Oncohypertension: Unraveling the Link Between Cancer and Hypertension.

Cancer therapies have reshaped survival, but have also unmasked hypertension as a common and clinically significant complication with distinct underlying mechanisms. This review uniquely integrates mechanistic insights, particularly those related to vascular signaling pathway inhibition, alongside emerging pathways implicated in other molecularly targeted therapies, with practical therapeutic considerations specific to oncology populations. We examine how cancer biology and treatment-related factors converge to drive hypertension, with a primary focus on agents targeting vascular signaling, wh

Carboxylesterase 1 is a core metabolic vulnerability of platinum-resistant high-grade serous ovarian carcinoma.

BACKGROUND: Platinum-resistant high-grade serous ovarian carcinoma (HGSOC) is a major clinical challenge, with high mortality rates and no effective therapeutic options. Metabolic adaptations-including enhanced lipid catabolism, oxidative phosphorylation (OXPHOS), and autophagy-are major drivers of chemoresistance; however, the molecular mechanisms underpinning this metabolic reprogramming remain poorly understood. Carboxylesterase 1 (CES1), an NF-κB-regulated neutral lipase, supports malignant cell survival under nutrient-depleted conditions in other cancers, but its role in HGSOC has not bee

White noise as a non-pharmacological intervention for enhancing the recovery of rats with wound-associated sleep disorders.

OBJECTIVE: Sleep disorders are one of the common symptoms in patients with wounds, which can hinder recovery. Commonly used hypnotic medications in clinical practice are short-acting but carry risks of adverse reactions and drug abuse. As a non-invasive acoustic intervention, white noise can regulate brain electroencephalographic rhythms to improve sleep disorders. However, the interventional effects and rehabilitation outcomes of white noise on wounds with sleep disorders remain unclear. METHOD: A wound model with sleep disorders was established in rats by integrating para-chlorophenylalanin

Acute Onset Orolingual Choreoathetosis and Auditory Hallucinations in a Healthy Young Adult Following High Melatonin Dose.

Melatonin is considered a safe OTC sleep aid with rarely reported adverse effects despite limited data. We present a case of acute orolingual choreoathetosis and auditory hallucinations following 132 mg ingestion of melatonin in an otherwise healthy, 22-year-old active-duty male in IBOLC training. The patient developed abrupt, predominantly orolingual choreoathetosis and mild auditory hallucinations, 1 hour after waking. Three days earlier, he began taking 24 mg of melatonin nightly for insomnia and ingested 132 mg the night before presentation. He denied alcohol, illicit drug, or other medica

Effects of Serotonin Transporter Genotype and (Un)Predictable Maternal Separation on Affective, Cognitive, and Social Behaviors in Female Rats.

Serotonin transporter (SERT) variation is linked to depression risk after stress, conferring an increased risk to SERT-linked polymorphic region (5-HTTLPR) short-allele carriers, who show heightened sensitivity to cumulative stress and childhood maltreatment. To elucidate the neurobiological mechanisms underlying SERT × early-life adversity interactions, animal models capturing this genetic vulnerability are of translational relevance. We evaluated four maternal separation (MS) paradigms to determine how different forms of early-life stress influence adult behavior in female SERT-deficient rat

Biological Aging, Adiposity, and Inflammation in Degenerative Spine Disease: Mendelian Randomization and Longitudinal UK Biobank Analyses.

BACKGROUND: Telomere length (TL) is a marker of cumulative biological aging, but whether it causally contributes to degenerative spine disease or reflects upstream metabolic and inflammatory burden remains unclear. METHODS: We performed a population-based study integrating two-sample Mendelian randomization (MR) with longitudinal UK Biobank analyses. MR used genetic instruments for leukocyte TL, body mass index (BMI), and C-reactive protein (CRP) against five degenerative spine GWAS outcomes. We estimated direct TL effects after adjustment for BMI and CRP. Observational analyses included 265,

Sex differences in responses to neurosteroid therapy after preterm birth.

Progesterone production by the placenta leads to elevated levels of neurosteroids in both the maternal and fetal circulations during late gestation. Allopregnanolone levels are crucial because they provide both neuroprotective and trophic support in the fetal brain. These include protecting against excessive excitation and supporting oligodendrocyte development and myelination. Pregnancy compromises and particularly premature birth reduce or totally deprive the newborn brain of this neurosteroid mediated protection and support. The premature fall in allopregnanolone levels may increase suscept

Understanding Food Insecurity in Westminster, UK: A Community-Informed Systems Thinking Approach.

INTRODUCTION: Food insecurity is increasing in the United Kingdom and is a growing public health concern. In Westminster, a highly diverse London borough, rates continue to rise in the most deprived areas, such as the Church Street ward, despite increasing food support and local policies. METHODS: This study used a community-informed systems thinking approach to understand the local dynamics. 26 stakeholders from 6 local services participated in group model-building (GMB) workshops and interviews to provide insight into how services and policies interact with the structural drivers of food in

Estimating the Economic Value of Supported Online Parent-Led Cognitive Behavioral Therapy to Prevent Child Anxiety Problems: Economic Evaluation of the MYCATS Cluster Randomized Controlled Trial.

