The puff adder (Bitis arietans) is a highly venomous viper responsible for many snakebite fatalities in Africa, yet there have been few geographically comprehensive analyses of its venom proteins, particularly of the proteases that play a key role in pathology of envenoming. To address this, we have isolated, identified and characterised the bioactivity of the venom metalloproteases of puff adders obtained from a range of localities. Prominent in all venoms was a PI snake venom metalloproteinase (SVMP), derived from a larger PII precursor. This protein existed as either non-glycosylated (21 kD

Snake bites remain major threats with only limited diagnostic and therapeutic options. Current antibody-based antivenoms show only limited effectiveness against local tissue damage and require complex manufacturing and cold chain logistics. To overcome these limitations, a fluorescence-based assay was established to enable the sensitive detection of snake venom metalloprotease (SVMP) and serine protease (SVSP) activities in crude viper venoms from Crotalus atrox, Bothrops jararaca, and Echis carinatus. The assays showed species-specific activity profiles with detection limits in the sub-microg

Accurate prediction of peptide-protein interactions (PepPI) is crucial for advancing peptide-based anticancer drug design. In this study, we introduce ProVenTL, a computer-aided molecular design framework that leverages transfer learning and protein language model embeddings to enhance PepPI prediction accuracy and interpretability. Two complementary strategies were explored: (i) fine-tuning a CAMP model pretrained on large-scale PepPI data from the Protein Data Bank (PDB) using a curated dataset of Calloselasma rhodostoma venom peptides and cancer-related proteins, and (ii) integrating ProtT5

Snake-venom phospholipases A2 (PLA2s) are small, structurally conserved enzymes that contribute significantly to the pathophysiology of envenomation. Here, we report the purification and crystal structure of an Asp49-PLA2 isolated from the venom of Lachesis muta, a pit viper from the Peruvian Amazon. The enzyme was purified using ion-exchange and size-exclusion chromatography and exhibited phospholipase activity in a dose- and time-dependent egg-yolk degradation assay. Pure protein crystals were obtained in space group P6222 and diffracted to 2.36 Å resolution, with two molecules in the asymme

Snakebite envenoming is a neglected tropical disease that causes high mortality and morbidity. The current treatment, intravenous antivenom, comes with numerous disadvantages, making new therapeutics important. Optimized small molecules offer the possibility for oral use at the onset of envenoming, and the highly pathogenic, zinc-dependent snake venom metalloproteinase toxin family represents an attractive target for drug discovery. Through systematic chemical modification guided by molecular modeling, we describe the development of hydroxamic acid 23 (DC-174), a molecule that displays potent

Snake venom metalloproteases (SVMP) play a major role in various pathophysiological effects of snakebite envenomation, and marimastat (MMT), a selective metalloprotease inhibitor, has been investigated as a potential ancillary treatment for attenuating SVMP-mediated effects. In this work, the effect of MMT on the coagulant, hemorrhagic and neuromuscular activities of Lachesis muta (South American bushmaster) venom was assessed experimentally in vitro and in vivo and compared with therapeutic anti-Bothrops/Lachesis antivenom (AV) produced by the Instituto Butantan. MMT (0.03-3.0 mM) significant

Snakebite envenomation remains a major cause of illness globally, resulting in complex local and systemic toxic effects. Although the biochemical functions of individual snake venom components have been extensively studied, the cellular mechanisms underlying progressive tissue damage and organ dysfunction remain less well understood. This review compiles contemporary mechanistic evidence associating notable venom toxin families, such as phospholipase A₂, snake venom metalloproteinases, and three-finger toxins, with oxidative stress-induced cellular damage. Mounting evidence highlights mitochon

Perinatal asphyxia (PA) is a primary cause of neonatal morbidity, with persistent neurological sequelae, associated with mitochondrial dysfunction and redox imbalance. The lack of experimental approaches for region-specific evaluation of mitochondrial bioenergetics in structurally preserved neural tissues has limited the identification of selective vulnerabilities and redox-based therapeutic targets. Here, we introduce a real-time metabolic profiling strategy that integrates Seahorse XFe96 respirometry with Cytation 5 image-based quality control to quantify mitochondrial respiration and glycol

Naja atra venom induces severe myotoxicity and inflammation largely driven by snake venom phospholipase A2 (SVPLA2). Although the SVPLA2 inhibitor varespladib shows great therapeutic promise, the underlying molecular mechanisms remain unclear. Herein, we uncover dual pathogenic pathways. First, a translational module in which RPL35A interacts with PTBP1 to selectively enhance IL-6 and TNF-α translation without altering mRNA levels. Second, a transcriptional axis where prostaglandin E2-activated p38 MAPK enhances NF-κB signaling and reactive oxygen species production, forming an inflammatory-ox

