Interspecific and intraspecific variation in venom proteomics, composition, and antivenom efficacy of Thai cobras (Naja spp.): Highlighting the distinct profile of the newly identified Naja fuxi.

Cobras (Naja spp.) account for a significant number of snakebite incidents in Thailand. The monocled cobra (Naja kaouthia) has historically been considered the only non-spitting species, but recent evidence indicates population-level diversification in central and southern regions. Additionally, a newly described non-spitting species, the mountain cobra (Naja fuxi), has been identified in mountainous areas. This study investigates venom variation among Thai Naja species and populations and evaluates the efficacy of monovalent and polyvalent Thai antivenoms. Proteomic analyses revealed that thr

Nephroprotective Effects of Wedelolactone against Snake Venom-Induced Acute Kidney Injury: Insights into Experimental Envenomation.

Snakebite-induced acute kidney injury (sAKI) is a severe clinical complication associated with Bothrops envenomation that can lead to kidney failure. Alternative therapies are needed due to the limitations of antivenom, including variable efficacy, risk of adverse reactions, and significant barriers to access, such as long distances, transportation difficulties, and high costs. This study evaluates the nephroprotective effects of wedelolactone (WEL), a bioactive coumestan derived from Eclipta prostrata, known for its anti-inflammatory and antioxidant properties, in a preclinical model of sAKI.

Inadequate immunorecognition and ineffective paraspecific neutralisation of lethality and toxicity of Trimeresurus erythrurus venom by Indian polyvalent antivenom compared to Trimeresurus popeiorum venom.

Snakebite envenomation remains a major public health challenge in India, where most fatalities are caused by the "Big Four" snakes and polyvalent antivenom (PAV) is produced only against their venoms. However, several regionally important species, including Trimeresurus erythrurus (red-tailed pit viper), are medically relevant in Northeast India but remain poorly studied. This study evaluated the cross-reactivity and neutralisation efficiency of Indian commercial PAVs against T. erythrurus venom (TEV). Immunological assays, including ELISA, spectrofluorometry, and Western blotting, demonstrate

Comparative Effects of Snake Envenomation on Rabbit Carcass Decomposition and Insect Succession in a Forensic Context.

BACKGROUND: Snake envenomation represents a significant health concern in some regions of the world, with fatal cases occasionally requiring forensic investigation to estimate the postmortem interval (PMI). However, the influence of venom on carrion decomposition dynamics and arthropod succession patterns remains poorly understood, potentially compromising postmortem interval (PMI) estimations in such cases. OBJECTIVES: This study investigated the effects of Naja haje and Cerastes cerastes venoms on decomposition progression and necrophagous arthropod succession. METHODS: Fifteen rabbits wer

Biochemical characterisation and substrate-specific proteolytic diversity of venom metalloproteinases in African puff adders.

The puff adder (Bitis arietans) is a highly venomous viper responsible for many snakebite fatalities in Africa, yet there have been few geographically comprehensive analyses of its venom proteins, particularly of the proteases that play a key role in pathology of envenoming. To address this, we have isolated, identified and characterised the bioactivity of the venom metalloproteases of puff adders obtained from a range of localities. Prominent in all venoms was a PI snake venom metalloproteinase (SVMP), derived from a larger PII precursor. This protein existed as either non-glycosylated (21 kD

Snake Venom Protease Detection and Inhibition in Serum.

Snake bites remain major threats with only limited diagnostic and therapeutic options. Current antibody-based antivenoms show only limited effectiveness against local tissue damage and require complex manufacturing and cold chain logistics. To overcome these limitations, a fluorescence-based assay was established to enable the sensitive detection of snake venom metalloprotease (SVMP) and serine protease (SVSP) activities in crude viper venoms from Crotalus atrox, Bothrops jararaca, and Echis carinatus. The assays showed species-specific activity profiles with detection limits in the sub-microg

ProVenTL: a transfer-learning framework for predicting peptide-protein interactions derived from snake venom for cancer therapeutics.

Accurate prediction of peptide-protein interactions (PepPI) is crucial for advancing peptide-based anticancer drug design. In this study, we introduce ProVenTL, a computer-aided molecular design framework that leverages transfer learning and protein language model embeddings to enhance PepPI prediction accuracy and interpretability. Two complementary strategies were explored: (i) fine-tuning a CAMP model pretrained on large-scale PepPI data from the Protein Data Bank (PDB) using a curated dataset of Calloselasma rhodostoma venom peptides and cancer-related proteins, and (ii) integrating ProtT5

Crystal structure and functional characterization of an Asp49 phospholipase A2 from the bushmaster (Lachesis muta).