BACKGROUND: Childhood anxiety disorders are common and lead to substantial negative long-term outcomes in various life domains. Although digital interventions for preventing childhood anxiety problems have been increasingly used in the recent decade, there is limited evidence documenting their cost-effectiveness (CE). OBJECTIVE: This study aimed to evaluate the CE of a parent-led online cognitive behavioral therapy with therapist support (OSI+TS) alongside usual school practice (USP), compared to USP alone for children aged 4-7 years at risk of anxiety disorders identified through universal s

Examining factors associated with catheter-defined unplanned dialysis initiation: a UK Renal Registry analysis.

BACKGROUND: In the UK, nearly half of patients with end stage kidney disease (ESKD) who initiate dialysis do so in an unplanned manner. A reduced quality of life, increased number of hospitalisations, and higher rates of mortality are just a few of the consequences of unplanned dialysis initiation (UDI). Our study examined factors associated with UDI in patients with ESKD in England, focusing on the effects of ethnicity and late presentation (LP). PATIENTS AND METHODS: We conducted a retrospective cohort study using data from the UK Renal Registry on 26 512 adult patients initiating dialysis

BMAL1 Overexpression in the SCN Confers Protection Against Anxiety-Like Behaviors of Chronic Sleep Restricted-Mice.

Children and adolescents globally experience inadequate sleep, which harms their cognitive functions and emotional regulation. The role of circadian clock genes in influencing the antioxidant and anti-inflammatory capabilities has been well-established. Previous studies reported that the impairment of neural activity within the suprachiasmatic nucleus (SCN) can lead to anxiety-like behaviors in mice. Our study was designed to explore the role of BMAL1 expression in SCN, which could be responsible for anxiety-like behaviors induced by chronic sleep restriction (SR) in young mice. Here, 5-week-o

Withaferin A induces a decrease in sacsin levels and the collapse of nestin networks: Relevance for glioblastoma.

Withaferin A (WFA) is a steroidal lactone derived from Withania somnifera, a plant cornerstone in Ayurvedic medicine for over 3000 years. WFA exhibits antitumoral properties against glioblastoma (GBM), but its mechanisms remain incompletely understood. We investigated its effect on the intermediate filament nestin, a marker of GBM stemness and poor prognosis. WFA treatment induced a partial, reversible collapse of nestin networks, characterized by juxtanuclear accumulation. WFA also decreased the elasticity of GBM cells, inhibited their proliferation and migration, increased their sensitivity

Across the Life Course and Into Custody: Understanding Incarceration Pathways of Deaf and Hard of Hearing Individuals.

Incarceration is rarely an isolated event but reflects cumulative institutional and interpersonal disadvantage across the life course. Although life-course criminology explains these trajectories for the general population, it has rarely been applied to d/Deaf and hard-of-hearing (D/HH) individuals, a population that is both understudied and difficult to access. This study addresses that gap using Life History Calendars to reconstruct the pathways of 16 incarcerated D/HH individuals in the southeastern United States. Findings identify early communication deprivation and family isolation as dis

Positive youth development and psychological outcomes: Longitudinal associations with curiosity, creativity, and well-being.

This study examined the longitudinal associations between positive youth development (PYD) and three key psychological outcomes in adolescence: curiosity, creativity, and well-being. Using two-wave survey data from 699 Chinese middle school students, we investigated whether four PYD dimensions, including love, aspiration, faith, and grit, predicted changes in these outcomes over time. Longitudinal measurement invariance was first established for curiosity and well-being, with partial support for creativity, allowing for meaningful interpretation of structural relations. Structural equation mod

'We don't need to check it, but we do check it': a qualitative study of the views of primary care healthcare professionals on blood test monitoring in three long-term conditions.

BACKGROUND: In the UK there is variation in which tests are used for monitoring Long Term Conditions (LTCs). Guidance that underpins testing practices for LTCs is often based on expert opinion rather than evidence. There is little insight into the views of primary care staff as to why such variation exists or on whether current LTC blood test monitoring is optimal. AIMS: To understand the views of Healthcare Professionals on blood test monitoring for three LTCs (type 2 diabetes, hypertension and chronic kidney disease), identify factors that shape monitoring practices for LTCs and inform the

A core-shell microneedle patch coordinates early anti-inflammatory modulation and sustained pro-angiogenic stimulation for cardiac repair after myocardial infarction.

Myocardial infarction (MI) involves a temporally evolving pathological cascade, in which acute NLRP3 inflammasome-mediated inflammation and insufficient reparative angiogenesis sequentially contribute to adverse ventricular remodeling. However, most current therapies lack the spatiotemporal control needed to match these evolving therapeutic demands. Here, we developed a core-shell microneedle (MN)-based epicardial patch for spatiotemporally programmed MI treatment through sequential delivery of the NLRP3 inhibitor MCC950 and basic fibroblast growth factor (bFGF). The rapidly hydrating methacry