Matrix metalloproteinase 2 (MMP-2) is the major MMP involved in hypertension and plays a central role in vascular remodeling in response to hemodynamic changes by promoting upregulation of transforming growth factor beta 1 (TGF-β1), vascular hypertrophy, fibrosis, and oxidative stress. BmooMPα-I, a metalloproteinase isolated from Bothrops moojeni venom, exhibits kininogenase activity, is capable of hydrolyzing angiotensin I, and exerts antihypertensive and cardioprotective effects. This study evaluated the effect of BmooMPα-I on MMP-2 activity and expression, TGF-β1 expression, redox balance,

Russell's viper envenomation is known to cause complications such as coagulopathy, acute kidney injury and haemolysis. Cardiac involvement, though rare, has been reported in the form of arrhythmias, myocardial infarction and ECG changes such as ST-segment elevation or T-wave inversions. We report a case of a middle-aged farmer who presented to our centre with Russell's viper envenomation, characterised by local envenomation, coagulopathy, haemolysis and ocular manifestations. His ECG showed a Brugada pattern, with no evidence of any structural cardiac abnormalities or elevated cardiac enzymes.

Sea snake venom systems provide exceptional models for studying marine adaptation and are rich sources of bioactive molecules for drug discovery. To comprehensively characterize the venom diversity of the medicinal sea snake Hydrophis cyanocinctus, this study employed an integrated multi-omics strategy. We performed deep proteomic profiling of its venom and venom gland using both data-independent acquisition (DIA) and tandem mass tag (TMT) techniques, underpinned by a previously established chromosome-level genome and PacBio full-length transcriptomics. Our analysis revealed a distinctive prot

BACKGROUND: Many clinical laboratories lack the methods or resources to quickly identify rare haemorrhagic diseases, such as genetic/acquired fibrinogen defects and factor XIII deficiency.
CASE REPORT: This report explored the value of human fibrinogen concentrate in the diagnosis and management of rare bleeding disorders through three case studies. Case 1 involved acquired hypofibrinogenemia caused by snake bite, where the coagulation abnormality was corrected by adding fibrinogen in vitro, confirming the mechanism of snake venom-specific degradation of fibrinogen. Case 2 utilised fibrinogen

Snakebite envenoming is a major neglected tropical disease, in which snake venom metalloproteinases (SVMPs) are key drivers of local tissue damage, microvascular disruption, and hemostatic imbalance. Closely related SVMPs can nevertheless range from highly hemorrhagic to essentially non-hemorrhagic while sharing a conserved Zn(II) catalytic site, and the structural causes of this divergence (particularly the role of the conserved methionine-containing turn (MET-turn) beneath the metal center) remain unclear. Here, we have combined a Protein Data Bank (PDB) inspection with theoretical calculati

The Indian cobra (genus Naja) is one of the 'Big Fours' responsible for dreaded snakebite incidents in India. Immediate anti-snake venom (ASV) treatment is the only solution for cobra envenomation. Currently available ASVs are not efficacious nationwide, probably due to geographical variations in venom. The lack of precise information on venom variation is the foremost hurdle to ASV improvement. Hence, mapping of geographical variation in snake venom proteome is of utmost importance. Here, we report mass spectrometry-based proteomic analysis of the venom of Naja naja and Naja kaouthia, collect

Snake venom proteins have attracted considerable attention as oral analgesics owing to their definite analgesic effects and lack of tolerance or addiction. Herein, we report the successful development of an optimized oral analgesic nanoparticle formulation (CS/ALG@NANT NPs), prepared by the self-assembly of chitosan/alginate andNaja atraneurotoxin (NANT) via polyelectrolyte complexation and optimized using a systematic Box-Behnken design. The resulting nanoparticles exhibited uniform spherical morphology, an appropriate particle size (278 nm), high encapsulation efficiency (>90%), and substant

Tigrin, a protein isolated from Rhabdophis tigrinus venom, is homologous to snake venom cysteine-rich secretory proteins (CRISPs); however, its biological function remains poorly understood. This study aimed to investigate whether tigrin induces pro-inflammatory cytokine production in human endothelial and monocytic cells. Tigrin was purified from the crude R. tigrinus venom and identified using SDS-PAGE. Human umbilical vein endothelial cells (HUVECs) and THP-1 monocytes were stimulated with either crude venom or purified tigrin. mRNA expression levels of IL-1β, IL-6, IL-8, and TNF-α were ana