Snake-venom phospholipases A2 (PLA2s) are small, structurally conserved enzymes that contribute significantly to the pathophysiology of envenomation. Here, we report the purification and crystal structure of an Asp49-PLA2 isolated from the venom of Lachesis muta, a pit viper from the Peruvian Amazon. The enzyme was purified using ion-exchange and size-exclusion chromatography and exhibited phospholipase activity in a dose- and time-dependent egg-yolk degradation assay. Pure protein crystals were obtained in space group P6222 and diffracted to 2.36 Å resolution, with two molecules in the asymme

Identification of DC-174 as a Novel Hydroxamic Precandidate for the Development of an Oral Snakebite Treatment.

Snakebite envenoming is a neglected tropical disease that causes high mortality and morbidity. The current treatment, intravenous antivenom, comes with numerous disadvantages, making new therapeutics important. Optimized small molecules offer the possibility for oral use at the onset of envenoming, and the highly pathogenic, zinc-dependent snake venom metalloproteinase toxin family represents an attractive target for drug discovery. Through systematic chemical modification guided by molecular modeling, we describe the development of hydroxamic acid 23 (DC-174), a molecule that displays potent

The action of marimastat, a metalloprotease inhibitor, on the coagulant, hemorrhagic, and neuromuscular activities of Lachesis muta (South American bushmaster) venom.

Snake venom metalloproteases (SVMP) play a major role in various pathophysiological effects of snakebite envenomation, and marimastat (MMT), a selective metalloprotease inhibitor, has been investigated as a potential ancillary treatment for attenuating SVMP-mediated effects. In this work, the effect of MMT on the coagulant, hemorrhagic and neuromuscular activities of Lachesis muta (South American bushmaster) venom was assessed experimentally in vitro and in vivo and compared with therapeutic anti-Bothrops/Lachesis antivenom (AV) produced by the Instituto Butantan. Snake venom-induced tissue damage as a paradigm of redox imbalance, ferroptosis, and inflammaging.

Snakebite envenomation remains a major cause of illness globally, resulting in complex local and systemic toxic effects. Although the biochemical functions of individual snake venom components have been extensively studied, the cellular mechanisms underlying progressive tissue damage and organ dysfunction remain less well understood. This review compiles contemporary mechanistic evidence associating notable venom toxin families, such as phospholipase A₂, snake venom metalloproteinases, and three-finger toxins, with oxidative stress-induced cellular damage. Mounting evidence highlights mitochon

Long-term region-specific mitochondrial respiration impairment after perinatal asphyxia is prevented by the NAD⁺ donor nicotinamide riboside: A real-time organotypic metabolic profiling approach.

Perinatal asphyxia (PA) is a primary cause of neonatal morbidity, with persistent neurological sequelae, associated with mitochondrial dysfunction and redox imbalance. The lack of experimental approaches for region-specific evaluation of mitochondrial bioenergetics in structurally preserved neural tissues has limited the identification of selective vulnerabilities and redox-based therapeutic targets. Here, we introduce a real-time metabolic profiling strategy that integrates Seahorse XFe96 respirometry with Cytation 5 image-based quality control to quantify mitochondrial respiration and glycol

Chinese cobra (Naja atra) SVPLA2 promotes muscle inflammation via RPL35A-mediated translational reprogramming.

Naja atra venom induces severe myotoxicity and inflammation largely driven by snake venom phospholipase A2 (SVPLA2). Although the SVPLA2 inhibitor varespladib shows great therapeutic promise, the underlying molecular mechanisms remain unclear. Herein, we uncover dual pathogenic pathways. First, a translational module in which RPL35A interacts with PTBP1 to selectively enhance IL-6 and TNF-α translation without altering mRNA levels. Second, a transcriptional axis where prostaglandin E2-activated p38 MAPK enhances NF-κB signaling and reactive oxygen species production, forming an inflammatory-ox

Brugada phenocopy induced by Russell's viper envenomation: resolution following anti-snake venom therapy.

Russell's viper envenomation is known to cause complications such as coagulopathy, acute kidney injury and haemolysis. Cardiac involvement, though rare, has been reported in the form of arrhythmias, myocardial infarction and ECG changes such as ST-segment elevation or T-wave inversions. We report a case of a middle-aged farmer who presented to our centre with Russell's viper envenomation, characterised by local envenomation, coagulopathy, haemolysis and ocular manifestations. His ECG showed a Brugada pattern, with no evidence of any structural cardiac abnormalities or elevated cardiac enzymes.

Unveiling the molecular diversity of toxins and non-toxins in the medicinal sea snake Hydrophis cyanocinctus through integrated venomics investigation.

Sea snake venom systems provide exceptional models for studying marine adaptation and are rich sources of bioactive molecules for drug discovery. To comprehensively characterize the venom diversity of the medicinal sea snake Hydrophis cyanocinctus, this study employed an integrated multi-omics strategy. We performed deep proteomic profiling of its venom and venom gland using both data-independent acquisition (DIA) and tandem mass tag (TMT) techniques, underpinned by a previously established chromosome-level genome and PacBio full-length transcriptomics. Our analysis revealed a distinctive prot
