Detorsion is not innocent: pirfenidone prevents contralateral ovarian damage following ovarian torsion.

Ovarian torsion is a gynecological emergency caused by rotation of the vascular pedicle, leading to ischemia, necrosis, and potential fertility loss. Management remains controversial, particularly regarding oophorectomy versus ovarian preservation. Detorsed ovaries continue cytokine secretion, impairing contralateral ovarian function, and no protective therapy currently exists. This study aimed to compare oophorectomy and detorsion strategies and to determine whether post-detorsion long-term pirfenidone therapy preserves ovarian function, thereby providing experimental resolution to the clinic

Coumarins from Tridax procumbens L. and their anti-inflammatory activity.

Twenty-two coumarins, including seven previously undescribed compounds, which comprised a pair of enantiomers (2a/2b) and five other compounds (1, 3-6), were isolated from the whole plant of Tridax procumbens L. Their structures were identified using spectroscopic techniques (NMR, IR, MS, and optical rotation), NMR calculation with DP4+ probability analysis, and ECD calculations. The anti-inflammatory effect of coumarins was evaluated by their inhibition of lipopolysaccharide-stimulated nitric oxide production in RAW 264.7 macrophages. nido-carborane conjugates with ibuprofen, flurbiprofen and fenoprofen: synthesis, characterization, COX inhibition potential and anticancer activity.

The search for novel anticancer drugs with reduced side effects and innovative therapeutic approaches remains a significant challenge. Non-steroidal anti-inflammatory drugs (NSAIDs), known to inhibit cyclooxygenase (COX) - an enzyme overexpressed in certain tumors - offer a promising avenue for cancer therapy. Additionally, icosahedral carboranes are emerging as a class of compounds with significant potential in drug design. Combining these two moieties into a single molecule presents a compelling strategy to develop drugs with dual mode of action and enhanced efficacy. Here, we report nido-ca

Mutual solubilization of rosiglitazone and ibuprofen: Investigation and mechanistic insight.

PURPOSE: We report for the first time that co-dissolved rosiglitazone and ibuprofen mutually enhance their aqueous solubility; the study was designed to quantify this phenomenon and clarify its origin. METHODS: Solubility was measured by HPLC. Fluorescence and FT-IR spectroscopy were used to probe molecular interactions, while classical molecular dynamics simulations provided atomistic insight. RESULTS: The fluorescence intensity of the mixed solution was markedly lower than that of either drug alone, indicating ground-state complexation or close association. FT-IR spectra showed no new peak

A unified biochemical-physical regulatory nanoparticle modulates tumor mechanics to augment nanomedicine penetration and antitumor efficacy.

The abnormal mechanical microenvironment in solid tumors is a self-reinforcing alliance between activated cancer-associated fibroblasts (CAFs) and dense extracellular matrix (ECM), forming an impenetrable barrier that shuts out nanomedicines. Biochemical approaches suppress CAFs but poorly penetrate stroma or relieve matrix barriers, whereas physical methods loosen ECM transiently but lack specificity and sustained CAFs inhibition. Here we report a responsive drug conjugate (PFD-TK-Cy) that self-assembles into nanoparticles (PFD-TK-Cy NPs) for integrated biochemical CAFs inhibition and physica

Orally delivered, pH-resistant dual coated extracellular vesicles restore intestinal barrier function and suppress colitis.

Oral therapy for inflammatory bowel disease (IBD) requires drug formulations that can withstand gastrointestinal stress and achieve localized action at the inflamed mucosa, yet existing extracellular vesicles (EVs)-based or small-molecule approaches often fail due to poor stability and limited colonic delivery. This study aimed to develop a dual-coated oral delivery platform that enhances gastrointestinal stability and targeted drug release. We engineered macrophage-derived EVs sequentially coated with a cationic lipid and sodium alginate (SA) to encapsulate 5-aminosalicylic acid (5-ASA). This

Renal Tubular Acidosis Leading to Severe Hypokalemia: An Uncommon Adverse Effect of Nonsteroidal Anti-inflammatory Drugs.

Nonsteroidal anti-inflammatory drugs (NSAIDs) are commonly used over-the-counter pain-relieving medications. They are not free from side effects, including effects on the kidneys. We present the case of an elderly woman who presented with severe fatigue and generalized weakness. Her medical history was significant for osteoarthritis, for which her pain was controlled with ibuprofen and naproxen. She was found to have renal tubular acidosis (RTA), causing severe, persistent hypokalemia. The underlying etiology was concluded to be secondary to NSAID use, making this case unique. She gradually im

Discovery of ferulic acid carbamate derivatives as dual-targeting agents of BuChE and Nrf2 for Alzheimer's disease.

Given the multifactorial aetiology of Alzheimer's disease, multi-target strategies have emerged as a promising therapeutic approach. In this study, we designed and synthesised a series of ferulic acid carbamate derivatives to selectively inhibit BuChE and stimulate Nrf2 pathway. The biological evaluation revealed that compound 5c and 5e were the most potent, exhibiting over 150-fold selectivity for BuChE. Also, 5c, 5g and 5h significantly reversed both H2O2- and Aβ-induced toxicity in HT22 cells. These compounds were further shown to eliminate ROS accumulation induced by Aβ and upregulated HO-

In vitro and preclinical assessment of drug interactions between fluoroquinolones and a nonsteroidal antiinflammatory drug predicting risk of seizure.

Seizures are a common reason for drugs to fail during development, but they are difficult to predict preclinically. Enoxacin, a fluoroquinolone, rarely causes convulsions as a monotherapy, but convulsions have been seen after combination therapy with fenbufen, a nonsteroidal antiinflammatory drug. The interaction between the drugs is thought to result from inhibition of GABAA receptors, which are not their primary targets. Here, we show that, among 15 marketed fluoroquinolones and 1 active metabolite, 6, including enoxacin, inhibited γ-aminobutyric acid (GABA)-evoked depolarization in cells ex

Acid-sensing ion channels and their role in controlling pain in the post-operative stage.

Acid-sensing ion channels (ASICs) are proton-gated cation channels that are widely expressed throughout the nervous system and play a central role in sensory processing, particularly in nociception. Due to their involvement in key physiological processes, ASICs have emerged as promising targets for the modulation of pain perception. Postoperative pain management is primarily dependent on pharmacological agents, such as opioids and nonsteroidal anti-inflammatory drugs (NSAIDs), to alleviate discomfort and facilitate recovery. However, the long-term use of these analgesics is associated with sig
