Intranasal Curcumin and N-Acetyl l-Cysteine (NAC) Attenuates Dibutyl Phthalate (DBP)-Aggravated Airway Inflammation by Targeting Ferroptosis via Nrf-2/GPx4-SLC7A11.

Ferroptosis plays a significant role in the pathophysiological development of several diseases. It is an iron-dependent type of controlled cell death triggered by oxidative stress and lipid peroxidation. Nrf2, a key regulator of the antioxidant response, protects cells from ferroptosis by regulating genes involved in iron metabolism and the synthesis and breakdown of glutathione (GSH). This study was undertaken to investigate the relationship between Nrf2-mediated oxidative stress and ferroptosis in allergic asthmatic mice, particularly when the environmental toxin DBP is present. DBP disrupts

RNF19A facilitates gastric cancer progression by regulating the Hippo/YAP axis.

BACKGROUND: RNF19A is a poorly characterized ring between ring finger (RBR) E3 ubiquitin ligase that acts as a tumor suppressor in bladder cancer, whereas as an oncogene in non-small cell lung cancer. However, the biological functions of RNF19A in gastric cancer cells and whether RNF19A targets other substrates involved in gastric cancer progression remain unclear. Here, we report that RNF19A expression is elevated in gastric cancer tissues and is indicative of poor prognoses. METHODS: Microarray gastric cancer tissue sections were subjected to immunohistochemistry (IHC). The effects of RNF19

Modulation of the NF-κB signaling pathway by the combined strategy of tocilizumab and dexamethasone for asthma therapy.

Asthma, a prevalent chronic inflammatory airway disease, is conventionally managed with glucocorticoids (GCs) such as dexamethasone (DEX). However, prolonged DEX use induces adverse effects, including immunosuppression and metabolic disruptions. Tocilizumab (TCZ), a humanized monoclonal antibody with immunomodulatory and anti-inflammatory properties, may enhance therapeutic efficacy while minimizing DEX-related toxicities. Herein, we evaluate a combined therapeutic strategy integrating TCZ with a reduced DEX dose (TCZ + DEX) for asthma management. In lung epithelial cells subjected to hydrogen

Relationship between dyspnoea and respiratory drive in mechanically ventilated patients: A prospective observational study.

BACKGROUND: Patients with high respiratory drive during mechanical ventilation (MV) may be at risk for ventilator-induced lung injury, making control crucial. While high respiratory drive is often associated with dyspnoea, the relationship remains unclear. OBJECTIVES: We examined the relationship between three dyspnoea-rating scales and respiratory drive, as measured by airway occlusion pressure (P0.1), in patients receiving MV. METHODS: A prospective observational study was conducted in the intensive care unit between February 2020 and March 2022. Adults (aged ≥20 years) expected to require

Suppression of CSF2RA macrophage polarisation impacts pathological cardiac remodelling in mice.

Granulocyte-macrophage colony-stimulating factor (GM-CSF; CSF2) has emerged as an important regulator of pro-inflammatory macrophage polarisation, with pro-inflammatory responses recognised to influence cardiac remodelling and repair after cardiac injury. We hypothesised that selective inhibition of CSF2RA, responsible for mediating CSF2 signalling in monocyte/macrophages, could promote reparative responses after cardiac injury through modulating macrophage/fibroblast communication. We demonstrate for the first time that pharmacological CSF2RA inhibition alters the inflammatory response and ca

Application and Progress of Functional Lung Avoidance Radiotherapy for Lung Cancer.

Lung cancer is a major disease that seriously threatens human life and health. As one of the main treatment methods for lung cancer, radiotherapy (RT), while combating cancer cells, inevitably causes damage to the surrounding normal lung tissues. Radiation-induced lung injury (RILI) is a common and serious complication of RT that can seriously affect the treatment process of patients, reducing their quality of life (QOL) and survival time. Conventional RT aims to reduce the incidence of RILI in patients by limiting the radiation dose to normal lung tissue. This approach assumes that lung funct

More is not better: The role of the leukocyte filter in ex vivo lung perfusion.

