The protein extracted from fermented Zanthoxylum bungeanum seeds mitigates DSS-induced ulcerative colitis by orchestrating inflammatory cytokine modulation, counteracting oxidative stress, and safeguarding gut microbiota homeostasis.

Zanthoxylum bungeanum seeds (ZBS) are protein-rich by-products with limited high-value utilization, and fermentation may provide an effective strategy to improve the functional properties and bioactivity of their proteins. In this study, fermented ZBS protein (FZP) was prepared, and its physicochemical properties, antioxidant activity, gastrointestinal digestibility, and protective effects against colitis were systematically investigated. FZP exhibited a high purity of 94.37% and was predominantly composed of proteins with molecular weights ranging from 15 to 25 kDa. Ultrafiltration revealed t

The Small GTPase RHO1 Regulates ROS Homeostasis and Differentially Responds to Pathogens Infection via Interaction With Aquaporin TIP1;1 in Nicotiana.

Small G-proteins are crucial regulators in plant growth and environmental responses. However, the mechanistic basis underlying their regulation of plant immunity to different pathogens is still poorly understood. Here, we showed that the small GTPase Ras homologous 1 (RHO1) responded differently to infection by distinct pathogens. Overexpression of RHO1 increased the plant susceptibility to chilli veinal mottle virus (ChiVMV) and necrotrophic Botrytis cinerea infections but enhanced plant resistance to the biotrophic bacterium Pseudomonas syringae. Further exploration revealed that there exist

Caulerpa peltata extract protects against Dextran sodium sulfate-induced acute ulcerative colitis via modulating Nrf2 pathway.

Ulcerative colitis (UC) is one of the inflammatory bowel diseases (IBD) and poses an increasing public health issue around the world because of the presence of genetic, immunological, and environmental causes. Antioxidants and anti-inflammatory therapy targeting oxidative stress and inflammatory pathways have become the main therapeutic approach with the use of the NRF2 pathway. In this study, the antioxidative, anti-inflammatory and NRF2 regulation activities of ethanolic extract of Caulerpa peltata (CPEE) were investigated using phytochemical screening, in vitro, in vivo, and in silico metho

Matrix metalloproteinase-13 selective inhibitors.

Matrix metalloproteinase-13 (MMP13) is the primary protease responsible for the irreversible degradation of type II collagen in the extracellular matrix (ECM), playing a pivotal role in the pathogenesis of osteoarthritis (OA), rheumatoid arthritis (RA), and various malignant cancers. While early clinical efforts with broad-spectrum zinc-chelating inhibitors were abandoned due to dose-limiting musculoskeletal syndrome (MSS), recent research has focused on achieving high isoform selectivity by targeting unique structural features such as the deep S1' specificity pocket and the MMP13-specific S1'

SIRT3-mediated DRP1 acetylation changes are associated with the restoration of mitochondrial function and alleviation of anterior cruciate ligament injury-induced osteoarthritis.

Mitochondrial dysfunction and endoplasmic reticulum stress (ERS) play critical roles in the development of osteoarthritis (OA). Sirtuin 3 (SIRT3), a mitochondrial deacetylase, has regulatory effects on OA, but whether it can influence OA progression by modulating the crosstalk between mitochondrial dysfunction and ERS remains unclear. Therefore, the aim of this study was to investigate the effects of SIRT3 on mitochondrial dysfunction and ERS in OA. Anterior cruciate ligament transection was performed to establish an OA rat model, and C28/I2 chondrocytes were treated with IL-1β to establish an

Differential regulation of SOCS signalling and immunoproteasome pathways by JAK inhibitors and TNF inhibitors in rheumatoid arthritis.

BACKGROUND: Janus kinase (JAK) inhibitors and TNF inhibitors (TNFi) are established therapies in rheumatoid arthritis (RA), yet their differential effects on the JAK/STAT/SOCS/proteasome axis remain incompletely understood. This pilot study systematically characterised treatment-specific effects of baricitinib, tofacitinib, and TNFi upon molecular signatures in RA patients. METHODS: Seventy-five patients with RA were enrolled across two prospective cohorts: an initial inception cohort treated with tofacitinib (n = 15) and a subsequent confirmatory cohort treated with baricitinib or TNFi (n =

FAN1 mutation and karyomegalic nephropathy in chronic kidney disease.

