Aurantiochytrium limacinum is a marine protist that produces high levels of omega-3 fatty acid DHA and is thus of biotechnological importance. Here, we describe a cryopreservation technique to preserve A. limacinum at two different temperatures (-80°C) and liquid nitrogen (-196°C). At -80°C, cells were preserved in 25% (v/v) glycerol; at -196°C (liquid nitrogen), in 10% (v/v) methanol, 15% (v/v) glycerol, or 15% (v/v) dimethyl sulfoxide. The cells were stable for up to 12 months of storage. The method preserves the genetic integrity and functionality of the cells for biotechnological applicati

Using Botrytis cinerea (ATCC 58025) as the material, genomic DNA was extracted using the SDS-CTAB method after liquid nitrogen freezing and extracted multiple times with organic reagents. Then the precipitate was washed with ethanol and dissolved in TE buffer. The cultured genomic DNA is high-yield, easy to extract, and has good purity, making it suitable for extracting from multiple samples at once. After experiments, the genomic DNA obtained by this method can be used for most basic molecular biology experiments such as PCR and DNA enzyme digestion.

Photoactive constituents in medicinal and edible plants can induce photooxidative reactions and pose potential safety risks, yet their rapid identification remains challenging. Herein, a field-induced droplet ionization mass spectrometry (FIDI-MS)-based strategy was developed to evaluate the photooxidative activity of Angelica dahurica. Ethanol extracts exhibited stronger photooxidative activity than aqueous extracts owing to their higher contents of lipophilic furanocoumarins. Six furanocoumarins, including imperatorin, isoimperatorin, psoralen, bergapten, byakangelicol, and phellopterin, wer

Chlorogenic acid (CA), the principal bioactive constituent of honeysuckle, is conventionally extracted by time-consuming methods using toxic organic solvents, which carry the risk of thermal degradation. This study provides a rapid and efficient microwave-assisted deep eutectic solvent (DES-MAE) method for CA extraction and elucidates its mechanism at the molecular level. Eight DESs were systematically screened, and extraction parameters were optimized via single-factor experiments and response surface methodology (RSM). Density functional theory (DFT) calculations were performed to reveal the

Granular cold-water-swelling oat starch was prepared under both low-temperature (90-130 °C) and high-temperature (150-190 °C) conditions using subcritical ethanol-water treatment, and the resulting structural and property changes were systematically investigated. The low-temperature treatment largely retained the native orthorhombic crystal structure and produced granules with finer morphology, whereas the high-temperature treatment induced crystallization of a new orthorhombic amylose phase and generated a swollen granular state with superior cold-water swelling capacity (11.60 g/g vs. 1.99 g

Coffee cherry pulp (CCP), a major by-product and solid waste of coffee processing, is rich in phenolic compounds and pectin but remains underutilized. This study evaluated supercritical CO2 extraction with ethanol co-solvent (SFE-CO2) as a green alternative for recovering bioactive compounds from CCP and investigated the subsequent recovery of pectin-rich fractions and pectic oligosaccharides (POS). Using a one-factor-at-a-time (OFAT) approach, the selected SFE-CO2 conditions were 50% (w/w) ethanol, 55 °C, and 300 bar. Under these conditions, the SFE-CO2 extract exhibited total phenolic (4.26

Brewer's spent grain (BSG), the major by-product of the brewing industry, is an abundant lignocellulosic biomass rich in phenolic compounds, although their association with the cell wall matrix limits their recovery and functional application. This study aimed to enhance phenolic compound recovery from BSG by integrating solid-state fermentation (SSF) with Aspergillus oryzae and green extraction technologies and to stabilize the recovered compounds using nanostructured lipid carriers (NLCs). Fermentation was evaluated for up to 7 days. Fermented and unfermented BSG were subsequently subjected

Three-dimensional (3D)-printed permanent crown resins are increasingly used in restorative dentistry, but their residual monomer elution compared with computer-aided design/computer-aided manufacturing (CAD/CAM) materials remains insufficiently characterized. This in vitro study compared residual monomer elution from three permanent crown materials at different storage times. A CAD/CAM hybrid ceramic (GC Cerasmart 270) and two 3D-printed permanent crown resins (Graphy Tera Harz and Saremco CrownTec) were evaluated. Specimens (12 × 14 × 2 mm; n = 5 per material) were stored in 75% ethanol, and