PURPOSE: A leukocyte filter has been empirically incorporated within ex vivo lung perfusion circuits since the advent of ex vivo lung perfusion. However, the utility of the leukocyte filter has never been fully defined, and studies have questioned the beneficial effects of the filter. Additionally, there have been suggestions that such filters may become quickly saturated in use requiring replacement for better effect. Thus, we sought to determine the effects of leukocyte filters on ex vivo lung perfusion and evaluated whether a conventional arterial filter would prove to be noninferior. METH

Influenza pneumonia mice under different immune conditions: changes in pulmonary microbiota and metabolites.

Pneumonia is the most common complication of influenza virus infection and is associated with a high mortality rate in immunocompromised individuals. Currently, neither vaccination nor antiviral therapy has achieved satisfactory therapeutic outcomes. Unlike other organs, the lungs harbor a distinct microbial community due to their unique exposure patterns to airborne particulates and pathogens. However, the characteristic alterations of the pulmonary microbiota and its metabolic products, as well as their potential association with immune suppression in influenza pneumonia, remain inadequately

Cell type-specific interactions induce tonic interferon production in endothelial cells in a pathogen-independent manner.

BACKGROUND: The endothelium promotes a non-adherent vascular surface that facilitates tissue perfusion, prevents clotting, and limits inflammation. Endothelial cells (ECs) execute these tissue-specific functions through the integration of signaling pathways promoted by growth factors, cytokines, extracellular matrix components, and signals from mechanosensory complexes. Furthermore, ECs secrete various molecular signals, leading to the establishment of a specific niche microenvironment. Importantly, ECs can serve as sentinels against invading viral pathogens, propagating anti-viral responses s

Lactate Dehydrogenase and Outcomes in Patients With HF and Reduced Ejection Fraction: Insights From GALACTIC-HF.

BACKGROUND: Lactate dehydrogenase (LDH) is a cytoplasmic enzyme found in most cells. Increased LDH levels are a nonspecific measure of cellular injury and may be prognostically important in heart failure (HF). OBJECTIVES: This study aims to assess the relationship between LDH and clinical characteristics and outcomes in heart failure and reduced ejection fraction (HFrEF). METHODS: Using data from GALACTIC-HF, a phase 3, randomized, placebo-controlled trial evaluating the efficacy and safety of omecamtiv mecarbil (OM) in patients with HFrEF, the relationship between LDH and clinical outcomes

Cerebral Tissue Pulmonary Embolism Masquerading as Primary Graft Dysfunction After Lung Transplantation.

Early graft dysfunction following lung transplantation is commonly attributed to primary graft dysfunction (PGD). However, other rare etiologies may present with similar clinical and radiologic features. We report a unique case of a 58-year-old male with idiopathic pulmonary fibrosis who underwent bilateral lung transplantation. The donor was a young adult with traumatic brain injury. Within hours post-transplant, the recipient developed severe hypoxemia and diffuse infiltrates on chest imaging, consistent with Grade 3 PGD. Despite supportive therapy, there was limited improvement. Histopathol

Inflammation-triggered self-immolative conjugates enable oral peptide delivery by overcoming gastrointestinal barriers.

Oral delivery of peptide therapeutics remains challenging due to gastrointestinal (GI) degradation and poor intestinal absorption. Here, we propose a self-immolative peptide prodrug conjugate (SIPPC) platform for inflammation-targeted oral delivery, integrating a hydrophilic polyethylene glycol segment, a reactive oxygen species (ROS)-responsive hydrophobic self-immolative module, and a hydrolyzable scaffold, which collectively enable spontaneous assembly into micelle-like nanoparticles. Using three anti-inflammatory peptides (KPV, Ac-QAW, and IRW), we demonstrated that the engineered conjugat

Respiratory oscillometry in monitoring lung transplant allograft function: a systematic scoping review.

Acute and chronic lung rejection are major barriers to the long-term survival of lung transplant recipients. Current spirometry-based monitoring of allograft function is limited by the low sensitivity in detecting distal airway abnormalities where allograft rejection evolves. Emerging evidence from haematopoietic stem cell transplant patients with chronic pulmonary graft-versus-host disease suggests that respiratory oscillometry may offer improved sensitivity in identifying small airway obstruction at earlier stages. In this scoping review, we explore the utility of oscillometry in monitoring

The ribosomal landscape in influenza A virus infection: from molecular mechanisms to clinical relevance.

