Targeting with toxins: an overview of venom peptides in drug delivery.

Due to the numerous issues associated with many current drug therapies, including lack of efficiency and off-target toxicity, targeted drug delivery has become a critical area of pharmaceutical research. Natural compounds show great promise for active drug targeting, since many of them possess distinct activity towards biomolecules in human body. That includes peptide toxins derived from venoms of various animals, which have often been tailored by evolution to target biomolecules in living systems with high potency and specificity. In this review, we discuss the current research on the use of

Hydrochalcogenation Reactions with Noncanonical Amino Acids as a Route to Increase Bioconjugate Valency.

Bioconjugation represents an efficient and rapid mechanism to enhance the already extensive utility of proteins; however, it requires the development of additional chemistries to prevent undesired reactivity with nascent biological functionalities. Moreover, the ability to conjugate multiple partners to increase the conjugate valency further expands potential applications. Herein, we describe the development and optimization of a thio-yne bioconjugation using rongalite coupled with noncanonical amino acids (ncAAs) to afford multivalent conjugates. The site-specificity of the ncAA partner provi

On the Photophysics and Contrasting Cavitand Binding of a Fluorescent Amphiphilic Benzothiazole.

Benzothiazoles, including the cationic thioflavin T and its variants, are of immense interest because of their therapeutic potential and applications as biomolecular fluorescence probes. This study investigates the photophysics of an interesting benzothiazole derivative, TEG-BTA-2, that combines a neutral thioflavin T type of moiety (BTA-2) with a tetraethylene glycol chain (TEG). Unlike thioflavin T, TEG-BTA-2 is highly fluorescent and shows multiple prototropic transformations. Monoprotonated TEG-BTA-2 is found to exist in two isomeric forms, each having characteristic absorption, emission,

Peptide interaction with mixed lipid bilayers alters packing and hydrocarbon chain conformations.

Cell-penetrating peptides (CPPs) with a cationic-hydrophobic character are recognized as carriers for delivering various therapeutics and diagnostic agents across cell membranes and into the cells. Among the most studied CPPs, nona-arginine (R9) exhibits superior penetration compared to its equally charged counterpart, nona-lysine (K9). This indicates that penetration ability relies not only on charge but also on the structure, distribution, and concentration of peptides, as well as the composition of lipid membranes. However, interactions of heptapeptides composed of arginine (R), lysine (K),

Small Molecule Heterocycles: Design and Synthesis of Novel Fluorescent Benzothiazoles for Early Detection of Alzheimer's disease.

Diagnostic agents capable of imaging β-amyloid (Aβ) plaques at prodromal stages could provide noninvasive tools to assess plaque burden and assist in determining efficacy of disease-modifying therapeutic interventions in vivo. To accomplish this goal, FDA approved 18F-incorporated tracers, such as Florbetapir, Flutemetamol, and Florbetaben may allow quantitative analysis of therapeutic efficacy of disease-modifying treatments that attenuate Aβ pathophysiology in vivo. Although promising, these tracers show high off-target binding. Importantly, current anti-amyloid therapy also targets diffuse

The barrier and protective functions of intestinal mucin in defense against Candida albicans.

Candida albicans (C. albicans) is an opportunistic fungal pathogen that typically colonizes intestinal mucosal surfaces asymptomatically. However, dysbiosis or disruption of the mucosal barrier can trigger its overgrowth, leading to mucosal infections and, in severe cases, systemic disease. Intestinal mucins, the main components of mucus, play critical roles in host defense by suppressing filamentation-associated gene expression, blocking the yeast-to-hypha transition, and inhibiting key virulence traits including adhesion, biofilm formation, and secretion of hydrolytic enzymes. While their fu

Osmolarity-Directed Encapsulation of Size-Tuned Nanoparticles into Red Blood Cells.

Red blood cells (RBCs) have emerged as promising carriers for therapeutic and diagnostic agents due to their long circulation time, biocompatibility, and immune-evasive properties. Hypotonic encapsulation is the most widely employed technique; however, the correlation between nanoparticle sizes and the hypotonic osmolarities required for efficient encapsulation remains unclear. In this study, the size-dependent osmolarity requirements governing nanoparticles into RBCs are investigated. Using monodisperse gold nanoparticles as model systems, the optimal osmolarity correlated with nanoparticle d

Development and characterisation of a thermoreversible sol-gel for intranasal delivery of MRI and PET imaging agents.

Central nervous system disorders remain particularly difficult to manage due to the blood-brain barrier preventing effective uptake and distribution of most therapeutic and diagnostic agents to brain parenchyma. Intranasal delivery using thermoreversible sol-gels is a non-invasive strategy proposed to bypass the blood-brain barrier, enhance mucosal residence, and enable sustained release of an agent. This study investigated poloxamer-based sol-gels for the intranasal delivery of Gd-DTPA for MRI and [64Cu]Cu2+ for PET. Formulations containing a blend of poloxamers and hydroxypropyl methylcellul

Harnessing Insights of Hepatocyte Growth Factor Kringle 3 Domain to Develop c-Met Targeted Positron Emission Tomography Probe.

The cellular mesenchymal-epithelial transition factor (c-Met) is overexpressed in multiple solid tumors and is normally driven by its native ligand, hepatocyte growth factor (HGF). Despite extensive structural studies, no diagnostic agents have been developed based on the individual HGF-Kringle 3 (K3) domain. Here, four HGF-K3-derived c-Met-targeted peptide radioligands were designed for positron emission tomography (PET) imaging. Among them, [68Ga]Ga-SMIC-1014 exhibited the most favorable pharmacokinetics and achieved different tumor uptake in HCT-116, HepG2, and LNCaP xenografts. Moreover, t
