MnO2-loaded hydrogel nerve guidance conduit for peripheral nerve regeneration: anin vivostudy in the rat sciatic nerve defect model.

Peripheral nerve defect repair remains a clinical challenge. Nerve guidance conduits (NGCs) are potential alternatives to autografts, but their performance can be compromised by the oxidative and inflammatory microenvironment after injury. Here, we fabricated MnO2-loaded hydrogel NGCs at concentrations of 10, 50, and 100 μg ml-1and evaluated them in a rat model of a 10-mm sciatic nerve defect. Among the three concentrations, the MnO2@NMgel-10 (10 μg ml-1) group yielded a higher sciatic functional index, denser and more orderly regenerated nerve tissue, and less gastrocnemius muscle atrophy tha

The Optimal Surgical Approach for Deep Infiltrating Endometriosis Involving Pelvic Nerves (type 1 and 2).

OBJECTIVE: Excision of deep infiltrating endometriosis in the parametrium involving pelvic nerves can be challenging and requires good anatomical knowledge and surgical strategy [1]. Based on our experience, we have identified two important types of nodule in this compartment, differing in location and the pelvic structures involved, and therefore propose this new classification. This video aims to show a standardized, reproducible method for each type. SETTING: The surgeries were performed with the DaVinci Xi system at the IFEMEndo center in Bordeaux, France. Illustrations were produced with

Characterization of sensory nerve conduction abnormalities at the time of ALS diagnosis.

OBJECTIVES: Although sensory nerve abnormalities have been reported in amyotrophic lateral sclerosis (ALS), their distribution at diagnosis, relative to motor involvement and clinical onset phenotype remains incompletely characterized. We aimed to systematically compare sensory and motor nerve conduction abnormalities at ALS diagnosis and determine whether sensory involvement follows an onset-dependent pattern similar to motor dysfunction. METHODS: In this prospective cross-sectional study, 40 newly diagnosed ALS patients enrolled in the Iran University ALS Registry (March 2022-March 2023) un

Longitudinal Single-Axon-Resolution Imaging of Peripheral Nerve Injury Response in Mice Using an Optical Window Implant.

Chronic, high-resolution imaging of peripheral nerve injury and regeneration remains a major technical challenge due to the optical inaccessibility of peripheral nerves embedded in soft tissue. Here, we present a broadly applicable approach for longitudinal, high-resolution optical imaging of peripheral nerves in vivo that combines a flexible, optically transparent polydimethylsiloxane (PDMS) window along with multimodal microscopy. The PDMS window is implanted in the skin above the mouse sciatic nerve, providing stable, motion-tolerant optical access for over one month without impairing nerve

Prior Authorization for Sacral Nerve Stimulation in the Era of WISeR: Evaluation of U.S. Payer Coverage Policies.

INTRODUCTION: Sacral nerve stimulation (SNS) is an established therapy for refractory urinary incontinence, yet the 2025 Wasteful and Inappropriate Service Reduction Model (WISeR) designated it as requiring mandatory prior authorization. No analysis has examined how U.S. payers translate SNS evidence into coverage policies or whether restrictions are evidence-based. We sought to: (1) compare covered and excluded indications, (2) evaluate restriction criteria and conservative therapy requirements, and (3) assess quality of evidence cited. METHODS: This was a systematic, retrospective analysis

Betula utilis (bark extract) speeds up sciatic nerve function restoration following a compression injury in a mouse model.

BACKGROUND: People who experience various traumas often develop peripheral nerve injuries (PNIs). Currently available strategies are insufficient to fully recover nerve damage. Plants are used to treat a variety of illnesses. OBJECTIVES: In this study, we investigated the possible effect of Betula utilis on the restoration of muscle function following an injury to a peripheral nerve. METHODS: Here, we treated PNI by using bark extract from Betula utilis. First, 32 healthy albino mice were equally divided into 4 groups. Bark extract (orally) at varying concentrations (25, 50, 100, and 200 mg/

Spared Nav1.8-positive nociceptors drive persistent tactile hypersensitivity after sciatic nerve crush injury in mice.

