A Comparison of Opioid Overdose Risk Behaviors by Race and Ethnicity Among Overdose Survivors in Boston, MA, and San Francisco, CA.

BACKGROUND: Black and Latine people are at high risk for opioid overdose mortality. Interventions focused on overdose risk behaviors may offer opportunities to reduce the risk of overdose, but little research has addressed racial and ethnic differences in risk behaviors. We explored differences in risk behaviors among Black, Latine, and White individuals who used opioids and had a history of prior overdose. METHODS: We used data from REBOOT 2.0 (the REpeated dose Behavioral intervention to reduce Opioid Overdose Trial) based in Boston, MA, and San Francisco, CA, which enrolled people with opi

Bromazolam Tablet Quantification and Analysis of Post-Mortem Cases From the National Programme on Substance Use Mortality (NPSUM).

Bromazolam is a new psychoactive substance (NPS) benzodiazepine commonly identified by drug checking services and in post-mortem toxicological analyses in the United Kingdom, Europe, and North America. At the time of writing, there are no studies that present quantitative analyses of bromazolam in street tablets. Here we describe the first quantitative analysis of bromazolam tablets, from samples submitted by UK drug checking services and police forces between 2022 and 2025. Using validated GC-EI-MS and 1H NMR methods, 47 tablet samples were quantified revealing a median bromazolam dose of 0.4

As-Needed Medication Use for Agitation Across Valproic Acid Formulations.

BACKGROUND: Pharmacokinetic differences between immediate-, delayed- and extended-release (IR, DR, ER) formulations of valproic acid (VPA) may impact therapeutic outcomes. As-needed, pro re nata (PRN) medications may serve as proxies for scheduled medication effectiveness for symptom management. This study compared the number of PRN doses of benzodiazepines and antipsychotics for agitation between VPA formulations. METHODS: A retrospective review was conducted for patients with bipolar I or schizoaffective disorder who received three or more days of VPA (IR, DR, ER) doses as maintenance thera

Intramuscular droperidol, olanzapine, midazolam, or lorazepam to treat methamphetamine intoxication in the emergency department.

INTRODUCTION: Emergency department (ED) visits for acute methamphetamine-associated agitation are increasing. Many cases require parenteral medications. Benzodiazepines are often recommended first-line, however randomized trials of intravenous medications suggest antipsychotics are also effective. High-quality data on intramuscular medications are lacking. OBJECTIVE: To compare the effectiveness of intramuscular droperidol, olanzapine, midazolam, and lorazepam to treat methamphetamine-associated agitation in the ED. METHODS: This was a secondary analysis of previously published data from 201

Impact of Comorbid Generalized Anxiety Disorder on rTMS/iTBS Clinical Outcomes in Major Depression: A Multicenter Registry-Based Observational Study.

Background: Major depressive disorder (MDD) is often accompanied by generalized anxiety disorder (GAD), a comorbidity linked to greater illness burden and potentially poorer outcomes. Repetitive transcranial magnetic stimulation (rTMS) and intermittent theta-burst stimulation (iTBS) are established treatments for MDD, yet the impact of comorbid GAD and concomitant medications remains unclear. This study aimed to compare rTMS/iTBS treatment outcomes between patients with MDD with and without comorbid GAD, and to examine the association between concomitant psychotropic medication use, stimulatio

Synaptic correlates of benzodiazepine tolerance.

Benzodiazepines are among the most widely prescribed psychotropic agents, exhibiting anxiolytic, hypnotic, anticonvulsant, and myorelaxant properties primarily mediated through positive allosteric modulation of type A γ-aminobutyric acid receptors (GABAARs)-the principal inhibitory receptors in the mammalian brain. However, prolonged benzodiazepine administration frequently results in the development of tolerance, a phenomenon characterized by reduced pharmacological efficacy and the need for escalating doses to sustain therapeutic benefit. Accumulating evidence indicates that benzodiazepine t
