Biomechanical Characterization of Human Pluripotent Stem Cell-Derived Cardiomyocytes by Use of Atomic Force Microscopy.

Atomic force microscopy (AFM) is not only a high-resolution imaging technique but also a sensitive tool able to study biomechanical properties of biosamples (biomolecules, cells) in native conditions-i.e., in buffered solutions (culturing media) and stable temperature (mostly 37°C). Micromechanical transducers (cantilevers) are often used to map surface stiffness geometry, elastic, and contact parameters of living cells; however, they can also be used to monitor time course of dynamic forces (beating), their rate, and other biomechanical properties of cardiomyocyte contractility. Here we descr

Current approaches to acute bronchiolitis management among pediatricians: guideline adherence and practice variation.

BACKGROUND: Although evidence-based clinical guidelines recommend supportive care as the mainstay of acute bronchiolitis management, significant variability in diagnostic and therapeutic practices among pediatricians persists. Identifying factors associated with guideline adherence may help reduce the gap between evidence-based recommendations and real-world practice. METHODS: This cross-sectional study was conducted between January and December 2023 using a structured, web-based questionnaire administered to pediatricians working in different clinical settings. Data were collected on demogra

Protistan grazer-mediated regulation of domoic acid: interactions between Noctiluca scintillans and Pseudo-nitzschia reshape toxin fate and risk in coastal waters.

Domoic acid (DA)-producing Pseudo-nitzschia and the heterotrophic dinoflagellate Noctiluca scintillans frequently co-occur during coastal harmful algal blooms, yet their interactions and consequences for toxin cycling remain poorly understood. Here, controlled co-cultures were conducted to examine grazer-induced defense, grazing dynamics, and DA accumulation-depuration. Sustained 3-d exposure stimulated a 3-4-fold increase in cellular DA production, without detectable changes in cell size or frustule silicification, indicating a primarily chemical defense response. Cellular DA induction exceed

Anesthetic Management of a Patient With Myasthenia Gravis Undergoing Thoracoscopic Thymectomy: A Case Report and Literature Review.

Myasthenia gravis is an autoimmune neuromuscular disorder characterized by fluctuating skeletal muscle weakness and fatigability. Perioperative management is challenging because patients may have bulbar or respiratory muscle involvement and altered sensitivity to neuromuscular blocking agents, increasing the risk of postoperative respiratory failure and myasthenic crisis. Thymectomy is an established therapeutic option in selected patients with myasthenia gravis and may be performed using minimally invasive thoracoscopic techniques. We report the anesthetic management of a 48-year-old man with

Half-organic substitution combined with dicyandiamide (DCD) reduces N2O and NO emissions from tea plantations: a field study and global assessment.

Acidic tea plantation soils with high nitrogen (N) inputs are recognized hotspots for nitrous oxide (N2O) and nitric oxide (NO) emissions. However, field evidence on the combined effects of organic manure and a nitrification inhibitor (dicyandiamide, DCD) on both gases remains scarce, and previous global budgets have considered N2O only, leaving combined NO + N2O unquantified. Here, a replicated field experiment using static closed chambers was conducted in a subtropical tea plantation. Seven treatments were established: urea (CN), composted chicken manure (OM), and their 1:1 combination (HOM)

Mechanistic Prediction of Maternal, Fetal, and Amniotic Fluid Betamethasone Concentrations to Support Dosing Optimization in Pregnancy.

Respiratory distress syndrome (RDS) remains a major cause of neonatal mortality following premature birth. Antenatal betamethasone is the standard of care to accelerate fetal lung maturation and improve neonatal outcomes; however, current dosing recommendations assume uniform maternal and fetal exposure across pregnancies. Gestational age (GA)-dependent physiological changes and maternal obesity may alter betamethasone disposition and fetal exposure. This study developed a maternal-fetal physiologically based pharmacokinetic (m-f-PBPK) model to quantify the effects of GA and maternal obesity o

Exogenous nitric oxide as an anti-infective agent.

Both before and after the discovery that the high-output isoform of nitric oxide (NO) synthase (iNOS; NOS2) contributes to control of many infectious diseases, investigators have been treating infections with exogenous NO or NO-related moieties. Here, we review the anti-infective potential of topically applied NO-releasing materials, systemically administered anti-infective drugs that generate NO-related moieties, and inhalation of NO gas. Evidence for the safety and efficacy of inhaled NO (iNO) at higher doses than typically used in cardiopulmonary medicine encouraged us to discuss prospects

γ-aminobutyric acid is associated with calcium, reactive oxygen species, and nitric oxide responses and enhanced cadmium tolerance in Salvadora persica.

BACKGROUND: γ-Aminobutyric acid (GABA) accumulates under stress, but its association with calcium (Ca2+), reactive oxygen species (ROS), nitric oxide (NO), and thiol responses during cadmium (Cd) exposure remains unclear in medicinal shrubs. METHODS: Rooted Salvadora persica L. cuttings were grown in sand-soil substrate. Screening selected 50mg Cd kg-1 substrate and 0.5  mM root-applied GABA. Eleven treatments had five independent pots each. Ca2+-sensitive Fluo-3 AM and NO-associated DAF-FM DA fluorescence and H₂O₂ were assessed in separate, destructively sampled plants at five post-applicati

Combining Pharmacological Therapy and Ultrasound-Guided Botulinum Toxin Injection Resulted in Clinical Improvement in Bilateral Arterial-Neurogenic Thoracic Outlet Syndrome: A Case Report.

BACKGROUND: Thoracic outlet syndrome (TOS) results from compression of the brachial plexus and/or the subclavian vessels. Botulinum toxin (BTX) has been shown to be useful in TOS both as a predictor of surgical outcome and as a therapeutic option. CASE REPORT: We present the case of a 21-year-old woman with a rare form of bilateral TOS with combined arterial and neurogenic involvement. After surgical treatment on the right side, the patient developed symptoms on the left side. We adopted a multimodal therapeutic approach that effectively reduced her symptoms and improved her quality of life.

Oculopharyngeal Muscular Dystrophy: From Molecular Pathogenesis to Gene Therapy.

Oculopharyngeal muscular dystrophy (OPMD) is a rare, late-onset, autosomal dominant muscular dystrophy caused by a trinucleotide GCN repeat expansion in the PABPN1 gene on chromosome 14q11.2. The disease is clinically characterised by progressive ptosis, dysphagia, and proximal limb weakness, with onset typically between the fifth and sixth decades. OPMD affects approximately 1:100,000 individuals in Europe, with substantially higher prevalence in specific founder populations, reaching 1:600 among Bukharan Jews and 1:1,000 in French Canadians. The expanded polyalanine tract in PABPN1 promotes
