Camelid-derived antibodies reduce myonecrosis and IL-1β release after treatment with Bothrops jararacussu venom in mice.

Snakebite envenoming in Latin America is both an occupational hazard and a neglected tropical disease. Globally, more than 420,000 envenomings occur annually, resulting in approximately 20,000 deaths. In Brazil, most cases are caused by snakes of the genus Bothrops, which comprises over 60 species distributed throughout the country. Although antivenom therapy efficiently neutralizes systemic effects, local manifestations such as myonecrosis, dermonecrosis, pain, and edema remain poorly controlled. These effects are largely mediated by snake venom phospholipases A₂ (svPLA₂s), which play a centr

Zebrafish as a translational model for Bitis arietans envenomation: Integrative proteomic and histopathological insights.

The genus Bitis, especially the species Bitis arietans, significantly contributes to snakebite incidents in Africa, causing a range of serious medical conditions. This study aimed to explore the pathological mechanisms underlying envenomations by B. arietans using zebrafish (Danio rerio) as a model organism. Blood samples were collected at 3- and 6-h post-envenomation, followed by plasma analysis through mass spectrometry. Key findings indicated that B. arietans venom caused a hemolytic effect leading to acute severe anemia by 6 h post-envenomation. A hematopoietic response was also observed,

Venomics across the Bothrops neuwiedi Species Complex Revealed a P-III Snake Venom Metalloproteases/K49-PLA2 Dichotomy and a Remarkable Paraspecific Neutralization of the Brazilian Pentabothropic Antivenom.

Snakes of the Bothrops neuwiedi complex are widely distributed and represent medically important species in Brazil. Here, we report compositional and functional profiles of the venom of seven species of Bothrops neuwiedi group: Bothrops mattogrossensis, Bothrops pauloensis, Bothrops pubescens, Bothrops diporus, Bothrops neuwiedi, Bothrops marmoratus, and Bothrops erythromelas. Toxin composition of individual and pooled venoms showed remarkable inter- and intraspecific variability of the relative abundance of toxins (evidenced by SDS-PAGE and RP-HPLC) and enzymatic activities (proteolytic, PLA2

Mechanism of Hyaluronic Acid Hydrolysis Catalyzed by Snake Venom Hyaluronidase.

Hyaluronidases are widely distributed in nature being ubiquitous in snake species (svHyal). They catalyze the hydrolysis of β-1,4-glycosidic bonds in hyaluronic acid, a critical constituent of the extracellular matrix. This facilitates the spread of venom toxins into the bloodstream, exacerbating tissue damage and systemic toxicity─a rationale for their common designation as "spreading factors". While svHyals are not directly toxic, they substantially contribute to the morbidity and mortality associated with snakebite envenomation, the world's most lethal neglected tropical disease. Despite th

Crotoxin B from the South American Rattlesnake Crotalus vegrandis Blocks Voltage-Gated Calcium Channels Independent of Its Intrinsic Catalytic Activity.

Neurotoxicity following South American Crotalus rattlesnake bite is primarily caused by crotoxin, the most abundant component in their venom. Despite the central role of voltage-gated calcium channels (CaV) in neurotransmission, direct targetability by crotoxin has been poorly explored. Crotoxin is a non-covalent heterodimer formed by an acidic subunit (CA) and a basic toxic phospholipase A2 subunit (CB). Here, we chromatographically isolated the CB subunit from Crotalus vegrandis and studied its effect on CaV heterologously expressed in tsA201 cells using the whole-cell patch-clamp technique.

Cathelicidin-like Peptide for Resistant Acinetobacter baumannii Control.

The growing global threat of antimicrobial resistance (AMR), particularly in cutaneous wound infections, represents a significant clinical and economic challenge. Biofilm formation by multidrug-resistant pathogens, such as Acinetobacter baumannii, often complicates healing and leads to therapeutic failure. Antimicrobial peptides (AMPs) are a promising alternative to conventional antibiotics due to their potent membrane-disrupting mechanism of action and lower propensity to induce resistance. Background/Objectives: This study aimed to evaluate the antibacterial, antibiofilm, and in vivo efficac

Comparative proteomic analysis reveals functional and evolutionary diversity in five Montivipera snake venoms.

