Effects of air pollution on disease activity in patients with rheumatoid arthritis.

OBJECTIVES: This study aimed to evaluate the associations of air pollution with disease activity and flare rate in patients with rheumatoid arthritis (RA). METHODS: This prospective cohort study included patients with RA who were treated at a tertiary medical centre in South Korea between January 2021 and December 2024. Air pollution exposure was estimated using monthly mean concentrations of 6 air pollutants (sulfur dioxide, nitrogen dioxide, ozone, carbon monoxide, particulate matter [PM]10, and PM2.5). Disease activity and flares were recorded longitudinally at each outpatient visit. Assoc

Cryopreservation Method for Aurantiochytrium limacinum.

Aurantiochytrium limacinum is a marine protist that produces high levels of omega-3 fatty acid DHA and is thus of biotechnological importance. Here, we describe a cryopreservation technique to preserve A. limacinum at two different temperatures (-80°C) and liquid nitrogen (-196°C). At -80°C, cells were preserved in 25% (v/v) glycerol; at -196°C (liquid nitrogen), in 10% (v/v) methanol, 15% (v/v) glycerol, or 15% (v/v) dimethyl sulfoxide. The cells were stable for up to 12 months of storage. The method preserves the genetic integrity and functionality of the cells for biotechnological applicati

Tissue Preservation and FFPE Samples: Optimized Nucleic Acids Isolation in Ewing Sarcoma.

Different methods have been described for the preservation of biopsy or resection samples. In routine pathology, the cheapest and most used method is fixation of samples in formalin and embedding in paraffin (FFPE samples). This method preserves tissue samples for a very long time and is suitable for several specialized techniques, such as fluorescence in situ hybridization (FISH) and immunohistochemistry, the latter being the most frequent and often the only additional method used for the establishment of the final diagnosis. However, in light of the growing need for next-generation sequencin

A Drosophila Study Model for EWS::FLI1-Induced Molecular Pathways.

Ewing sarcoma (EwS) is an aggressive, human-exclusive tumor typically driven by the EWS::FLI1 fusion protein. The paucity of genetically modified animal models hinders in vivo functional studies of this oncogene. Here, we describe the generation of transgenic Drosophila lines expressing different levels of human EWS::FLI1 and fluorescent reporters of conversion of quiescent GGAA-repeat microsatellites (GGAAμSats) into neoenhancers. We also describe experimental setups to test EWS::FLI1 activity, and to carry out genetic and chemical screens. Altogether, the collection of transgenic Drosophila

Functionalization of AFM Tips and Supports for Molecular Recognition Force Spectroscopy and Recognition Imaging.

Linking sensor molecules (e.g., antibodies) to an AFM tip converts it into a biosensor by which single target molecules (e.g., antigens) can be detected and localized on the sample surface. Moreover, the mechanism of interaction can be studied by force spectroscopy if purified target molecules are linked to an ultra-flat surface, such as mica or silicon (nitride). Rapid imaging of the binding sites and force spectroscopy studies are greatly facilitated if 6-10 nm long polyethylene glycol (PEG) chains are used as flexible tethers between the sensor molecule and the tip. Here, we describe a set

Biochemical Identification of Salmonella Species: Classical Tools for Accurate Diagnosis.

Biochemical testing plays a crucial role in the detection and confirmation of Salmonella spp. Since the cultural characteristics of Salmonella often resemble those of other enteric pathogens, differentiation based solely on growth features can be challenging. Biochemical tests allow for accurate identification by exploiting the distinct metabolic traits of Salmonella. In this study, we described three key tests, including methyl red, hydrogen sulfide production, and urease production, which are particularly important for the confirmation of Salmonella.

Protocol for the Extraction of High-Quality DNA from Botrytis cinerea.

Using Botrytis cinerea (ATCC 58025) as the material, genomic DNA was extracted using the SDS-CTAB method after liquid nitrogen freezing and extracted multiple times with organic reagents. Then the precipitate was washed with ethanol and dissolved in TE buffer. The cultured genomic DNA is high-yield, easy to extract, and has good purity, making it suitable for extracting from multiple samples at once. After experiments, the genomic DNA obtained by this method can be used for most basic molecular biology experiments such as PCR and DNA enzyme digestion.