Peripheral nerve injury can lead to chronic mechanical hypersensitivity, yet the severity and persistence of pain are strongly influenced by the extent of axonal damage. Notably, partial sciatic nerve crush injury (PCI) produces persistent tactile hypersensitivity despite a less severe anatomical insult than full crush injury, yet the identity and postinjury state of the fibers that persist after PCI remain unclear. To define sensory neuron populations contributing to PCI-induced tactile hypersensitivity, we combined fiber-specific transgenic labeling (Thy1-YFP for Aβ mechanoreceptors and Nav1

The therapeutic effects of ligustilide in cuprizone-induced sciatic nerve injury in rats via activation of autophagy and inhibition of apoptosis.

PURPOSE: Ligustilide, an active ingredient from Rhizoma Chuanxiong, exhibits anti-inflammatory, antioxidant, and neuroprotective effects. We aim to evaluate the protective effects of ligustilide against cuprizone-induced sciatic nerve injury in rats by assessing autophagy, fibrosis and apoptosis. METHODS: Cuprizone-induced sciatic nerve injury was investigated in rats. The severity of injury was evaluated using the open field test, the rotarod test, the wire grip test, and nerve conduction velocity measurements. Additionally, sections of the sciatic nerve were stained with Masson trichrome an

COL4A3 Promotes Invasion and act as Biomarker in Pituitary Neuroendocrine Tumor.

INTRODUCTION: The Transgluteal approach (TGa) to the hip, proposed by BAUER, and often used for total arthroplasties, could be responsible, according to some authors for bad clinical results, due to injury of the nervus gluteus superius (NGS). The aim of this study was to verify the nerve's anatomical condition and evaluate the risks of injury during TGa. MATERIAL AND METHODS: Thirty three dissections of fresh corpses in the lateral position (preceded by a TGa) permitted the estimation of the risk and the measurement of the distances between the nerve and the trochanter major (TM). The NGS wa

Sciatic nerve compression as a cause of severe chronic pain after total knee replacement: a case report.

BACKGROUND: Persistent pain following total knee replacemnt (TKR) remains a challenging and often multifactorial issue. While common causes include infection, malalignment, or complex regional pain syndrome, less typical origins are easily overlooked. We report a unique case of chronic, disproportionate posterior thigh pain 1 year after TKR, caused by a complete proximal hamstring rupture leading to symptomatic sciatic nerve compression. To our knowledge, this is the first published case describing this specific postoperative complication and its successful surgical management. CASE PRESENTAT

Analgesic Effects of Novel Spinal Cord Stimulation Waveforms in Rats With Painful Tibial Nerve Injury.

OBJECTIVES: Neuropathic pain is associated with hyperexcitability of spinal cord dorsal horn neurons. Spinal cord stimulation (SCS) can reduce this hyperexcitability and provide analgesia. We evaluated the effects of two novel SCS waveforms, designed with long pulse widths and short interpulse intervals with slowly varying amplitudes, on neuropathic pain behaviors and dorsal horn neuronal activity in a rat model of tibial nerve injury (TNI). MATERIALS AND METHODS: Rats underwent TNI followed by implantation of epidural stimulation leads. Novel waveforms, specifically stochastic amplitude modu

From anthelmintic to neurotherapeutic: exploring the regenerative potential of niclosamide in sciatic nerve injury.

Peripheral nerve injury (PNI) remains a major clinical challenge, often leading to chronic neuropathic pain and persistent motor and sensory dysfunction. Despite advances in microsurgical repair, effective pharmacological strategies to attenuate neuroinflammation and promote functional recovery are still lacking. Niclosamide, a well-known FDA-approved anthelmintic agent, has recently emerged as a potent modulator of multiple intracellular signaling pathways implicated in neuroinflammation. In the present study, we investigated the neuroprotective and anti-inflammatory effects of niclosamide in

Peroneal Nerve Dysfunction Following Vascularized Fibular Epiphyseal Transfer (VFET): A Prospective Case Series Study.