Karyomegalic Interstitial Nephritis (KIN) is a rare hereditary cause of chronic kidney disease (CKD), resulting from autosomal recessive mutations in the FAN1 gene. Renal biopsy typically reveals severe interstitial fibrosis, tubular injury and glomerulosclerosis. Early and accurate diagnosis can guide appropriate management. We present a middle-aged euglycaemic, normotensive female who underwent renal biopsy, which revealed tubular vacuolisation, patchy acute injury and enlarged, bizarre nuclei (karyomegaly) with occasional intranuclear cytoplasmic inclusions. Clinical exome sequencing confir

PARP7 drives angiotensin II-induced vascular remodeling via ADP-ribosylation and inhibition of GPX4.

Pathological vascular remodeling is driven by metabolic imbalance in vascular smooth muscle cells (VSMCs). NAD+ homeostasis is essential for sustaining VSMC contractility, quiescence, and genomic stability. ADP-ribosylation, catalyzed by poly(ADP-ribose) polymerase (PARP) enzymes, is an NAD+-dependent modification that links NAD+ consumption to cellular stress. Here, we identify the mono-ADP-ribosyltransferase PARP7 as a critical regulator of vascular remodeling, with elevated expression in VSMCs. Parp7 deficiency attenuates angiotensin II-induced remodeling, while VSMC-specific overexpression

Glutaminase 1-mediated glutaminolysis regulates tuberculosis progression by modulating Th1 and Th17 immune responses.

INTRODUCTION: Mycobacterium tuberculosis-mediated tuberculosis (TB) is an infectious disease that results in approximately 1.2 million deaths annually. Glutaminase (GLS1) is a metabolic enzyme involved in glutaminolysis. The current study examined the roles and mechanisms of GLS1 in TB progression. MATERIAL AND METHODS: Peripheral blood mononuclear cells (PBMCs) were extracted from TB patients and healthy individuals, and CD4+ T and CD14+ monocytes were sorted. PBMCs were incubated with M. tuberculosis strain H37Rv lysate to stimulate immune responses and treated with BPTES, a GLS1 inhibitor.

Research progress on resource distribution, extraction technology, anti-inflammatory mechanisms, and structure-activity relationships of natural anti-inflammatory pectins.

Inflammation is a fundamental protective response, yet its chronic form drives many non-communicable diseases. Conventional anti-inflammatory drugs carry significant side effects, prompting interest in natural alternatives such as pectin polysaccharides (PPs), which are generally recognized as safe, structurally diverse, and anti-inflammatory in preclinical models. This narrative review offers a comprehensive overview of anti-inflammatory PPs, from sources to functions. We discuss conventional and emerging green methods and their effects on yield, structural integrity, and bioactivity. Anti-in

Identificaton of novel Cystoisospora suis enolase 2 interaction proteins in IPEC-J2 cells using Co-IP/LC-MS.

Enolase 2 (ENO2) is a crucial an important glycolytic enzyme involved in glycolysis and is also recognised as a multifunctional protein. However, its interacting proteins in intestinal porcine epithelial cells (IPEC-J2 cells) remain unclear. This study investigates the interacting proteins of Cystoisospora suis (Biester et Murray, 1934) ENO2 (CsENO2) in IPEC-J2 cells. According to our previous research, CsENO2 is highly expressed in sporulated oocysts. Co-immunoprecipitation/liquid chromatography-mass spectrometry (Co-IP/LC-MS) was conducted using the pcDNA3.1-Flag vector carrying the enolase

Sulfated chitooligosaccharide selenium alleviates deoxynivalenol-induced intestinal injury in neonatal chicken through modulation of ULK1/LC3-mediated autophagy.

Deoxynivalenol (DON) disrupts intestinal epithelial integrity through oxidative stress and dysregulated autophagy. This study evaluates the efficacy of sulfated chitooligosaccharide selenium (S-COS-Se) in protecting chicken embryos and intestinal epithelial cells from DON-induced intestinal injury. Five hundred 9-day-old embryonated eggs were allocated into 10 groups: Control, Na₂SeO₃, chitosan sulfate, chitosan-selenium, S-COS-Se, DON, and 4 combination groups. Additionally, intestinal epithelial cells were treated with DON alone or combined with various selenium formulations for 24 h. S-COS-

Fractal kinetics unveils the dual regulatory mechanism of sophorolipid in overcoming spatial heterogeneity during lignocellulose hydrolysis.

Fractal kinetics was employed to elucidate how sophorolipid enhances enzymatic hydrolysis of lignocellulosic substrates. Graded bagasse substrates with varying lignin and accessible surface areas were prepared by controlled alkaline hydrogen peroxide pretreatment. Non‑productive cellulase adsorption onto lignin was quantified by Langmuir isotherms, and accessible surface areas of cellulose and lignin were separately measured by dye adsorption. Sophorolipid reduced the Langmuir adsorption constant by 73.8-85.7 % and binding affinity by 77.8-88.3 %, with reductions positively correlated to ligni