Influenza A virus (IAV) infections continue to represent a significant global health concern, both in terms of severe individual cases of acute respiratory distress syndrome (ARDS) and the potential for the emergence of pandemics. Despite decades of research, therapeutic options remain limited and the pathogenesis of severe disease is not yet fully understood. One critical yet underappreciated aspect is how IAV reprogrammes the ribosomal landscape of the host to facilitate viral replication and evade immune responses. Ribosomes take centre stage during infection, as both the immune response an

Heterogeneous causes of acute respiratory distress syndrome correlate with distinct peripheral polyunsaturated fatty acid metabolites.

Acute Respiratory Distress Syndrome (ARDS), a heterogeneous syndrome of hypoxic respiratory failure secondary to dysregulated pulmonary inflammation, is caused by diverse insults. Because of this heterogeneity, mechanisms and treatments are difficult to study. As a treatment, n-3 polyunsaturated fatty acid (PUFA) supplementation has had mixed results. PUFAs and downstream oxylipins are important to pulmonary inflammation but are not well defined in ARDS. We hypothesized that differences in fatty acid metabolism, as measured by levels of n-3 and n-6 PUFAs and oxylipins, are associated with diff

Tetramethylpyrazine Targets HDAC6-Mediated PINK1 Degradation to Alleviate Chronic Intermittent Hypoxia-Induced Injury in Human Bronchial Epithelial Cells (16HBE).

Obstructive sleep apnea (OSA) is characterized by recurrent chronic intermittent hypoxia (CIH), which induces oxidative stress, inflammatory responses, and mitochondrial damage in bronchial epithelial cells. Tetramethylpyrazine (TMP) has been shown to exert lung-protective effects in other pathological models, but its role in mitigating CIH-induced 16HBE cell injury and the underlying molecular mechanisms have not been previously investigated. PTEN-induced putative kinase 1 (PINK1)-mediated mitophagy is a critical endogenous mechanism that defends against CIH-induced epithelial damage. However

Comparative evaluation of single-artery cannulation with passive venous drainage versus traditional dual-cannula ex vivo lung perfusion in a rat model.

BACKGROUND: Ex vivo lung perfusion (EVLP) has emerged as a critical technique for lung preservation and evaluation prior to transplantation. While conventional rat EVLP systems utilize closed-loop dual cannulation of pulmonary artery (PA) and vein, the effect of the simplified model using single PA cannulation with passive venous drainage is unknown. METHODS: We developed two EVLP models in rats: a semi-closed circuit with PA-only cannulation and left atrial incision for passive venous drainage (SC-EVLP), and a closed circuit employing both arterial and venous cannulation (C-EVLP). Donor lung

Genetic and chemical correction of cystic fibrosis reduces airway susceptibility to SARS-CoV-2.

The airway epithelium, a primary target for viral infection, plays a critical role in disease response-particularly in individuals with preexisting airway conditions such as cystic fibrosis (CF). At the onset of the SARS-CoV-2 pandemic, individuals with CF were expected to have severe outcomes based on earlier viral outbreaks; however, those on effective CF transmembrane conductance regulator (CFTR) modulators showed milder disease. Patients with CF on the CFTR modulator combination elexacaftor/tezacaftor/ivacaftor (ETI) combination therapy showed attenuated viral infection and reduced airway

Cisplatin as a Xenobiotic Agent: Molecular Mechanisms of Actions and Clinical Applications in Oncology.

Cisplatin, a platinum-based compound, is a cornerstone of modern chemotherapy and remains widely used against a variety of solid tumors, including testicular, ovarian, lung, bladder, and head and neck cancers. Its anticancer activity is primarily attributed to the formation of DNA crosslinks, which obstruct replication and repair, ultimately leading to apoptosis. However, the clinical value of cisplatin is constrained by two major challenges: its toxic profile and the development of resistance. Cisplatin toxicity arises from its interaction not only with tumor DNA but also with proteins and nu
