genomes of 127 adult heartworm specimens collected from mammalian carnivore hosts across Australia, the USA, Central America, Europe, and Asia. Dirofilaria immitis is a parasitic nematode responsible for canine heartworm disease. Current heartworm treatment options include melarsomine chloridrate (an arsenical) to treat adult parasites and the macrocyclic lactones. Unfortunately, resistance to macrocyclic lactones is emerging which is highlighting the need for the discovery of new anthelmintics. Cys-loop ligand-gated ion channels are an untapped source for novel drug targets essential for nematode neurotransmission. This work presents the isolation and preliminary pharmacological characterization of a glutamate-gated chloride channel,

Lymphatic filariasis (LF) is a parasitic disease caused by filarial nematode worms, transmitted by mosquitoes. Infection damages the lymphatic system which leads to limb lymphedema and hydrocele. LF continues to remain one of the major neglected tropical diseases. Patients with filarial lymphedema are prone to nutritional deficiencies due to poverty, poor nutrition, old age, and frequent adenolymphangitis. However, studies on micronutrient deficiencies in affected patients remain scarce. In this study, we aimed to assess the levels of vitamin A and D in patients visiting a specialized clinic f

Zoonotic Onchocerca lupi (Spirurida, Onchocercidae) has attracted the interest of the scientific community worldwide, by causing severe ocular infections in domestic animals (dogs, cats) and can infect wild carnivores (wolves, coyotes), as well as humans. Though recent advancements in scientific knowledge have been gained, gaps still remain about the biology of this filarioid, as well as its genetic structure. Based on mitochondrial genes, two highly divergent genotypes were identified, in the Iberian Peninsula (genotype 2) and Europe, Asia, and the United States (genotype 1), meanwhile only a

Eosinophilia is a valuable biomarker for estimating the likelihood of parasitic infection in immigrants from tropical or subtropical regions. This study aimed to evaluate the frequency and etiology of imported eosinophilia in patients attending the Tropical Medicine Unit (TMU) of Salamanca, Spain, between 2008 and 2023. A total of 773 immigrant patients were assessed: 450 (58.2%) from Africa, 306 (39.6%) from Latin America, and 17 (2.2%) from Asia. Eosinophilia was observed in 338 patients (43.7%), of whom 15 (4.4%) had noninfectious causes. Among the remaining 323 evaluated for infections, 17

BACKGROUND: The interaction between helminth and viral infections has important implications for understanding viral disease outcomes and vaccine efficacy in helminth-endemic regions. We previously demonstrated that helminth seropositivity is associated with reduced Th1/Th17 cytokine levels and reduced COVID-19 severity; however, the underlying immunological mechanisms remain unclear. This study further investigated these mechanisms by assessing how helminth antigens influence SARS-CoV-2-induced T-cell responses in individuals infected with filarial parasites in vitro.

INTRODUCTION: India targets to eliminate lymphatic filariasis (LF) in alignment with the global goals. By 2024, 106 out of a total of 345 endemic districts have passed all three serial transmission assessment surveys (TAS) and are under post-mass drug administration (MDA) surveillance for a variable period. However, the current epidemiological situation of LF is not known in these districts. With increased mobility of population from the endemic districts currently under MDA to these post-MDA areas, resurgence of LF in these areas cannot be ruled out. Therefore, a study is planned to understan

Global efforts to control heartworm disease are challenged by increasing drug resistance and inadequate early detection methods, which hinder the effective diagnosis and treatment of Dirofilaria immitis infection. This study presents an in vivo model for investigating D. immitis development and host-parasite interactions. The growth and migration of the D. immitis JYD-34 isolate were analyzed in immunodeficient NSG mice over 177 days, revealing parasite migration to the heart and lungs, which mirrors infection dynamics in the natural canine host. Pathophysiological examination of host tissues

The Death to Onchocerciasis and Lymphatic Filariasis (DOLF) project conducts clinical and translational research to accelerate the global elimination of lymphatic filariasis (LF) and onchocerciasis (river blindness). Research needs were identified during several meetings of experts and helped study design. Over the last 15 years the project has conducted mass drug administration (MDA) studies and clinical trials with outstanding research teams in 11 countries that have led to many important discoveries. For example, DOLF studies showed that annual MDA was as effective as semiannual MDA for LF

The current guidelines for stopping treatment and verifying elimination of onchocerciasis recommend entomological and epidemiological surveys to be performed during the peak transmission season, which is generally assumed to occur during the rainy season, although this may not necessarily hold true in all transmission zones (TZs). This review has synthesized data, from a literature search, on the effects of seasonality on transmission in West and Central Africa. Seasonality affects vector population dynamics by altering terrestrial and aquatic ecology. In TZs with perennial rivers, peak transm

BACKGROUND: The END Fund's Field Entomology Mentorship Program addresses a critical gap in trained entomologists supporting onchocerciasis elimination in Africa. Unlike conventional training, this hands-on mentorship equips early to mid-career entomologists with technical expertise in critical onchocerciasis blackfly surveillance activities.
METHODS: The 3-y program follows a phased approach, gradually increasing mentees' independence while ensuring strong mentorship.
RESULTS: In its inaugural year, six mentees engaged in field activities across multiple countries, contributing to the develo