Proteomic characterization of snake venoms is essential for understanding the molecular basis of their evolution and for identifying bioactive compounds of therapeutic interest. The Montivipera species endemic to the Near and Middle East region remain poorly studied despite their interesting biological activities. Previous analyses of Montivipera venoms have provided only partial proteomic profiles, with notable discrepancies between studies. To address this gap, we conducted a proteomic analysis of five Montivipera species, including M. bornmuelleri, M. bulgardaghica, M. albizona, M. raddei a

Forensic identification of Agkistrodon halys bite by the combination of ELISA and immunohistochemistry: a case report and literature review.

Snakebites constitute a significant global public health concern. Based on various toxic components and action mechanisms, snake venoms are typically classified into hemotoxic, neurotoxic, and histotoxic types. The pathological changes caused by different toxicities vary, which is often one of the key points in the forensic identification of snake bites. However, relying solely on this aspect often has certain limitations in the identification of snake bite cases, with poor qualitative and quantitative results. This article presents a case of identifying death caused by an Agkistrodon halys bi

Metalloproteinases Atroxlysin-I (Atr-I) and Atroxlysin-III (Atr-III) from Bothrops atrox snake venom bind to murine macrophage CD11b altering cytokine production.

Atroxlysin-I (Atr-I) and atroxlysin-III (Atr-III) are snake venom metalloproteinases (SVMPs) isolated from Peruvian Bothrops atrox. These proteins were previously characterized as hemorrhagic, fibrin(ogen)olytic and platelet aggregation inhibitors. In this work, we evaluated the response of murine macrophages treated with Atr-I (23 kDa, P-I class), Atr-III (56 kDa, P-III class), and B. atrox venom. Neither SVMPs nor crude venom decreased cell viability, whereas treatment with a higher concentration of Atr-I and B. atrox venom increased cellular metabolism. The SVMPs and B. atrox venom differen

Comparison of proteomes and biofunctional properties of male and female common adder (Vipera berus) venoms.

Snake venom is an ecologically critical functional trait, primarily applied for foraging and accordingly shaped by selective pressures. Recent insights underpinned the high variability of snake venoms down to the intraspecific level, with regional, ontogenetic, and seasonal variation being mostly investigated. In contrast, sex-based venom variation has received considerably less attention so far, and its influence on venom compositions is poorly described. Here, we compare venom profiles and bioactivity from pooled male and female samples of Central European adders (Vipera berus) to provide in

Conserved Enzymatic Peptides in Bitis arietans Venom Revealed by Comparative Proteomics: Implications for Cross-Reactive Antibody Targeting.

Snakebite envenoming remains a critical public health issue, and the molecular variability of venoms limits the cross-species efficacy of conventional antivenoms. Here, we conducted a comparative proteomic analysis of Bitis arietans venom to identify conserved peptide regions derived from enzymatic toxins and evaluate their potential relevance for complementary immunotherapeutic applications. Enzyme-enriched venom fractions were isolated through sequential affinity and ion-exchange chromatography and were subsequently characterized using fluorogenic FRET substrates and inhibitor assays. Omics-guided discovery and functional profiling of Da-29, a bioactive serine protease from Deinagkistrodon acutus venom, with antithrombotic activity.

BACKGROUND: Snake venom serine proteases (SVSPs) are critical toxic components that interfere with the hemostatic balance and serve as a key candidate library for anticoagulant drug development. Despite the diversity of SVSPs in Deinagkistrodon acutus venom, systematic studies of low-abundance SVSPs with thrombin-like activity profiles are limited. Hence, this research aimed to isolate and characterize a bioactive SVSP candidate from D. acutus venom using integrated multi-omics and to assess its antithrombotic effects and bleeding risk. METHODS: PacBio full-length transcriptome sequencing com

Bioinformatics-Driven Design and Evaluation of Recombinant Multi-Epitope Immunogens Derived From Snake Venom Toxins as Potential Antivenom Candidates.