BACKGROUND: In skeletally immature individuals, vascularized epiphyseal transfer allows for reconstruction of complex bony defects while preserving longitudinal growth and providing a reliable articular surface for effective joint function. The shape and size of the proximal fibular epiphysis allow for gradual remodeling of the articular surface, facilitating favorable reconstruction and rehabilitation over time. Moreover, the proximal fibula is highly expandable, with less weight-bearing function, contributing to low donor-site morbidity. However, peroneal nerve dysfunction-related foot drop

Sex-related analgesic responses to photobiomodulation therapy in rats with neuropathic pain.

Neuropathic pain is associated with sensory abnormalities and remains difficult to treat. Photobiomodulation therapy (PBMT) has emerged as a non-pharmacological approach with analgesic potential; however, sex-related differences in treatment responses remain poorly understood. We investigated the effects of PBMT in male and female rats subjected to chronic constriction injury (CCI) of the sciatic nerve. Animals received infrared PBMT administered three times per week during ten sessions, and nociceptive responses were assessed using mechanical hyperalgesia and tactile allodynia throughout trea

Chronic Groin Pain in Female Cyclists: Consider Pudendal Neuroma as a Possible Cause.

A 58-year-old female cyclist is experiencing chronic right groin pain, adversely affecting her quality of life and athletic performance. Despite undergoing comprehensive diagnostic evaluations, including pelvic MRI and specialized physical examinations, the primary findings were chondrolabral separation and ischiofemoral impingement. Notably, a pudendal neuroma was identified through ultrasound imaging. The patient received ultrasound-guided corticosteroid injections (Depomedrone 20 mg) and local anesthetic (0.25% bupivacaine 1 mL, with 1% lidocaine for skin and subcutaneous tissues), substant

Comparison of the effects of intensities and sessions of extracorporeal shock wave therapy on nerve regeneration in rats with experimentally induced sciatic nerve injury.

Extracorporeal shock wave therapy (ESWT) has emerged as a promising treatment for neurological disorders but optimal protocols regarding energy density and session frequency remain unclear. This study aimed to evaluate the effects of different ESWT energy levels and treatment sessions on peripheral nerve regeneration in rats with sciatic nerve injury. Fifty rats were randomized into five groups: four experimental groups received ESWT at low energy (0.098 mJ/mm2) or high energy (0.229 mJ/mm2), either once or three times, and one control group received no treatment. Assessments included protein

Radioanatomical study of the proximity of the sciatic nerve and the pelvic bone.

PURPOSE: Percutaneous thermal ablation of pelvic bone tumours carries a risk of iatrogenic injury to the sciatic nerve. This study aimed to determine the proximity of the sciatic nerve to the iliac bone as it exits the pelvis and evaluate its anatomical variability. METHODS: This retrospective, non-interventional study reviewed consecutive MRI scans of the lumbar spine performed at our institution between September and October 2021. Two experienced musculoskeletal radiologists independently measured the shortest distance between the sciatic nerve and the adjacent bone at six predefined levels

Rare Common Peroneal Nerve Neuropraxia after Removal of Upper Tibial Pin Traction.

Common peroneal nerve (CPN) injury is an uncommon complication associated with skeletal traction. Neurological deficits occurring after removal of upper tibial pin traction (UTPT) are exceedingly rare and sparsely reported in the literature. A 27-year-old male sustained a closed left subtrochanteric femur fracture following a road traffic accident. Preoperative UTPT was applied with no immediate neurological deficit. UTPT was removed intraoperatively before definitive fixation. Postoperatively, the patient developed left-sided foot drop. Electromyography and nerve conduction velocity studies a

Schwann Cells in Development, Regeneration, and Therapeutic Applications: Insights From the Sciatic Nerve Model.

The sciatic nerve is the largest nerve in the human body and is frequently affected by lesions, causing pain and even loss of sensitivity. The sciatic nerve injury model provides a robust platform to study the degeneration-regeneration process. In this context, Schwann cells (SC) are central regulators of peripheral nervous system (PNS) development, homeostasis, and regeneration. During development, SC arise from neural crest-derived precursors and undergo tightly regulated lineage progression, culminating in myelinating and non-myelinating phenotypes. This process is orchestrated by coordinat