Preparation of Postmortem Human Formalin-Fixed Paraffin-Embedded Frontal Cortex Tissue for Profiling Pyramidal Neurons Via Digital Spatial Profiling.

Digital spatial profiling (DSP) facilitates RNA sequencing (RNA-seq) of small populations of cells, specifically ~20 pyramidal neurons in postmortem formalin-fixed paraffin-embedded (FFPE) human brain tissue. This approach enables precise expression profiling via RNA-seq data linked to spatially characterized samples utilizing tissue-bound probes. We present a comprehensive protocol for manual slide preparation using the GeoMx DSP system as preliminary steps to spatial characterization of lamina-specific pyramidal neurons in human postmortem Brodmann area 9 (BA9) frontal cortex brain tissue.

Cell-Based Multisensor Array for Vapor-Phase Detection of Cancer-Related Compounds in Human Urine.

Detecting volatile biomarker candidates in complex biological samples is a key challenge in sensor science, particularly for noninvasive disease screening applications. Biohybrid sensors based on olfactory receptor-expressing cells offer high molecular selectivity; however, their use with physiological samples is often limited by weak fluorescence signals and variability in cellular responses. Here, we present a cell-based multisensor platform in which insect olfactory receptor-expressing cells are encapsulated in hydrogel and immobilized within slit-integrated microwells. This microstructural

Enhancing the extraction efficiency, functional activity, and freeze-stability of milk-derived extracellular vesicles via rennet coagulation combined with polyethylene glycol precipitation.

Milk-derived extracellular vesicles (MEV) hold considerable promise for immune regulation and nutrient delivery; however, their scalable production and long-term storage stability present major challenges. This study compared 2 whey pretreatments-rennet (EC 3.4.23.4) coagulation and acetic acid precipitation-combined with polyethylene glycol (PEG) precipitation or ultracentrifugation (UC) for MEV isolation. The yield, physicochemical properties, and storage stability of the extracted MEV were systematically evaluated. Results indicated that the PEG-rennet-coagulated (R) method demonstrated opt

ROS-Responsive ECM Mimetic Hyaluronic Acid-PEGDA Hydrogel Enables Palmitoleic Acid-Mediated Suppression of Oxidative Stress and Atherosclerosis via AMPK/NF-κB/NLRP3 Signalling.

Oxidative stress and inflammasome-driven inflammation are key contributors to atherosclerotic disease progression. Here, we report a ROS-responsive hyaluronic acid-PEGDA hydrogel designed for the sustained delivery of palmitoleic acid. In the study, catechol-functionalized hyaluronic acid was cross-linked with polyethylene glycol diacrylate to create an extracellular matrix mimic that possessed tunable mechanical strength and redox-sensitive degradability. It showed controlled release of palmitoleic acid under normal physiological conditions and a faster rate of release under oxidative conditi

Survival Outcomes Associated With Short-Acting Versus Long-Acting G-CSF Use During First-Line Chemoimmunotherapy for Advanced Lung Cancer: A Retrospective Study.

While granulocyte colony-stimulating factor (G-CSF) is used to prevent and treat chemotherapy-induced neutropenia, it also regulates key immune cells and may therefore affect the efficacy of immunotherapy. Short-acting and long-acting G-CSF have similar efficacy in managing neutropenia, but they differ in effects on immune cells. However, the overall impact of G-CSF and its different formulations on survival in advanced lung cancer patients receiving chemoimmunotherapy remains unclear. This retrospective study enrolled patients with advanced primary lung cancer receiving first-line chemoimmuno

Presynaptic Optical Modulation Enabled by Dielectric-Embedded Type-II PbS/PbSe Quantum Dots for Near-Infrared Neuromorphic Vision.

In nighttime perception and near-infrared (NIR) neuromorphic vision, a central challenge lies in encoding weak optical inputs into stable and controllable temporal states at the sensor front end. In nocturnal mammalian retinas, AII amacrine cells achieve this function by relying on stable chemical intermediate states, presynaptic modulation, and subthreshold temporal integration. Functionally inspired by selected information-processing principles of this mechanism, we propose and demonstrate a bio-inspired device design strategy. Through a chemical-structural codesign approach, type-II PbS/PbS

Spatially Refined Analysis of Emission Burdens and Equity from Light-, Medium-, and Heavy-Duty Vehicle Traffic in the United States.