In situ assimilation of off-gases (H2/CO2) into liquid metabolites during anaerobic fermentation presents a promising approach to maximize waste biomass valorization. However, mechanisms governing the interaction between microbial carbon fixation and chain elongation (CE) through headspace dynamics remain unclear. This study explored gas-liquid metabolic flux and microbial response by varying headspace pressure/composition in batch CE bioreactors. Autogenic pressure reshaped the physicochemical microenvironment, which potentially induced specific biological inhibition and constrained ethanol u

To isolate and characterize aroma-producing microorganisms from tobacco and evaluate their fermentation potential, 12 strains obtained from tobacco leaves and root soil were identified as members of the genera Enterobacter, Burkholderia, and Pseudomonas. High amylase activity was detected in most Enterobacter and Pseudomonas strains, whereas low cellulase activity and limited ethanol tolerance were generally recorded across all isolates. A total of 174 volatile compounds were detected by HS-SPME-GC-MS, among which alcohols were the most abundant class. Seventy-four odor-active compounds were i

Bio-aerogels are a class of ultra-lightweight, extremely porous, solid materials that are engineered from biopolymers, e.g., polysaccharides, proteins, or peptides. Such biomaterials are of interest for medical and environmental applications, such as molecular filtration, wound healing, or tissue engineering. In this context, ultrashort peptides, defined as sequences of up to ten amino acids, are attractive self-assembling building blocks for bio-aerogels, due to their low-cost synthesis, chemical tunability, and potential biocompatibility. Building on our previous report demonstrating the int

BACKGROUND: Alcohol-containing emulsions face severe colloidal instability because ethanol perturbs biopolymer conformation and interfacial organization. To overcome this, we fabricated ethanol-tolerant Pickering emulsions stabilized via the interfacial assembly of propylene glycol alginate (PGA)-modulated lactoferrin/zein (LZ) composite nanoparticles. This study systematically elucidated the role of the oil-phase volume fraction (φoil, 200-700 mL L-1) in regulating the physicochemical properties, rheological behavior, and β-carotene protection of these complex colloidal systems in an aqueous

Granular activated carbon (GAC) is widely used to remove perfluorooctane sulfonate (PFOS) from contaminated water, yet its sustainable regeneration remains challenging. Solvent-based regeneration preserves the carbon matrix but may shift environmental burdens toward solvent production, chemical use, energy demand, and downstream waste management. Here we present a life cycle assessment (LCA) of PFOS-laden GAC regeneration using deionized water (GAC-WR) and ethanol (GAC-ER). We identify electricity-driven elution and downstream incineration as the dominant environmental hotspots across 18 midpo

The rapid anion exchange between chloride (Cl-) and bromide (Br-), accompanied by remarkable photoluminescence (PL) shifts, enables CsPbBr3 perovskites to exhibit excellent selectivity for Cl- detection. However, CsPbBr3 nanocrystals stabilized by organic ligands usually exhibit limited applicability for Cl- detection in water, because the hydrophobic ligands prevent direct contact with aqueous Cl- and restrict interfacial ion-exchange kinetics. Ligand-free CsPbBr3-based perovskites synthesized by solid-state methods generally suffer from poor stability in aqueous environments and weak fluores

Internal states like hunger, pain, thirst, and arousal can bias behavior by affecting sensory and memory processing. Internal states are critical to understand in the context of alcohol addiction because they influence cravings, reinstatement, and relapse. Norepinephrine plays a key role in both hunger and alcohol-induced arousal and preference, but the circuit-level mechanisms through which it modulates the influence of hunger on alcohol preference are not well understood. We sought to address this using intersectional genetic tools for manipulating neurons expressing octopamine, the inverteb

Additive manufacturing of engineered living materials (ELMs) enables the spatial patterning of living cells or organisms within three-dimensional (3D) polymer matrices. However, effective biocontainment, defined as the ability to retain cells within 3D-printed matrices during the application period, remains a key limitation for ELMs requiring physical confinement. While synthetic biology provides cell-specific biocontainment strategies, materials-based approaches can enable generic cell retention capabilities. Therefore, we applied a materials-based strategy that combines pre-printing formulat

Smartphone-based colorimetric sensor arrays using natural dyes are promising for on-site meat freshness assessment, but varying illumination and imaging angles compromise reliability. To address this, we developed an anthocyanin-curcumin array incorporating an α-Fe2O3 internal standard and an internal-standard-guided dynamic feature-space switching (IS-DFSS) strategy and validated its effectiveness under complex imaging conditions using ammonia as the representative analyte because it is the dominant component of total volatile basic nitrogen (TVB-N). Eight colorimetric films and the internal
