Comparative transcriptomic and co-expression analyses enable the discovery of key enzymes responsible for oleuropein biosynthesis in olive (Olea europaea).

Olive (Olea europaea) is one of the most important crop trees, with olive oil being a key ingredient of the Mediterranean diet. Oleuropein, an oleoside-type secoiridoid, is the major determinant of olive oil flavor and quality. Iridoid biosynthesis has been elucidated in Catharanthus roseus, which produces secologanin-type secoiridoids, but iridoid biosynthesis in other species remains unresolved. In this work, we sequenced RNA from the fruit mesocarp of six commercial olive cultivars with various oleuropein contents during maturation and ripening. Using these data, we discovered three polyphe

Blood Pressure Polygenic Score Predicts Long-Term Blood Pressure Control and Treatment-Resistant Hypertension.

BACKGROUND: Suboptimal blood pressure (BP) control remains a major cardiovascular disease risk factor. Whether genetically predicted BP independently predicts long-term BP control is unknown. We examined the associations of BP polygenic scores (PGSs) with long-term BP control and treatment-resistant hypertension. METHODS: We identified 22 456 Mass General Brigham Biobank participants with hypertension. Longitudinal BP control was defined as the percentage of time above-target systolic BP (SBP) ≥130 mm Hg or diastolic BP (DBP) ≥80 mm Hg over 5 years. Using multivariable regression, we assessed

Unlocking neuronal health: leveraging C. elegans for drug repurposing studies.

Aging affects neuronal function, leading to both structural and functional changes in the human brain. The concomitant cognitive decline and neurodegenerative diseases drive efforts to find pharmaceutical drugs to ameliorate the consequences of neuron-associated deficits during aging. C. elegans is an excellent model to combine studies of the nervous system and drug screening. C. elegans has a well-characterized and relatively simple nervous system that allows for precise mapping of neuronal circuits. Neuronal activity plays a crucial role in regulating lifespan in C. elegans. Various genetic,

Resistant Hypertension in Adults: Evaluation and Treatment.

Difficult-to-control hypertension is a common issue in primary care. Resistant hypertension is defined as a blood pressure (BP) measurement above goal despite the use of maximum or optimal dosages of three antihypertensive agents, including a diuretic. Before diagnosing resistant hypertension, family physicians should evaluate patients with difficult-to-control BP for comorbid conditions, medication nonadherence, white coat hypertension, secondary hyper-tension, and suboptimal therapy. Attention should be focused on ensuring accurate BP measurement technique in the office and confirmatory BP m

Dual role of ACE2 in regulating inflammation triggered by Omicron S1 and other SARS-CoV-2 Spike variants.

Since the emergence of SARS-CoV-2 in late 2019, substantial efforts have been made to understand its mechanisms of pathogenicity. Although angiotensin-converting enzyme 2 (ACE2) has been identified as the main receptor for viral entry, the complexity of the host immune response to different Spike protein conformations and variants remains poorly understood. Using zebrafish larvae as an in vivo model, we show that the monomeric S1 domain of the Omicron variant triggers a potent proinflammatory response characterized by elevated Nfkb activity and increased expression of key cytokines, despite re

Time-resolved proteomic and phosphoproteomic profiling of Angiotensin-(1-7) signaling in A549 cells.

Angiotensin-(1-7) [Ang-(1-7)] is a heptapeptide of the renin-angiotensin system (RAS) with antitumoral effects reported in various tumoral cell lines, including the human lung adenocarcinoma A549 lineage. While previous studies have shown that Ang-(1-7) modulates MAPK and PI3K-AKT signaling, the precise molecular mechanisms involved remain incompletely understood. To investigate the signaling events of Ang-(1-7) in lung cancer-derived cells, we employed an integrated proteomic and phosphoproteomic approach in A549 cells. We analyzed early (minutes) and late (hours) molecular responses to Ang-(

Early blood pressure management in acute ischaemic stroke before and after reperfusion treatment.

Patients with acute ischaemic stroke often present with elevated blood pressure. Evidence has emerged that challenges the longstanding recommendation across clinical guidelines that blood pressure should be controlled to a systolic pressure of less than 185 mm Hg before initiating reperfusion therapy in these patients. RECENT: developments Major clinical trials (INTERACT4, MR-ASAP, and RIGHT-2) and an observational study (TRUTH) indicate that intensively lowering blood pressure before initiating reperfusion therapy for acute ischaemic stroke is associated with increased risk of death and disab

Involvement of Adenosine A2A Receptors in Anxiety-Like Behaviors in Tetrahydrocannabinol-Treated Mice.

BACKGROUND: Previous studies have suggested that adenosinergic system in the central nervous system may play a role in both behavioral changes and the physiopathology induced by Δ9-tetrahydrocannabinol (THC), and this is thought to be mediated by adenosine A2A receptors (A2ARs). However, the contribution of the adenosinergic system to the anxiety-like behaviors in response to THC in mice is not well understood. AIMS: In this study, we aimed to investigate the possible role of the adenosinergic system in THC-treated mice. METHODS: For that purpose, we combined behavioral tests and molecular a