Snakebite envenomation is a major public health concern, particularly in low- and middle-income regions where access to safe and effective antivenoms is limited. Traditional antivenoms, derived from immunization with crude venom, often trigger adverse reactions and lack specificity against key venom components. This study presents a bioinformatics-driven approach to design, construct, and evaluate a novel panel of recombinant multi-epitope toxin-derived immunogens targeting the most clinically significant snake venom toxin families. Five principal toxin families-three-finger toxins (3FT), phos

In vitro-in vivo discord: A preclinical study of AZD2716 and its racemate with comparison to varespladib for the development of snake venom sPLA2 inhibitors.

We evaluated a family of repurposed sPLA2 inhibitors as novel candidate snakebite envenoming therapeutics. Stereospecific (R)-7 AZD2716 and its racemic mixture were compared to varespladib in an in vitro sPLA2 assay against a sample of 26 venoms from medically important snake species from five continents. All compounds demonstrated potent nano-to picomolar IC50 values, comparable to the benchmark inhibitory profile of varespladib. Surprisingly, however, this in vitro efficacy did not translate to survival in an in vivo mouse model under GLP standard conditions at an independent third party lab

Establishing the Kenya National Antivenom Quality Control Laboratory: Preclinical Efficacy Results of Four Antivenoms Against Venoms from the "Big Five" Snake Species in Kenya.

Antivenom administration is currently the only therapy for snakebite envenoming. However, in sub-Saharan Africa, inadequate quality control systems have led to deficits in the availability, accessibility, efficacy and safety of regionally available antivenoms, which, in turn, hinder snakebite treatment and management in the region. To address this impediment to snakebite treatment in Kenya, this study aimed to assess the preclinical neutralising potencies of four different antivenoms previously or currently available in Kenya (SAIMR polyvalent, AFRIVEN, PANAF-PremiumTM and InoserpTM) against k

Inter- and Intraspecific Venom Variation in the Reclusive Rear-Fanged Black-Striped Snakes (Coniophanes).

Our current understanding of snake venom is highly biased towards species known to be medically significant in human envenomations. This vastly under-represents the true evolutionary and ecological breadth of snake venom, with gaps spanning entire clades and unique lifestyles. As a result, many genera of rear-fanged snakes lack well-understood venom profiles despite these taxa composing around 65% of known extant snake species. Methodological challenges associated with venom extraction have long been a key reason responsible for the lack of venom research on this group. Modern advancements in

New Insights into the Bioenergetic and Immunomodulatory Properties of Phospholipases A2 from Bothrops diporus Venom.

Phospholipases A2 (PLA2s) are key mediators of the cytotoxic and inflammatory activities of snake venoms. While PLA2 isoforms from Bothrops diporus venom have been characterized and shown to possess antimetastatic and antiangiogenic properties, their impact on mitochondrial bioenergetics and immune modulation has not yet been investigated. In this study, we examined the bioenergetic and immunomodulatory effects of B. diporus PLA2s using integrated biochemical, metabolic, and multiplex cytokine analyses. In MDA-MB-231 breast cancer cells, pooled PLA2s induced a dose-dependent decrease in MTT-re

A commercial insecticide-induced neurotoxicity and snake venom nerve growth factor-inspired peptides-mediated neuroprotection in Caenorhabditis elegans: Mechanistic, safety, and pharmacokinetic (in vivo imaging) evaluation of peptides.

Agriculture workers are especially at risk from insecticide (INSECT) exposure, which is associated with neurological disorders, such as Parkinson's disease (PD). The present study evaluated the mechanism of neurotoxicity induction in a commercial insect containing chlorpyrifos (50%) and cypermethrin (5%) in Caenorhabditis elegans models, as well as the neuroprotective efficacy of two custom peptides (CPs), HNP (heptadeca-neuropeptide) and TNP (trideca-neuropeptide), derived from snake venom nerve growth factor.