We estimate block-level emission intensities of nitrogen oxides (NOX) and fine particulate matter (PM2.5) from on-road vehicle traffic for every census block in the United States, combining MOVES-based emission factors with street level traffic data. We use this spatially refined, computationally efficient, and policy-sensitive metric to evaluate emissions burden. Despite comprising less than 11% of vehicle kilometers traveled, we find that medium- and heavy-duty trucks account for 46% of NOX and 50% of PM2.5 emission burden nationally ─ with PM2.5 burden from trucks dominated by exhaust emiss

Metal-Induced Localized Microcrystals Buffer Poisoning in NOx Catalytic Reduction.

Metal impurities in flue gas severely threaten the service life of NOx abatement catalysts. While traditional acid-base interaction strategies rely on sacrificial sites, such passive defense mechanisms merely mitigate activity loss rather than fundamentally resolving poisoning. This study establishes an active defense mechanism by fabricating amorphous high-entropy metal oxides capable of sequestering metal poisons by inducing localized microcrystallization. These resulting microcrystals are subsequently transformed into extra active centers for NOx reduction. Systematic investigation reveals

A Formaldehyde-Free, Soy Protein-Based Adhesive with P-N-Si Synergistic Flame Retardancy for Plywood Application.

To overcome the flammability and formaldehyde-emission problems associated with plywood, a novel soy protein-based intumescent flame-retardant adhesive with phosphorus-nitrogen-silicon (P-N-Si) synergism (C@Si@APP) was developed in this study. C@Si@APP was initially prepared by coating silane-modified ammonium polyphosphate with collagen. Subsequently, C@Si@APP was combined with lignin and adenine and incorporated into a soy protein, collagen, and isocyanate (MDI)-based adhesive matrix to fabricate a flame-retardant adhesive for eucalyptus plywood. Cone calorimeter (CONE) analysis demonstrated

Evolutionary Inferences Based on Thoracic Limb Muscles of South American Coati (Nasua nasua): Brachium and Craniolateral Antebrachium.

The South American coati (Nasua nasua) is a mammal of the order Carnivora and family Procyonidae, characterized by scansorial locomotion. Accordingly, several anatomical and functional studies examined its thoracic limbs. Nevertheless, the anatomical descriptions and terminology differ among those studies, and none considered the evolutionary derivations of the brachium and antebrachium muscles based on topology, innervation and anatomical variants. Therefore, the present study aimed to infer the muscle derivations and reconstruct muscle maps of the brachial and craniolateral antebrachial musc

Simultaneous valorization of nitrate and formaldehyde waste streams to hexamethylenetetramine via photo-electro-thermo-catalysis.

To remediate nitrate (NO3-) pollution and recover resources from formaldehyde wastewater, this work presents a waste valorization strategy integrating photoelectrocatalytic nitrate reduction (PEC NO3RR) to ammonia (NH3) with downstream chemical upgrading. Herein, a hollow MoO2@CuFe2O4@MnFe2O4 Z-scheme-Schottky dual heterojunction nanoreactor array is developed to spatially separate proton and nitrate adsorption sites. The MoO2@CuFe2O4@MnFe2O4 improves photogenerated charge separation and retaining robust redox capability, while the oxygen vacancy-rich MoO2 layer exhibits favorable photothermal

In Vivo Chemical Cross-Linking Coupled with Reversed-Phase Co-Fractionation Mass Spectrometry Enables Protein Complex Analysis.

Understanding protein-protein interactions (PPIs) and the architecture of protein complexes is essential for elucidating cellular functions and drug action mechanisms. Co-fractionation mass spectrometry (CF-MS) has emerged as an effective tool for analyzing protein complexes under near-native conditions, however, its resolution and structural fidelity are often compromised by complex dissociation during biochemical processing. Here, we present an analytical strategy that integrates in vivo formaldehyde cross-linking with reversed-phase cofractionation mass spectrometry (XL-RP-CF-MS) to identif
